    Notes: Summary Dopamine-β-hydroxylase (DBH) immunohistochemistry was used to demonstrate the noradrenergic fibers and terminals in the anterior column of the rat lumbosacral spinal segments. PAP-positive varicose fibers were widely distributed in the gray matter with preferential accumulation in the nuclear regions containing motoneurons involved in the contraction of perineal striated muscles. Unmyelinated DBH fibers were composed of nodular enlargements (varicosities, 0.4–3.0 μm in diameter) and very fine, short intervals (intervaricose segments, 0.1–0.2 μm in diameter and 1.0–4.0 μm in length). DBH-positive dense products were electron microscopically often confined within small granular particles and less frequently within large granules. Additionally, in order to characterize the innervation pattern of noradrenergic fibers on dendritic bundles organized in the motoneuronal pools innervating the pelvic small muscles, semi-quantitative analysis was done in the area of the dorsolateral nucleus endowed with especially well-developed dendritic bundles. DBH terminals contacting with unreactive dendrites were more common (67.9%) than those with neuronal somata (15.1%), and the remainder (17%) had no contacts with surrounding neuronal elements. Furthermore, specialized synaptic formations were observed in only 20.1% of these nodules. The results suggest that bulbospinal descending noradrenergic neuron systems influence the functioning of pelvic muscles principally via the neuronal contacts with dendritic bundles in the spinal cord.

    Notes: Antigen recognition by T cells is the key event for the antigen specific immune responses to be triggered. This recognition is initiated by the binding of the T cell receptor (TCR) to antigen peptide/major histocompatibility complex (MHC) on the surface of the antigen presenting cells. TCR on most of the T cells is a heterodimer composed of α and β chains which are associated with CD3 γδε as well as ζ chains, the signal transmission molecules. The dynamics of this TCR complex upon antigen/MHC recognition, however, has not been well understood. In this paper the authors analyse the configuration of TCR complex on T cells from a TCR β chain gene transgenic mouse (TGM) strain. Unlike many other TGM strains reported, a considerable proportion of T cells from this TGM expresses both transgene-encoded (Vβ3) and endogenous TCR β chains on their surface. By immunoprecipitation and immunoblotting analysis of T cells stimulated with a superantigen, staphylococcal enterotoxin B (SEB), the authors found that Vβ3 was coprecipitated with Vβ8, demonstrating the clustering of TCR αβ upon superantigen/MHC recognition.

    Notes: Summary In our previous study we found that the ARH 77 human B lymphoblastoid cell line, originating from a patient with multiple myeloma, produced both human interferon-α (HuIFN-α) and HuIFN-β after induction with Sendai virus. In order to examine whether IFN-α-producing ARH 77 cell clones can be separated from IFN-β-producing ones, the ARH 77 line was cloned by the soft agar method. Twelve clones chosen at random were examined for IFN production and the antigenic types of IFN produced were determined. All examined clones simultaneously produced both HuIFN-α and HuIFN-β, although the ratio of HuIFN-α to HuIFN-β production was variable among the clones. This result suggests that one lymphoblastoid cell can produce both HuIFN-α and HuIFN-β.

    Notes: Summary.  To investigate the role of apoptosis in the pathogenesis of HIV infection we used macaques infected with simian immunodeficiency virus (SIV) as a primate model and examined the characteristics of the apoptosis of lymphocytes in SIV mac-infected macaques. In vitro apoptosis was more strongly induced in peripheral blood mononcuclear cells (PBMC) from SIV mac239- infected macaques than those from uninfected controls. We found that the frequency of Fas antigen-positive cells was higher in PBMC from SIV mac-infected macaques than from uninfected controls, and in vitro apoptosis of PBMC was suppressed by an inhibitor of the interleukin-lβ converting enzyme (ICE) family proteases. In biopsied lymph nodes, the number of apoptotic nuclei in T cell-dependent areas was higher in SIVmac-infected macaques than in unifected controls. A higher number of apoptotic nuclei in lymph nodes of SIVmac-infected macaques was observed in the stage of persisent general lymphadenopathy than in those with AIDS-related complex, while there was no significant difference in the extent of apoptosis of cultured PBMC among the SIVmac-infected macaques. These results suggest that in vitro apoptosis is mediated by the Fas/Fas ligand and ICE system and that apoptosis in lymph nodes may be more closely related to the stage of SIVmac infection than is that of cultured PBMC.

    Notes: Summary Interferon-α (IFN-α) production was investigated in whole-blood cultures of 66 bladder cancer patients and 65 control subjects. IFN synthesis was induced with Sendai virus, and IFN activity was assayed in FL cells challenged with vesicular stomatitis virus (VSV). The mean levels of the IFN-α produced were 5,724±2,288 IU/ml in the control subjects and 4,800±2,353 IU/ml in the bladder cancer patients. IFN-α production was significantly suppressed in the bladder cancer patients compared with that in the control subjects (P (0.05). The impairment in IFN-α production correlated with the tumor grade, and it was shown that the tendency toward decreased IFN-α production was closely associated with the advancement of the tumor stage. Our results suggested that the decreased IFN-α production may contribute to the disordered immunoregulation in bladder cancer patients.

    Notes: Summary High doses of rabbit interferon (2×106 and 2×105 units/ml, a drop of about 0.1 ml administered topically into the eye twice daily) suppressed the rejection of rabbit corneal xenograft, whereas a low dose (2×103 units/ml) enhanced it. However, neither heat-inactivated rabbit interferon nor mouse L cell interferon showed any effects on rabbit corneal grafts.

    Notes: Abstract This study reports the establishment of cloned hybridomas between mouse myeloma cells (P3-X63-Ag8, 6·5·3) and spleen cells from BALB/c mice immunized with lung homogenates of nude mice (BALB/c nu/nu) heavily infected withPneumocystis carinii. A hybridoma subclone, designated 1E12-8, produced antibodies of an IgG1 subclass. Using indirect immunofluorescent antibody techniques, the monoclonal antibody was found to be directed toward the pellicle antigen of air-driedP. carinii both in trophozoite and cyst forms, and to recognize theP. carinii antigen not only from nude mice but also from rats. The antibody did not cross-react with other components of the infected host lung tissue and showed little cross-reactivity with fungi examined. The monoclonal antibody should be useful in the isolation and identification of corresponding antigenic substances ofP. carinii and may provide a useful tool in epidemiology, taxonomy, and diagnosis of this organism.
