    Notes: Summary It has been suggested, based on animal experiments and limited human data, that the antihypertensive drug hydralazine might be carcinogenic, and among the sites of concern are the lung and colon. To assess the possible relationship between the use of hydralazine and lung and colorectal cancers in humans, we compared 1006 cases of lung cancer with 3531 hospital control subjects, and 972 cases of colorectal cancer with 3276 controls. Data were collected by interview in hospitals in the United States and Canada. Overall, 1.1% of the lung cancer cases, 1.6% of the colorectal cancer cases, and 1.5% of the controls had used hydralazine. Compared with those who had never used hydralazine, the relative risk estimate of lung cancer for those who first took the drug at least 18 months before hospital admission was 1.1 (95% confidence interval 0.4–2.9). The estimate for use for at least 1 year was 1.4 (0.5–3.8) and for use for at least 5 years the estimate was 0.9 (0.2–4.3). The corresponding relative risk estimates for colorectal cancer were 1.2 (0.5–2.5) for any use, 1.7 (0.8–3.7) for use for at least one year, and 2.4 (0.8–6.9) for five or more years of use. Other antihypertensive treatments and risk factors, including cigarette smoking in the analysis of lung cancer, were taken into account in these estimates. Although the effect of use after long latent intervals could not be evaluated, the results provide no support for the hypothesis that hydralazine use increases the risk of lung cancer. There is also no evidence that hydralazine increases the risk of colorectal cancer, but an effect after extended durations of use cannot be ruled out.

The present study examined the effect of systemic administration of lipopolysaccharide (LPS; 100 and 250 μg/kg, i.p.) on tyrosine hydroxylase (TH) and tryptophan hydroxylase (TPH) activities in frontal cortex, striatum and midbrain of the rat. Enzyme activities were determined by measuring accumulation of the transient intermediates 5-hydroxytrptophan (5-HTP) and L-dihydroxyphenylalanine (L-DOPA) following in vivo administration of the decarboxylase inhibitor, NSD 1015. TPH activity was increased 2 hours after administration of LPS (100 and 250 μg/kg) in both frontal cortex and midbrain, and a secondary increase was seen in the midbrain 12 hours after challenge. LPS provoked an increase in TH activity in the midbrain only, and this was evident for up to 24 hours after LPS administration. Thus in addition to previous studies demonstrating that LPS increases in vivo NA, DA and 5-HT release, this study shows that LPS increases the activity of the rate-limiting enzymes responsible for their synthesis.

Changes to the binding properties of cortical N-methyl-D-aspartic acid (NMDA) and beta-adrenergic receptors have both been reported as potential indicators of antidepressant activity. In the present investigation we examined the effects of the noradrenaline reuptake inhibitor, reboxetine, the serotonin reuptake inhibitor, sertraline, alone and in combination on the binding properties of cortical NMDA receptors and cortical β1-adrenoceptors following 14 days of treatment in the olfactory bulbectomized rat model of depression. A decrease in the potency of glycine to displace the strychnine insensitive glycine antagonist [3H] 5,7 dichlorokynurenic acid (5,7 DCKA) was observed in cortical homogenates of OB rats when compared to sham-operated controls. Similarly, treatment with the combination of reboxetine and sertraline for 14 days produced a decrease in the potency of glycine when compared to vehicle treated controls. By contrast neither olfactory bulbectomy or drug treatment significantly altered basal or glycine enhanced binding of the non-competitive NMDA antagonist [3H] MK-801 in cortical homogenates. Reboxetine alone, and in combination with sertraline, down-regulated [3H]-CGP 12177 (a selective β-adrenoceptor antagonist) binding in both OB and sham-operated animals. The lack of a bulbectomy effect in the [3H] CGP-12177 binding assay, and the fact that olfactory bulbectomy and antidepressant treatments produce a similar change to the potency of glycine at the NMDA receptor, suggests that these tests do not provide a neurochemical marker for either the behavioral hyperactivity deficit or antidepressant response in the model.

    Notes: Summary The general objective of this study was to evaluate the risks of agranulocytosis and aplastic anemia in relation to drug use. Other potential risk factors, including history of infectious mononucleosis, were also evaluated. In an international populationbased case-control study, cases of agranulocytosis and aplastic anemia and controls, who were patients with selected diagnoses, were identified from the same hospitals. Information on drug use was obtained in a structured interview. Limited information on environmental exposures and on selected items of medical history, including infectious mononucleosis, was also recorded. The possible effects of these factors on the risk of the two blood dyscrasias were evaluated. Relative risks were estimated by stratified analysis and by multiple logistic regression. Among 319 cases of arganulocytosis, 12 patients (4%) gave a history of infectious mononucleosis at least 1 year previously; among 2180 controls the corresponding number was 11 (0.5%) (multivariate relative risk estimate, 6.2; 95% confidence interval, 2.3–17). A nonsignificant elevation in the relative risk was observed for aplastic anemia. The data suggest that infectious mononucleosis may confer a long-term increase in the risk of agranulocytosis. However, the association was an unexpected one, identified in the course of multiple comparisons, and it must be independently confirmed.

    Description / Table of Contents: Zusammenfassung Die Technik des „kontrollierten Rundens“ (Controlled Rounding) wird von Behörden wie dem United States Bureau of Census benutzt, um in mehrdimensionalen Tabellen oder Matrizen erfaßte statistische Daten durch Approximation so zu verändern, daß sowohl die Anonymität von Einzeldaten (Matrixeinträgen) als auch die Integrität der Gesamtdaten (Zeilensummen, Spaltensummen etc.) gewährleistet ist. In dieser Arbeit behandeln wir die Komplexität und Lösbarkeit des dreidimensionalen Rundungsproblems und diskutieren eine Hierarchie von Relaxationen, die bei Nichtlösbarkeit des Ausgangsproblems alternative, nahezu zulässige Lösungen liefern.
    Notes: Summary Controlled rounding is a procedure that perturbs tabular data collected from respondents in such a way as to preserve the anonymity of the respondents while maintaining the integrity of the data. Controlled rounding techniques are regularly used by the United States Bureau of the Census and its counterparts in other countries. This paper discusses the complexity of the three-dimensional controlled rounding problem. In particular, the three-dimensional, zero-restricted controlled rounding problem is shown to be NP-complete. As zero-restricted controlled roundings may fail to exist, various relaxations of this basic rounding problem have been defined. The paper introduces a sequence of such relaxations and proceeds to address the existence of solutions and complexity issues for the relaxed problems.
