Notes: Summary The effects of the inhibition of steroidogenesis by trilostane on oocyte maturation were examined by studying spontaneous maturation and fertilization in vitro. 10−6 M trilostane had no influence on the meiotic process, whether the oocytes were naked or not. At a concentration of 10−6 M and 10−7 M trilostane, low normal pronuclear formation and high polyspermy were found during in vitro fertilization. However, no retarded male pronuclear development could be detected in the trilostane-treated group. Thus, steroid producing activity within ova is apparently necessary to prevent multiple sperm penetration, but it has no effect on meiosis or the action of the so-called male pronucleus growth factor (MPGF).

Notes: Summary An autopsy case with clinical features of progressive parkinsonism and dementia of presenile occurrence was characterized by the following neuropathologic findings: (1) severe degeneration in the striationigral system and (2) widespread occurrence of numerous senile plaques and Alzheimer's neurofibrillary tangles in the cerebral cortex. We believe this to be a very rare case of striatonigral degeneration combined with Alzheimer's disease, because, as far as we know, only two similar cases have been reported and they had not such typical characteristics of Alzheimer's disease as ours did. In addition, in our case the biochemical examination revealed that the activity of one of the dopamine-synthesizing enzymes, tyrosine hydroxylase, was greatly decreased in the nigro-striato-pallidal system.

Notes: Summary The case of a 14-month-old boy afflicted with central neurofibrobromatosis is presented which included the following dysgenetic disorders: glioma of the optic nerve; diffuse cellular and fibrillar gliosis with deficient myelinization; subpial, subendymal and perivascular fibrillar gliosis with hypermyelinization; diffuse cellular and fibrillar gliosis in the cortex mainly of the limbic system and the paleocerebellum; ectopic subarachnoid glia; paraendymal spongioblast nodules and a single blastomatous glial nest. The diffuse dysgenetic gliosis did not appear to be blastomatous, and thus it cannot be compared with diffuse glioblastomatosis. The question of special dysgenetic disturbances of myelinization—with respect both to retarded and to paradox myelinization—is discussed in relation to findings made in recent electron microscopic studies and animal experiments. Bielschowsky's term: glial dysgenesia with blastomatosis, (Gliadysgenesien mit blastomatösem Einschlag) applies to tuberous sclerosis and central neurofibromatosis as well as to a very large group of singular, unifocal lesions for which only relatively rarely genetic connections with the two principal groups, i.e. Reckling-hausen's and Bourneville's disease, can be found. Epileptogenic, tumor-like, “paratumoral”, dysgenetic uncus gliosis and dysgenetic aqueduct stenosis for example, should be included here.

Notes: Summary A 57 year old male complained of tiredness, apathy, dysarthria, unsteadiness of gait. The symptoms became more and more severe. Temperatures of 39° C (102.2° F) appeared and the patient died after 4 months illness. At autopsy pulmonary embolism was found. The neuropathological examination revealed changes typical of neuroreticulosis, such as tumor-like proliferation of reticulum cells in the medio-basal parts of the brain with abundant formation of reticulin fibres and diffuse astro- and microglial proliferations also in the cerebral cortex with perivascular cuffs. Electron microscopic study performed after 9 months of formalin-fixation of the tissue showed: 1. “virus-like”-filamentous structures in the cytoplasm of proliferated reticular cells near the vascular wall, which had an outside diameter of about 140–150 Å, and a length of 2300 Å. 2. peculiar distribution of nucleoprotein in the proliferative cells, such as filamentous or net-like structures of about 160 Å outside diameter. 3. intraplasmatic and nuclear crystalline structures of oblong rhombic shape with differentiated dark and bright stripes with a periodicity of 300–350 Å. There is some possibility that mixoviruses may play an aetiological role in neuroreticulosis.

Notes: Summary The integrity of cholinergic and monoaminergic neuronal systems in Alzheimer-type dementia (ATD) was studied using a combination of morphological and biochemical procedures applied to samples from seven ATD brains and ten control brains. On morphological examination, the number of neurons was counted in the nucleus basalis of Meynert (nbM), the locus coeruleus (LC) and the nucleus centralis superior (NCS). Biochemically, the activities of choline acetyltransferase (ChAT) and the concentrations of noradrenaline (NA) and serotonin (5-hydroxytryptamine; 5-HT) were measured in the nbM, LC, NCS, frontal cortex, temporal cortex and occipital cortex. Compared with the controls, the mean number of neurons in the nbM and LC were significantly reduced in the ATD brains. The neuronal loss in the NCS in the ATD brains was not significant. The ChAT activities in all regions from the ATD brains had a tendency to be reduced. Marked reductions in the NA concentrations in the LC and 5-HT concentrations in the temporal and occipital cortices were found in the ATD brains. These findings suggest that various neurotransmitter systems, including cholinergic, noradrenergic and serotonergic systems, are affected in the ATD brains.
