    Description / Table of Contents: Zusammenfassung Es wurden Anreicherungsmethoden für die penicillinresistenten Serratia-Stämme HY und CV/rc 3 entwickelt. 0,04 mg Colistin/ml Mi-Medium inaktiviert wachsende HY-Zellen auf ca. 10-2 in 8 Std, auxotrophe Zellen werden nicht getötet. Der Anteil auxotropher Zellen in einer Mischung mit Prototrophen wurde durch 8stündige Colistinbehandlung von 1% auf 35–70% gesteigert. Ähnliche Resultate wurden mit Flavomycin an CV/rc 3 gewonnen: 9stündige Behandlung mit 0,2 mg Flavomycin/ml Mi-Medium und anschließende Verdünnung in dest. H2O steigerten den Auxotrophenanteil von 6 auf 32%. Flavomycin erzeugt osmotisch sensible Sphäroplasten, Colistin dagegen nicht.
    Notes: Summary Enrichment methods — modified penicillin-techniques — were employed for Serratia marcescens HY and CV/rc 3. This was done because Serratia is highly resistant to penicillin. the incubation of HY cells under growth conditions in the presence of 0.04 mg colistin/ml for 8 hrs was found to be effective in inactivating the population to more than 10-2. Auxotrophic mutants under nongrowth conditions were not affected by the antibiotic. The mutant yield of a mutagenized HY culture (NG for 15 min; nearly 80% survival) was increased from 1% to 37–70% after 8 hrs of colistin treatment. Spheroplast formation was not caused by the enrichment procedure. Nearly the same enrichment method was elaborated for CV/rc 3 with flavomycin, a new antibiotic produced by some strains of Streptomyces, which interacts with cell wall biosynthesis: 9 hrs of incubation in minimal-medium with 0.2 mg flavomycin/ml and following dilution in dest. H2O rose the mutant yield from 6% to 32% after NG treatment of CV/rc 3. Dilution in buffer-saline after flavomycin treatment was less effective because the spheroplasts produced by flavomycin are osmotically fragile. Four monoauxotrophic HY mutants were tested for selective enrichment after colistin treatment. Selection could not be recognized. Some experiments which are only preliminary show that selection may occur after flavomycin treatment.

CV/rc3 and E. coli Harvard the frequency of spontaneous auxotrophic mutants of many different types is high enough (0,3 and 1%) in a culture for isolating many mutants by simple replica plating without selective enrichment. By platings from series of parallel cultures in complete medium the distribution of the frequencies of the different spontaneous mutation types (spontaneous “spectrum of auxotrophs”) was obtained. They are different in both organisms (Table). When cells of Serratia grown on minimal medium were incubated for $$1{\raise0.5ex\hbox{$\scriptstyle 1$}\kern-0.1em/\kern-0.15em\lower0.25ex\hbox{$\scriptstyle 2$}}$$ -2 generations in complete medium and then heated for $$7{\raise0.7ex\hbox{$1$} \!\mathord{\left/ {\vphantom {1 2}}\right.\kern-\nulldelimiterspace}\!\lower0.7ex\hbox{$2$}}$$ min at 52°C to a survival of 10-5 about 0,45% auxotrophic mutations are induced. The type spectrum was significantly different from the spontaneous one due to a relatively lower frequency of the types “met, cys”, “leu”, “poly”, and “val+ile” and a higher frequency of “arg” “hyx”, “thia” and perhaps “ad, hyx” (Table). When E. coli H. was treated in the same way, but at 58°C, to a survival of 10-5 no auxotrophic mutations were induced at all. It is concluded from the findings that heat agitation is very probably not the main cause of spontaneous mutations in E. coli H., and further that in Serratia either the same is true for several mutation types or that thermic agitation produces mutations not by a single mechanism (e.g. splitting of guanine from DNA) but by at least 3 reaction types with different Q 10.-From Serratia 3 new types of “jump-mutations” are isolated mutating forth and back spontaneously from one auxotrophic state to an other one (met⇄his; ad⇄β-ala; arg⇄met). In E. coli none was found.

CV/rc3 und bei E. coli Harvard ist die Häufigkeit spontaner Auxotrophiemutanten vieler verschiedener Typen hoch genug in einer Kultur (0,3 bzw. 1%), so daß große Zahlen von Mutanten ohne selektive Anreicherung durch bloßes Stempeln gewonnen werden können. Durch Ausplattungen aus Serien von Parallelkulturen in Komplettmedium wurde so die Häufigkeitsverteilung der verschiedenen Spontanmutantentypen (spontanes Spektrum der Auxotrophen) erhalten. Diese Spektren sind verschieden in beiden Organismen. Wenn auf Minimalagar gewachsene Zellen von Serratia zunächst für $$1{\raise0.5ex\hbox{$\scriptstyle 1$}\kern-0.1em/\kern-0.15em\lower0.25ex\hbox{$\scriptstyle 2$}}$$ -2 Generationen in Komplettmedium bebrütet und dann für $$7{\raise0.7ex\hbox{$1$} \!\mathord{\left/ {\vphantom {1 2}}\right.\kern-\nulldelimiterspace}\!\lower0.7ex\hbox{$2$}}$$ min auf 52°C erwärmt wurden (Überlebensrate 10-5), so wurden 0,45% Auxotrophiemutationen induziert. Dies Typenspektrum war signifikant unterschieden vom spontanen, wofür die geringere Häufigkeit der Typen “met, cys”, “leu”, “poly” und “val+ile” sowie die größere von “arg”, “hyx”, “thia” und wahrscheinlich auch “ad, hyx” verantwortlich war. Wenn E. coli H. ebenso behandelt wurde, jedoch bei 58°C, wurden bei der Überlebensrate 10-5 gar keine Auxotrophiemutationen induziert. Es wird aus den Befunden geschlossen, daß Wärmebewegungen wahrscheinlich nicht die Hauptursache der Spontanmutabilität von E. coli H. ist und daß bei Serratia entweder dasselbe für wenigstens einige Mutationstypen gilt oder daß Wärmebewegung nicht durch einen einzigen Mechanismus (z. B. Guaninabspaltung aus DNS) Mutationen erzeugt, sondern durch mindestens drei mit verschiedenen Wärmekoeffizienten Q 10.-Bei Serratia wurden drei neue Typen “Sprungmutationen” isoliert, die spontan zwischen zwei verschiedenen Auxotrophien hin- und hermutieren (met⇄his; ad⇄β-ala; arg⇄met). Bei E. coli H. wurden keine gefunden.

    Description / Table of Contents: Summary The “photoproduct” after illumination of UV-irradiated Cytidylic acid (CMP) reported by Park and Kaplan earlier could not be reproduced by the 2nd author. Both, postincubation with and without light at about 25°C led to the formation of obviously the same substance with the absorption spectrum of normal CMP.

Kaplan berichtet worden war, konnte in neuen Versuchen des zweiten Autors nicht wiedergefunden werden. Nachbehandlung mit Licht wie auch Dunkelheit bei ca. 25°C ergaben offenbar die gleiche Substanz mit dem Absorptionsspektrum von normalem CMP.

    Notes: Summary A study presenting findings of focal hippocampal x-irradiation in early infancy and late behavior at two ages was carried out using Long-Evans hooded rats (Rattus norvegicus). Animals from eleven litters received 150-R irradiation treatments for 5 or 15 consecutive days and were then tested in an open field situation and in a two-way active avoidance apparatus. The results indicated that the behavioral deficits paralleled those seen in animals with classical hippocampal lesions. The results were offered in support of a response suppression model for hippocampal functioning.

    Notes: A cutaneous ciliated structure from the lower neck of a 61-year-old man having seromucous, sebaceous and respiratory-type differentiation is reported. The finding of continuity between endodermal and ectodermal structures in the reported lesion suggests an origin from the branchial apparatus. Many lesions with similar appearance have been labeled bronchogenic though these, in fact, may have branchial origins. The term cutaneous bronchogenic cyst, implying an origin from the tracheobronchial tree, maybe misleading and should be reconsidered for those lesions with respiratory-type differentiation in the lower neck.
