Notes: Abstract Eighty-six children (44 males, 42 females) were identified as having non-tuberculous mycobacterial lymphadenitis. The diagnostic criteria were either culture of the organism from the affected lymph node (n=68), or, when culture was negative, a positive skin test with non-tuberculous mycobacterial antigens and negative skin test responses to tuberculin purified protein derivative (PPD) in association with typical histological features (n=18). All children had histopathological findings of granulomatous inflammation with caseation and/or acid-fast bacilli. Eighty-two percent of the children were under 5 years of age at presentation and 30% were less than 2 years old. Most (79%) were city dwellers. Lymph node enlargement had been present for less than 6 months in almost all children (97.5%) and was almost exclusively in the face and neck region (97%). Disease was confined to the involved lymph nodes in 56% but had extended beyond the confines of the infected node to form a collar stud abscess in 38% and 6% presented with a skin sinus. Extranodal extension did not show any statistically significant association with the duration of lymphadenopathy. The duration of lymphadenopathy had been greater in those children in whom an organism was not isolated on culture resected tissue χ2, P〈0.01). All children were treated surgically, and recurrence occurred in five patients. This study describes the clinical and demographic characteristics of non-tuberculous mycobacterial lymphadenopathy in children in a population in which tuberculous adenitis is rare. Recognition of these features may allow earlier diagnosis and appropriate surgical therapy.

Notes: The η refinement of Rogers [Acta Cryst. (1981), A37, 734-741] has been applied to a wide range of noncentrosymmetric structures containing medium to strong anomalous scatterers; it has been shown to be an effective and robust method. The use of the general term 'absolute structure' (to signify a structure successfully distinguished from its inverse by, for example, analysis of anomalous scattering effects) is recommended.

Notes: A survey of 102 structures flagged with 'absolute configuration' in the Cambridge Crystallographic Data Centre files for 1982 reveals many unsatisfactory features in the original publications. These are associated with data collection (lack of Friedel opposites and of absorption corrections), misinterpretation of the term 'absolute configuration' (claimed for achiral molecules and space groups), methods used to determine absolute configuration (over-optimistic interpretation of discriminatory tests) and presentation of results (little experimental detail, no prominent statement that an absolute configuration was assigned, no clear description of the methods used). Some confusion arises because of unintentional flagging of absolute configurations determined by solely chemical means.

Notes: Thirteen non-centrosymmetric structures with absorption coefficients ranging from 45 to 0.6 mm-1 were refined using both absorption-corrected and uncorrected data. The absorption was in no case severe, since \bar \mu r was less than 1 for each structure. In these circumstances, the absolute-structure parameter η could be determined equally clearly in both cases, although its e.s.d. increased somewhat for uncorrected data. More serious were the effects of uncorrected data on the precision of molecular dimensions.

Notes: The photosynthetic capacity and Rubisco levels of primary wheat leaves were reduced significantly in plants grown on media supplemented with 1 % glucose. Neither the chlorophyll content nor the growth rate of the primary leaf was affected by this treatment. In glucose-fed plants a decrease in levels of mRNA encoding several Calvin cycle enzymes (fructose 1,6-bisphosphatase, sedoheptulose 1,7-bisphosphatase, phosphoglycerate kinase and Rubisco Small Subunit) was found to correspond to an increase in the internal levels of glucose. In control plants endogenous levels of glucose and sucrose were measured in leaf sections from the base to the tip of the primary leaf, which revealed a gradient of each of these sugars within the leaf, with the highest levels at the tip. Conversely, Calvin cycle transcripts were seen to decrease in the oldest tip sections. In glucose-fed plants, the endogenous glucose levels were increased throughout the leaf but Calvin cycle RNA levels were most markedly reduced in the mid-leaf section. These data indicate that the changing metabolic status of the cells from the base to the tip of the primary wheat leaf may play a role in the regulation of Calvin cycle gene expression during normal leaf development.
