    Notes: Summary Cryostat sections of 12 gliomas and of 3 peritumoral brain tissue samples were investigated for mononuclear cell infiltration by immunohistochemistry, concentrating on cells expressing monocyte/macrophage markers. Only low numbers of T cells were detected in the tumors, whereas in average 20%–30% of all cells present in the samples were recognized by various macrophage markers. These cells carried surface epitopes with known function, like Fc-γ (Fcg) and complement receptors. Microglial cells, in comparison to typical debris laden macrophages, were only recognized by a restricted panel of macrophages markers (anti-Fcg receptors 1, 2, 3, complement receptor CR3, HLA DR, common leucocyte antigen CD45 and the monocyte marker RM3/1). In peritumoral tissue mainly dendritic, microglia-like cells were present, which revealed decreased expression of antigens CD4, RM3/1 and Fcg receptors in comparison to those in gliomas. A significant positive correlation was found between the number of RM3/1 or CR3 (CD11b)-positive cells and the proliferation rate of the tumors as documented by the number of bromodeoxyuridine-positive or Ki-67+ cells.

    Description / Table of Contents: Zusammenfassung Die biologische Aktivität von 5,6-trans-25-Hydroxycholecalciferol wurde bei Patienten mit terminaler Niereninsuffizienz untersucht. Alle Patienten erhielten über einen Zeitraum von 6 Tagen täglich 100 U dieser Substanz intravenös. Nach dieser Therapie fand sich ein Anstieg der Absorption von47Calcium bei 4 Patienten sowie bei allen ein Anstieg des Serumcalciums. Die Ergebnisse lassen hoffen, daß 5,6-trans-25-Hydroxycholecalciferol schon in niedriger Dosierung eine nützliche Therapie der urämischen Osteopathie sein könnte, um so mehr, als diese Substanz kein Calcium aus dem Knochen mobilisiert.
    Notes: Summary Investigations were performed in order to test the biological effectiveness of 5,6-trans-25-hydroxycholecalciferol in patients suffering from terminal renal failure. All patients received for six days a daily i.v. injection of 100 U of this compound: The absorption of47calcium increased in four of five patients, the serum calcium level rose in all. The data suggest that 5,6-trans-25-hydroxycholecalciferol may be useful in the treatment of uremic bone disease, especially because this compound has little, if any, activity in stimulating the mobilization of calcium from the bone.

    Description / Table of Contents: Summary Elimination of ampicillin and gentamicin by hemofiltration was measured in 5 and 4 patients, respectively, being treated for end stage renal failure. Serum half-life time of ampicillin after a single I.V. dose of 2 g, was found to be 2.97±0.73 h, and the filtrate concentrations declined parallel to the serum ampicillin levels, as was to be expected. The serum half-life time after a single I.V. dose of 1 mg/kg body weight gentamicin was 3.26±0.42 h, and considerably shorter than T/2 of gentamicin in conventional hemodialysis, while the filtrate concentrations increased paradoxically with declining serum levels. This could be explained by binding of this aminoglycoside to the polyacrylnitril membrane of the RP 6® dialyser used for hemofiltration.
    Notes: Zusammenfassung Bei 5 bzw. 4 Patienten mit terminaler Niereninsuffizienz wurde die Elimination von Ampicillin bzw. Gentamicin durch die Hämofiltration gemessen. Die Serum-Halbwertzeit von Ampicillin betrug nach einer Einzeldosis von 2 g i.v. 2,97±0,73 h und die Filtratkonzentration nahm, wie zu erwarten war, parallel den Serum-Ampicillin-Spiegeln ab. Die Serum-Halbwertzeit von Gentamicin betrug nach einer Einzeldosis von 1 mg/kg Körpergewicht i.v. 3,26±0,42 h und war damit beträchtlich kürzer als T/2 von Gentamicin bei der konventionellen Hämodialyse, während die Filtratkonzentrationen paradoxerweise mit abfallenden Serum-Spiegeln anstiegen. Dieses Verhalten könnte durch eine Bindung des Aminoglykosids an die Polyacrylnitrilmembran des RP 6® Dialysators, wie er für die Hämofiltration verwendet wurde, erklärt werden.

    Description / Table of Contents: Zusammenfassung Bei 8 gesunden Versuchspersonen (Durchschnittsalter 25,6 Jahre) wurde nach oraler Applikation von 500 mg und 1000 mg Calcium die Calcium-Resorption bestimmt. Hierzu wurde ein Doppelisotopenverfahren mit angereicherten stabilen Calciumisotopen verwendet, das, da im Gegensatz zu Radiotracermethoden jegliche Strahlenbelastung vermieden wird, uneingeschränkt anwendbar ist. Die oral verabreichten Calciumsalze wurden mit48Ca, das intravenös injizierte CaCl2 mit46Ca markiert. Die Bestimmung der beiden Isotope in Serum- und Urinproben erfolgte mit Hilfe der Neutronenaktivierungsanalyse. Für die Resorptionsquote wurde, unabhängig von der Calcium-Dosis, ein Wert von 30% gefunden. Aus dem46Ca-Gehalt im Serum wurde für das in 24 h ausgetauschte Körper-Calcium ein mittlerer Wert von 6,4±1,0 g Calcium oder 98,8±15,4 mg Ca/kg Körpergewicht berechnet.
    Notes: Summary The calcium absorption was determined in 8 healthy test persons (average age 25.6 years) after oral administration of 500 mg and 1000 mg of this element. A double isotope method with enriched stable calcium isotopes was used which can be applied without any restriction as, unlike in radiotracer techniques, any exposure to ionizing radiation is avoided. The calcium administered orally was labelled with48Ca, the CaCl2 injected intravenously with46Ca. The determination of both isotopes in serum and urine samples was carried out by means of neutron activation analysis. Independent of the calcium dose given, an absorption of 30% was found. From the46Ca content in the serum samples a mean value of 6.4±1.0 g calcium or 98.8±15.4 mg Ca/kg body weight was calculated for the 24 hr-exchangeable calcium body pool.

    Notes: Sera from 204 patients with acute or former reactions following penicillin treatment were investigated by RAST with specificity for benzylpenicilloyl and phenoxymethyl-penicilloyl. The IgE antibody levels were followed up for of more than 2 years. Positive results could be observed at the time of the acute reactions and the weeks following; later on the amounts of specific IgE decreased, with negative RAST results after months or years in many cases. Occasionally negative RAST results occurred within 30 days; on the other hand high levels of specific IgE persisted for years in some patients. Total serum IgE measured by RIST and the RAST results showed similar patterns, with mean levels of 568 u/ml between 0 and 30 days after the penicillin allergic reaction, 286 u/ml between 30 days and 1 year, and 195 u/ml in cases after 1 year corresponded well with the observed decline in specific IgE.Skin tests performed with penicilloyl-polylysine in 118 patients showed an overall correlation of 91.5% with the RAST results. Differences between the two test systems were observed in patients with a long interval between the test and the last allergic attack. None of the patients with proved penicillin allergy had atopic diseases or a history of atopy.On the basis of these findings it is suggested that the determination of both circulating specific IgE and total IgE should be of great value for diagnostic purposes during acute drug reactions and in the months immediately afterwards.

    Notes: Background: Interleukin (IL)-4 and IL-13 have been shown to be potent switch factors for IgE synthesis in human B cells.Objective: In this study we investigated the effects of recombinant human IL-4 and IL-13 on total and allergen specific IgE synthesis by peripheral blood inononuclear colls (PBMC) from pollen allergic patients and healthy control individuals.Methods: Peripheral blood mononuclear cells (PBMC) from allergic patients were investigated for their capacity to produce allergen specific IgE in vitro. Total protein extracts from birch pollen and timothy grass pollen as well as purified recombinant birch pollen allergens, Bet v I, birch profiling (Bet v II) and recombinant timothy grass pollen allergens. Phi p I, Phi p II, and Phi p V were used to measure specific IgE-antibody synthesis in cell culture supernatants by IgE-immunoblot and ELISA. Reults PBMC obtained from allergic patients spontaneously secreted allergen specific IgE in the culture supernatants. Addition of Interleukin 4, Interleukin 13 and anti-CD40 antibody to the cultures alone or in combinations significantly induced total IgE production whereas allergen specific IgE production was not affected.Conclusion: Our results indicate that the peripheral blood of allergic individuals contains long lived allergen specific B cells which have already switched to IgE production and which are not sensitive to IL-4 and IL-13 treatment. These results may have implications on attempts to use cytokines or cytokine antagonists in therapy of Type I allergy.
