    Notes: Linkage maps of perennial ryegrass were constructed from F2 and BC1-type populations using, predominantly, restriction fragment length polymorphism data based on heterologous probes used in mapping other grass species. The maps identified seven linkage groups, which covered a total of 515 cM (F2) and 565 cM (BC1). They were aligned using 38 loci identified in both populations (common loci) and a possible marker order for all mapped loci in either population was identified in an integrated map. The estimated recombination frequencies and map distances between adjacent common loci were compared between the two data sets and regions of heterogeneity identified. Overall, the common markers identified a map distance of 446 cM in the F2 population and 327 cM in the BC1 population, reflecting a higher recombination frequency in the former, although the difference was not evenly spread over the seven linkage groups.

    Notes: Salt-affected soil is a major world-wide problem with many hectares of land lost to cultivation each year .To combat this problem, the development and assessment of a novel salt tolerant cereal, tritipyrum, was carried out. This is a hybrid between wheat and Thinopyrum bessarabicum. a very salt-tolerant member of the Triticeae.A range of tritipyrums derived from crosses between tetraploid wheat, Triticum durum, and Th. bessarabicum was produced. Although meiosis in ihe tritipyrums was generally regular, chromosome pairing failure was observed in each of the genotypes. In addition, the level of fertility was relatively low, the fertility of bagged spikes ranging from 29% to 51%. One iritipyrum produced multiple seeds in some florets. Three tritipyrums selected at random performed better than their wheat parents in hydroculture experiments in each of three treatments (150, 200 and 250 mol/ m3 Nacl). The potential for the exploitation of tritipyrum in salt-affected soils is discussed.

    Notes: Lines of Triticum aestivum cv. Chinese Spring carrying an additional chromosome 5Eb from Thinopyrum bessarabicum or having chromosome 5A or 5D replaced by chromosome 5Eb were screened in hydroculture for tolerance to salt. The previously reported tolerance of the 5Eb addition line was confirmed and the two substitution lines were shown to have a higher level of survival in 175 mol/m3 NaCl than both the addition and the ‘Chinese Spring’ parent. Reasons for the better tolerance of the substitutions are discussed.

    Notes: Abstract Genomic in-situ hybridization (GISH) was used to determine the amount of wheat-rye chromosome pairing in wheat (Triticum aestivum) x rye (Secale cereale) hybrids having chromosome 5B present, absent, or replaced by an extra dose of chromosome 5D. The levels of overall chromosome pairing were similar to those reported earlier but the levels of wheat-rye pairing were higher than earlier determinations using C-banding. Significant differences in chromosome pairing were found between the three genotypes studied. Both of the chromosome-5B-deficient hybrid genotypes showed much higher pairing than the euploid wheat hybrid. However, the 5B-deficient hybrid carrying an extra chromosome 5D had significantly less wheat-rye pairing than the simple 5B-deficient genotype, indicating the presence of a suppressing factor on chromosome 5D. Non-homologous/non-homoeologous chromosome pairing was observed in all three hybrid genotypes. The value of GISH for assessing the level of wheat-alien chromosome pairing in wheat/alien hybrids and the effectiveness of wheat genotypes that affect homoeologous chromosome pairing is demonstrated.

    Notes: Abstract Two novel 46-chromosome doubled haploid lines, W66 and M17, derived from separate hexaploid triticale x bread wheat crosses, were characterised using cytological and biochemical markers. Both lines were shown to be relatively stable cytologically, over 11 and 8 generations of selfing, respectively. By examining mitotic and meiotic chromosomes, the stabilities of the two lines were shown to be similar with frequencies of 2n=46 in 74.2–85.5% of cells. However, over selfed generations, the rye chromosomes were shown to have lost some of their heterochromatin, which made it difficult to establish their continued presence using cytological techniques, such as C-banding alone. Cytological evidence from pairing studies, C-banding, and fluorescence in-situ hybridization, showed that both M17 and W66 are wheat/rye multi-addition lines with rye chromosome constitutions of 1R+6R, and 1R+4R, respectively. These conclusions were confirmed by isozyme and storage-protein analysis.

    Notes: Summary Genomic in situ hybridization was used to identify alien chromatin in chromosome spreads of wheat, Triticum aestivum L., lines incorporating chromosomes from Leymus multicaulis (Kar. and Kir.) Tzvelev and Thinopyrum bessarabicum (Savul. and Rayss) Löve, and chromosome arms from Hordeum chilense Roem. and Schult, H. vulgare L. and Secale cereale L. Total genomic DNA from the introgressed alien species was used as a probe, together with excess amounts of unlabelled blocking DNA from wheat, for DNA:DNA in-situ hybridization. The method labelled the alien chromatin yellow-green, while the wheat chromosomes showed only the orange-red fluorescence of the DNA counterstain. Nuclei were screened from seedling root-tips (including those from half-grains) and anther wall tissue. The genomic probing method identified alien chromosomes and chromosome arms and allowed counting in nuclei at all stages of the cell cycle, so complete metaphases were not needed. At prophase or interphase, two labelled domains were visible in most nuclei from disomic lines, while only one labelled domain was visible in monosomic lines. At metaphase, direct visualization of the morphology of the alien chromosome or chromosome segment was possible and allowed identification of the relationship of the alien chromatin to the wheat chromosomes. The genomic in-situ hybridization method is fast, sensitive, accurate and informative. Hence it is likely to be of great value for both cytogenetic analysis and in plant breeding programmes.

    Notes: Summary The transmission of chromosome 4S l from Aegilops sharonensis was observed in a range of wheat genetic backgrounds. Chromosome 4S l was transmitted at a very high frequency (at least 97.8%) in all crosses. The genetic background appears to only have a small effect on transmission. The frequency of transmission of chromosome 4S l was the same in each genetic background through both the male and female gametes.

    Notes: Abstract Ten random amplified polymorphic DNA (RAPD) markers specific to chromosome 5Eb of Thinopyrum bessarabicum were detected. Genomic in situ hybridization and standard cytological observations revealed that six of the markers are located on the 5Eb short arm and four are located on the 5Eb long arm. These RAPD markers have been used to confirm the identity of putative 5Eb (5A) and 5Eb (5D) substitution individuals. The potential of RAPDs for the detection of wheat/alien recombinants is discussed.

    Notes: Summary Wheat plants nullisomic for chromosome 4B are male sterile due to the absence of the male fertility gene Ms1. However, plants in which chromosome 4B has been substituted by the preferentially transmitted chromosome 4S1 of Ae. sharonensis are male fertile due to the compensating effect of Ms4 on the alien chromosome. This substitution line has been mutated and three recessive mutation of Ms4 have been selected. Plants homozygous for these mutations are male sterile. The implication of these mutations for hybrid wheat production is discussed.
