    Description / Table of Contents: Summary By using an enzyme-bridge immunoperoxidase (PAP) technique, localization of so-called pregnancy-specific β1-glycoprotein (SP1) and placental-specific tissue proteins (PP5, PP10, PP11, PP12) was investigated in 19 cases of breast cancer, 24 cases of testicular malignant tumours, 12 cases of gastric cancer and some cases of other malignant tumours. These five glycoproteins isolated from human term placentae and characterized by Bohn are localised mainly in the cytoplasm, or nucleus of syncytiotrophoblast and appear to be specific for the trophoblast. But to a certain percentage these proteins could also be detected in the cytoplasma of malignant cells: In breast cancer, SP1 was present in 52.6% of cases, PP5 in 63.2%, PP10 in 68.4%, PP11 in 55.6% and PP12 in 31.6%. In testicular malignant tumours the detection rates of these proteins varied from 20.8 to 75.0% and in gastric cancer from 41.7 to 66.7%. In total 50% of the tumours tested were SP1-positive, 58.3% PP5-positive, 55.6% PP10-positive, 38.0% PP11-positive and 31.9% PP12-positive. In addition, it was found that mononuclear histiocytes showed a strong cytoplasmic staining for PP10 and PP12 and that a few other non-cancerous cells (i.e. striated cells in gastric metaplasia, gastric polyps etc.) stained for PP5, PP10, PP11 and/or PP12. All control sections were negative in malignant cells as well as in other tissue cells. This study confirms the reports that some kinds of malignant tumours produce SP1 and indicates that the ectopic production of proteins normally produced by the trophoblast is a common finding in malignant tumours. It furthermore suggests that such proteins may be useful as markers in monitoring patients with malignant diseases. The possible mechanisms of ectopic production of these inappropriate proteins in malignant tumours are discussed.
    Notes: Zusammenfassung Unter Verwendung der ‚Enzyme-Bridge‘-Immunoperoxidase (PAP)-Technik wurde das sogenannte schwangerschaftsspezifische β1-Glykoprotein (SP1) und einige placentaspezifische Gewebsproteine (PP5, PP10, PP11, PP12) im Gewebe von malignen Tumoren lokalisiert. Diese von Bohn aus menschlichen Placenten isolierten und charakterisierten fünf Glykoproteine wurden in der Placenta hauptsächlich im Zytoplasma oder in den Zellkernen des Syncytiotrophoblast lokalisiert und scheinen für die Placenta spezifisch zu sein. Zu einem gewissen Prozentsatz konnten diese Proteine aber auch im Zytoplasma von malignen Zellen nachgewiesen werden. Untersucht wurden 19 Fälle von Brustkrebs, 24 Fälle von malignen Hodentumoren, 12 Fälle von Magen-Carcinomen und einige Fälle von anderen malignen Tumoren: Bei Brustkrebs war SP1 in 52,6%, PP5 in 63,2%, PP10 in 68,4%, PP11 in 55,6% und PP12 in 31,6% der Fälle positiv. Bei malignen Hodentumoren lag die Nachweisrate dieser Proteine zwischen 20,8% und 75,0% und bei Magen-Carcinomen zwischen 41,7% und 66,7%. Insgesamt waren von den getesteten malignen Tumoren 50% SP1-positiv, 58,3% PP5-positiv, 55,6% PP10-positiv, 38,0% PP11-positiv und 31,9% PP12-positiv. Zusätzlich wurde gefunden, daß mononukleare Histiozyten im Krebsgewebe eine starke zytoplasmatische Färbung auf PP10 und PP12 zeigten und daß einige wenige andere nichtkanzeröse Zellen (Saumzellen bei intestinaler Metaplasie, Magenpolypzellen) auf PP5, PP10, PP11 und/oder PP12 anfärbten. Alle Kontrollschnitte waren negativ, sowohl bei malignen Zellen als auch bei anderen Gewebszellen. Diese Untersuchung bestätigt die Berichte über das Vorkommen von SP1 bei einigen malignen Tumoren und zeigt, daß auch andere Proteine die normalerweise nur im Trophoblast vorkommen, von malignen Tumoren ektopisch produziert werden. Sie weist außerdem auf die Möglichkeit hin, solche Proteine als Marker bei der Überwachung von Patienten mit malignen Erkrankungen zu verwenden. Die möglichen Mechanismen der ektopischen Produktion dieser Proteine in malignen Tumoren werden diskutiert.

    Notes: By using an epitaxial growth technique, Co73Cr15Pt12(112¯0) bi-crystalline thin films formed on Cr–Tix(001)[x=0–15 at. %] single crystalline underlayers were prepared to investigate the relationship between the magnetic properties and the lattice distortion in the magnetic layer. X-ray diffraction measurement shows that the best lattice matching between the c axis of the CoCrPt and the [110] direction of the Cr–Ti underlayer is realized and the CoCrPt layer shows an ideal symmetrical hcp. structure when the Ti concentration is 10 at. %, where the maximum magneto-crystalline anisotropy energy and coercivity values are observed. Distortion in the hcp. CoCrPt magnetic crystal grains decreases the uniaxial magneto-crystalline anisotropy energy, thus lowering the attainable medium coercivity. © 1996 American Institute of Physics.

    Notes: The magnetic properties of CoCrPt/CoCrPtSi dual-magnetic-layered thin films are investigated in terms of the crystalline microstructures using a vibrating-sample magnometer, transmission electron microscope, and scanning electron microscope. The magnetic crystal grains of the films are aligned with forming chainlike clusters. Each stacked magnetic crystal grain is epitaxially grown on individually isolated columnar Cr crystals with a relationship of hcp (101¯1)Co-based alloy //bcc (110)Cr. The direction of magnetic easy axes of two stacked magnetic crystals are aligned in a same direction. Single-crystalline dual-magnetic-layered thin films, which are epitaxially grown on MgO(110) single-crystal substrates, are prepared to estimate the Ku of the dual-layered magnetic grains. The Ku is determined to be 4×106 erg/cm3, whose value is about 2 times as large as that of single-layered CoCrPtSi. This large anisotropy energy is presumed to lead the increase of Hc in the dual-layered magnetic films.

    Notes: The magnetization process of ferrofluids with carrier fluids of water, paraffin, and alkylnaphtalene was investigated in a temperature range from 77 to 300 K as functions of the freezing rate and the intensity of cooling magnetic fields. A uniaxial magnetic anisotropy is induced by field cooling in frozen ferrofluids. This induced anisotropy which is caused by the formation of clustering of magnetic particles disappears on heating near the melting point. The magnetization of the frozen fluids exhibits an anomalous increase below the melting point of the carrier fluids. In a frozen state, no relative motion of particles, such as Brownian motion, occurs. With increasing temperature, the inhibited motion will be released gradually by the recovery of viscosity. The anomaly may be related to glassy transition and premelting. These magnetic properties are discussed in terms of mesoscopic phase transition between liquid and solid.

    Notes: Summary The production of human chorionic gonadotropin (hCG), pregnancy-specific beta-1 glycoprotein (SP1), and the various placental tissue proteins (PP4, PP5, PP10, PP11, PP12, MP1) was examined in human choriocarcinoma cell lines using radioimmunoassay (RIA) and an immunoperoxidase (PAP) staining method. In 6 gestational and 2 nongestational choriocarcinoma cell lines, hCG was produced in vitro. A significant heterogeneity of hCG production was noted among the cell lines. Production of hCG in the xenograft tumors was consistent with the hCG production of the same cell lines in vitro. SP1 was demonstrated both in the culture supermatants and the xenograft tumors of 3 cell lines, and the production was found to be inversely correlated with the hCG production of the cell lines. Positive staining for PP4 and MP1, and weakly positive staining for PP5, PP10, and PP12 was found in the xenograft tumors by immunoperoxidase (PAP) stain, but the staining for PP11 was negative. These placental proteins showed positive staining in portions of choriocarcinoma cells. These results indicate that placental proteins are not equally produced in heterogeneous choriocarcinoma cells.

    Notes: Summary The patient was a 57-year-old woman with ovarian serous cystadenocarcinoma in FIGO clinical stage IV. Cancer antigen 125 (CA 125), tissue polypeptide antigen (TPA) and carcinoembryonic antigen (CEA) were immunohistochemically demonstrated in tumor cells, and the variations of serum CA125 and TPA levels reflected the clinical course. The tumor tissue obtained at exploratory laparotomy was minced with scissors, and transplanted subcutaneously into female nude mice for in vivo maintenance. The tumor cells from 5th generation nude mice were dispersed in Eagle's minimal essential medium supplemented with 10% fetal calf serum, and incubated in Falcon tissue culture dishes at 37°C in 5% CO2 in air for in vitro maintenance. The results were as follows: Histopathologically the tumor transplanted into nude mice showed a cystadenocarcinoma, which closely resembled the original human tumor. Immunohistochemically CA125, TPA and CEA were demonstrated in the tumor transplanted into nude mice as well as in the original human tumor. From the growth curve in nude mice, the doubling time was estimated to be about 3.5 days. Serum TPA levels in nude mice were increased in proportion to the tumor growth after transplantation, but serum levels CA125 and CEA were normal. The concentrations of CA125 and TPA were increased in the conditioned media compared with the control media, although the elevated values were decreased with subsequent passages. CEA concentrations in the conditioned media were unchanged.
