Summary 1. Diphenylhydantoin, 5,5-Diphenylhydantoin-3-sodiumacetat, 5,5-Dimethyl-dithiohydantoin, 3-Methylhydantoin and 5,5-Dimethyl-3-nitrohydantoin abolish the 2,4-dichlorphenoxyacetate induced myotonia in a directly stimulated rat diaphragm. Thus has been proved a direct effect of these substances on the muscle-cells. 2. Thiohydantoin and 2-Imino-thiazolidon-4 have no antimyotone effect. 3. With Calcium-intake in the incubation medium we need less of the substances mentioned under 1. The cause of the disappearance of myotonia through the derivates of hydantoin is a lessening of the irritability of the muscle cell membrane system, because more sodium is released from the muscle cells. The mechanism of the release of sodium is not completely understood.

Zusammenfassung 1. Diphenylhydantoin (Zentropil®), 5,5-Diphenylhydantoin-3-essigsaures Natrium, 5,5-Dimethyl-dithiohydantoin, 3-Methylhydantoin und 5,5-Dimethyl-3-nitrohydantoin beseitigen die experimentelle 2,4-D Myotonie am direkt stimulierten Rattenzwerchfellpräparat dosisabhängig. Damit ist eine direkte Wirkung dieser Substanzen auf die Muskelzelle bewiesen worden. 2. Thiohydantoin und 2-Imino-thiazolidin-4 zeigen keinen antimyotonen Effekt. 3. Calciumzugabe zum Inkubationsmedium hat eine dosissparende Wirkung. Die Beseitigung der Myotonie durch Hydantoinderivate kommt am ehesten durch eine Verminderung der Irritabilität des Muskelzellmembransystems infolge verstärkter Auswärtsbewegung des Natriums zustande. Die Mechanismen, die den Auswärtsfluß des Natriums hervorrufen, sind im einzelnen nicht bekannt.
