    Notes: Summary A highly sensitive enzyme immunoassay for arginine vasopressin (AVP) was described. The enzyme used was β-D-galactosidase fromEscherichia coli, and it was coupled to AVP by using the N-hydroxysuccinimide ester of N-(4-carboxycyclohexylmethyl)-maleimide. The complex was then utilized for the enzyme immunoassay, which could detect 0.5 fmoles (or 0.5 pg) of AVP in the assay tube.

    Notes: Summary To elucidate the localization of the subunit C of aldolase (aldolase C) in peripheral neuroendocrine cells, we made an immunohistochemical study with monospecific antibodies against human aldolase C. Aldolase C was found to be localized in various types of neuroendocrine cells; in the pituitary gland, thyroid, pancreas, adrenal gland, bronchus, and gastrointestinal tract.

    Notes: Abstract Aims/hypothesis. Although increased polyol pathway activity has been implicated in the pathogenesis of diabetic microangiopathy, the relation with diabetic macroangiopathy remains unclear. Galactose feeding is known to stimulate the polyol pathway and to develop abnormalites similar to those in diabetic microangiopathy. Our study was conducted to investigate whether an activation of polyol pathway by long-term treatment with galactose produced morphological changes in coronary arteries of dogs and the effect of an aldose reductase inhibitor, epalrestat, was also studied.¶Methods. Dogs received either normal chow or chow containing 30 % galactose with or without epalrestat given orally (20 or 50 mg · kg–1). After 44 months, morphometric analyses of coronary arteries were carried out and the galactitol contents in aortas were measured.¶Results. The ratio of areas of the intimal layer to those of the medial layer, an indicator of intimal thickening, was statistically significantly increased in galactose-fed dogs compared with control dogs. Galactose-fed dogs had a remarkable accumulation of galactitol in their aortas. These morphological and biochemical deficits were reduced by treatment with epalrestat.¶Conclusion/interpretation. This report morphologically shows diabetes-like macrovascular abnormalities in galactosaemic animals, suggesting that polyol pathway hyperactivity is closely related to the development of diabetic macroangiopathy, which could be prevented by aldose reductase inhibition.     Notes: Summary The effect of dibutyryladenosine 3′,5′-cyclic monophosphate (dibutyryl cAMP) on the expression of glutathione S-transferase placental type (GST-P) was examined in rat glioma cell line using an immunohistochemical technique. Cultured T9 glioma cells were negative for GST-P activity under normal conditions. However, treatment with 1 mM dibutyryl cAMP produced GST-P expression in about 50% of the cells, as well as some morphological changes. The expression of GST-P was increased with addition of dibutyryl cAMP together with 1 μg/ml allyl isothiocyanate (AITC) or 0.1 μg/ml benzyl isothiocyanate (BITC). With these combinated treatments, almost all cultured cells showed a strong positive reaction for GST-P, although AITC or BITC alone elicited GST-P in only 5% of the cultured cells. The results of the present study indicate that dibutyryl cAMP causes functional as well as morphological differentiation of T9 glioma cells.

    Notes: Abstract  Analytical instruments used for measurements of air and water pollution are calibrated by using reference materials such as standard gases and standard solutions. In Japan, since the middle of the 1970s, those reference materials which are traceable to the national standards maintained at national research institutes have been supplied to users by reference material producers. In order to establish the primary standards and to secure the traceability from the working standards to the national ones, various analytical methods such as coulometric, titrimetric and gravimetric analyses for purity determination and highly sensitive atomic spectrometry for trace analysis have been developed as the primary methods and reference methods. The Japanese Measurement Law, revised in 1992, has introduced a new traceability system in which a public organization, a "designated calibration body", can also prepare and maintain the national standards under the advice and instruction of national research institutes. The designated calibration body can provide calibration services to reference material producers (accredited calibration bodies) by using the national standards. The reference materials supplied in conformity with the traceability system include standard gases, pH standard solutions, metal standard solutions and non-metal ion standard solutions.

    Notes: Summary We have demonstrated the histochemical fiber types of human skeletal muscle in paraffin sections by the immunohistochemical method, together with compatible observations previously made in frozen sections that carbonic anhydrase III is mainly localized in type 1 fibers (Shima et al. 1983), and muscle-specific enolase in type 2 fibers (Ibi et al. 1983). This method is useful to analyze the fiber types when frozen muscle samples at biopsy or autopsy cannot be obtained.

    Notes: Abstract A sensitive analytical method was established for the determination of Th and U in activated concrete samples. The method combines an anion-exchange separation step with an ICP-MS determination technique. In the ICP-MS measurement, a few μg mL–1 of Al and Ca, a few ng mL–1 of Mn, La, Ce, Nd and Pb and pg mL–1 amounts of Li, Zr, Nb and Ba coexisting in the anion-exchange fraction of Th and U did not interfere. No adverse interference effects were observed in real sample analyses. The obtained detection limits (3σ, n = 10) of Th and U were 2.3 and 1.8 pg mL–1, respectively. The analytical precisions for ca. 5 μg g–1 Th and ca. 1 μg g–1 U in real activated concrete samples were equally less than 7% RSD. The accuracies obtained by the analysis of GSJ rock standard samples were –18.1 to 0.4% for the Th determination and –14.0 to –5.7% for the U determination. The method uses the conventional absolute calibration curve. The internal standard calibration is unnecessary.

    Notes: Abstract To define the effect of cyproterone acetate (CPA) on statural growth, 25 girls with idiopathic precocious puberty who had been treated with CPA were studied retrospectively. The final height SDS was −1.12±1.16 (mean±SD). The daily CPA dose was negatively related to the final height SDS. We divided our subjects into two groups according to the daily CPA dose [low dose, 84.9±15.4 mg/m2 (n=19) vs high dose, 135.8±17.1 mg/m2 (n=6)]. In the low dose group, the difference of the final height SDS minus height SDS for bone age at the initiation of CPA treatment was 0.55±1.16 and final height SDS was −0.82±1.05. The final height was not significantly different from the target height in the low dose group subjects whose target heights were obtained. Since the increment of height age to the increment of bone age during the treatment was significantly less in the group needing and treated with high dose CPA, high doses of CPA may reduce growth velocity more than its suppressive effect on bone maturation. These results suggest that CPA has an effect on statural growth in girls whose clinical symptoms can be controlled with a low dose of CPA, although they have not been compared with final height in untreated Japanese girls.

    Notes: In the Fock–Bargmann space, the complete harmonic-oscillator basis for the three-cluster system is constructed. The indices of the reduction U(6)&supuline;U(2)×U(3) are chosen as the quantum numbers. The Pauli-forbidden states are eliminated by the orthogonal transformation of basis functions. The basis states are obtained in terms of hypergeometric functions and the spherical Wigner functions. Their simple form allows one to solve the problem of calculating the matrix elements of the microscopic Hamiltonian needed for the study of three-cluster systems within the algebraic version of RGM. © 1995 American Institute of Physics.

    Notes: Abstract Three colonial variants of Actinobacillus actinomycetemcomitans, which formed transparent rough (TR)-, transparent smooth (TS)-, and opaque smooth (OS)-surfaced colonies, were described in relation to their fimbriation. TR- and TS-cells were adhesive to agar and glass surfaces but not the OS-cells. The examination by electron microscopy revealed that TR-cells were highly fimbriated but not TS- and OS-cells. Thus, TS-cells seemed to be an intermediate type. The fimbriae were isolated from TR-cells by suspending in 0.15 M ethanolamine-HCl buffer (pH 10.5) and purified by dissolving non-fimbrial components in 0.5% deoxycholate and 0.7% n-octyl-β-d-glucopyranoside. The relative molecular mass of the fimbrial subunit protein was 54 000.
