    Notes: Abstract Canola meal was used in channel catfish Ictalurus punctatus diets at levels of 0, 15.4, 30.8, 46.2 and 61.6%, by progressively replacing (on an equal nitrogen basis) 0, 25, 50, 75, or 100% of solvent-extracted soybean meal in the control diet. The feeds were formulated to contain approximately 29% crude protein and 2,650 kcal of digestible energy/kg on an air-dry basis. Each diet was fed to juvenile channel catfish to satiation twice daily for 10 wk. Fish fed the diets containing the two lowest levels of canola meal (15.4 and 30.8%) had similar weight gains, feed intakes, feed utilization efficiencies, and percent survivals relative to the group fed the control diet. Weight gains and feed intakes declined significantly as the dietary levels of canola meal were increased to 46.2% or higher, probably because of reductions in diet palatability and some impairment of feed utilization due to the presence of increased levels of antinutritional factors, particularly glucosinolates. Whole body percentages for moisture and crude protein were unaffected by the dietary treatments. Body ash contents, however, were lowest for fish fed the control diets but were essentially the same for fish fed the other diets. Fish fed the diet containing 30.8% canola meal had lowest body fat content but this effect may not have been diet related. Values for red blood cell concentration, hemoglobin, mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration were not affected by dietary canola meal level, but hematocrit was higher (although not always significant) for fish fed the control diet. The results of this study suggest that canola meal can comprise about 31% of the diet of channel catfish by replacing half of the amount of soybean meal used in the control diet without adversely affecting growth or any other aspect of performance.

    Notes: Juveniles from all possible crosses among USDA 102. USDA 103, and Norris channel catfish Ictalurus punctatus strains were compared for: 1) survival and mix-Edwardsiella ictaluri antibody after exposure to live E. ictaluri bacterium (isolate 597-458); 2) antibody level after injection with formalin-killed E. ictaluri (597-458); and 3) pre-stress. post-stress, and stress-recovery serum Cortisol levels. Purebred and crossbred USDA 102 strain fish had higher survival (mean of five genetic groups = 87%) and lower anti- E.ictaluri antibody (mean optical density (OD) of five genetic groups = 0.167) 30 d after live E. ictaluri challenge than purebred Norris and USDA 103 strains and their crosses (means of four genetic groups = 60% survival and 0.210 OD antibody level). Significant general combining ability, line effects, and heterosis indicated that the USDA 102 strain contributed additive and dominance genetic effects for increased survival and lower antibody level after live E. ictaluri challenge. Antibody response to formalin-killed, intra-peritoneally injected E. ictaluri was not different among genetic groups (overall mean = 0.198 OD). Serum Cortisol was measured prior to (pre-stress), immediately after (post-stress), and 2 h after (stress-recovery) a standard stressor. Serum Cortisol level was highest in post-stress fish (35.8 ng/mL), intermediate in stress-recovery fish (10.9 ng/mL), and lowest in pre-stress fish (6.5 ng/mL), but was not different among genetic groups within a stress time period. Results indicate diat differences exist among genetic groups of channel catfish for survival and antibody production after live E. ictaluri challenge, but these differences were not related to antibody response to killed E. ictaluri or serum Cortisol levels.

    Notes: The dietary iron requirement for normal growth and optimum hematological values and bioavailability was determined for channel catfish Ictalurus punctatus fingerlings using egg-white based diets supplemented with 0,5,10,20,60, and 180-mg iron/kg from iron methionine or 20, 60, and 180-mg iron/kg from iron sulfate. The basal diet which contained 9.2-mg iron/kg, 34% crude protein and 3.1 kcal of digestible energy/g was fed to channel catfish fingerlings (8.5 g) in triplicate flow-through aquariums to satiation twice daily for 8 wk. Fish fed the basal diet without iron supplementation exhibited poor growth throughout the 8-wk period. Fish fed iron-supplemented diets did not differ with regard to final weight gain. Survival, feed conversion, total blood cell count, mean corpuscular hemoglobin concentration, serum iron, total iron binding capacity, and transferrin saturation were not significantly affected by dietary iron level. Hemoglobin, hematocrit, mean corpuscular hemoglobin, and mean corpuscular volume were significantly lower in fish fed the basal diet. These values were also consistently lower for fish fed diets with 5 and 10-mg iron/kg from iron methionine. However, differences were not always significant. Results of this study indicate that supplementation of 5-mg iron from iron methionine was sufficient for growth. However, a supplemental iron level of 20-mg/kg diet or a total iron level of 30-mg/kg of diet appeared to be needed for optimum hematological values. Iron methionine and iron sulfate were equally effective in preventing anemia in channel catfish.

    Notes: Channel catfish Ictalurus punctatus fingerlings were fed purified diets supplemented with iron at levels of 0, 20, 60, and 180 mg/kg from iron sulfate (FeS) or 5, 10, 20, 60, and 180 mg/kg from iron methionine (FeM) in triplicate tanks for 8 wk. Fish were then divided into two groups and subjected to different assays to measure disease resistance and individual immune functions. Representative fish from each dietary treatment were challenged by bacterial immersion with virulent Edwardsiella ictaluri, and mortality due to enteric septicemia was recorded. Other fish were immunized with 0.2–mL formalin-killed E. ictaluri and boosted 21 d post-immunization. Antibody response was determined by FAST-ELISA. Chemiluminescent and chemotaxis assays were performed using peritoneal macrophages. Supplementation of the diet with various levels of iron from FeS or FeM did not significantly affect antibody production. Chemotactic migration by macrophages was depressed in iron-deficient fish and a level of 60 mgkg from either FeS or FeM provided the highest chemotactic indexes. A deficiency of dietary iron was found to increase mortality of channel caffish due to enteric septicemia of catfish (ESC). However, more studies should be conducted to better understand the effects of sources and levels of dietary iron on immune responses and disease resistance in channel caffish.

    Notes: Streptococcus agalactiae is a major bacterial pathogen that is the cause of serious economic losses in many species of freshwater, marine and estuarine fish worldwide. A highly efficacious S. agalactiae vaccine was developed using extracellular products (ECP) and formalin-killed whole cells of S. agalactiae. The vaccine efficacy following storage of S. agalactiae ECP and formalin-killed S. agalactiae cells at 4 °C for 1 year was determined. The stored ECP containing S. agalactiae formalin-killed cells failed to prevent morbidity and mortality among the vaccinated fish, and the relative percentage survival was 29. Serum antibody responses of the stored ECP and freshly prepared ECP against soluble whole cell extract of S. agalactiae indicated that significantly less antibody was produced in fish immunized with stored ECP and S. agalactiae cells than in those fish immunized with freshly prepared ECP and S. agalactiae cells at day 31 post-vaccination. Silver staining of sodium dodecyl sulphate-polyacrylamide gels and immunostaining of Western blots with tilapia antiserum to S. agalactiae revealed that predominant 54 and 55 kDa bands were present in the freshly prepared ECP fraction. The 55 kDa band was absent from the stored ECP and new bands below 54 kDa appeared on the Western blot. The results of this study on S. agalactiae ECP provide evidence for a correlation between protection and antibody production to ECP and for the importance of the 55 kDa ECP antigen for vaccine efficacy.

    Notes: This study explored the existence of apoptosis (programmed cell death) in Ichthyophthirius multifiliis Fouquet (Ich) theronts and determined the effect of cutaneous antibodies in skin culture fluid from fish immune to Ich on theront apoptosis. Apoptosis was detected in theronts and was clearly distinguished by fluorescent microscopy after staining with acridine orange and propidium iodide. The apoptotic theronts showed characteristic chromatin condensation and nuclear fragments containing chromatin pieces. The externalization of phosphatidylserine on the plasma membrane of apoptotic theronts was detected with fluorescein isothiocyanate-conjugated annexin using flow cytometry. Theront apoptosis was induced using the skin culture fluid from fish immune to Ich, which contained cutaneous antibodies against Ich. The highest apoptosis appeared in theronts exposed to immune skin culture fluid at a 1:10 dilution, compared with those at 1:20 and 1:40 dilutions. A direct correlation was noted between the percentage of apoptotic theronts and exposure duration to immune skin culture fluid. The study indicated that antibody reaction with theronts (immobilization) played an important role in theront apoptosis, but it could not be excluded that other components released from the excised skin had effects on theronts.

    Notes: The humoral immune responses and host protection of channel catfish, Ictalurus punctatus (Rafinesque), against Ichthyophthirius multifiliis (Ich) were determined after immunization with live theronts and sonicated trophonts. Immunizations with live theronts or sonicated trophonts were carried out by both bath immersion and intraperitoneal (i.p.) injection. Cutaneous and serum immunoglobulin (Ig) levels and anti-Ich antibodies were measured 12 and 21 days post-immunization. The level of Ich infection and survival of catfish were determined after theront challenge. Cutaneous and serum anti-Ich antibodies were significantly higher (P 〈 0.05) in fish immunized with live theronts by immersion or i.p. injection, or with sonicated trophonts administered by i.p. injection, than in fish immunized with sonicated trophonts by immersion, with bovine serum albumin by i.p. injection, or non-immunized controls. Host protection was noted only in fish immunized with live theronts by immersion or i.p. injection or with sonicated trophonts by i.p. injection. There was a positive correlation between higher levels of anti-Ich antibodies and host survival in the immunized fish.

    Notes: Withholding feed has been suggested as a strategy to manage infectious disease of channel catfish, Ictalurus punctatus (Rafinesque). In a previous study, we demonstrated that deprivation of feed for as little as 7 days reduced innate resistance of catfish to Flavobacterium columnare. This study was conducted to evaluate the effect of feeding regimens [no feeding (NF), fed once every other day to satiation (FEOD) and fed once daily to satiation (FD)] on organosomatic indices, physiological changes and susceptibility of channel catfish to F. columnare. Fish that were not fed for 2 and 4 weeks had a significant increase (P 〈 0.05) in gutted weight:wet weight ratio and decrease in other organosomatic indices [gut index (GI), mesenteric fat index (MFI) and hepatosomatic index (HSI)]. Haematology was not effected by feeding regimen except at week 4, when a significantly higher haemoglobin level was observed in the NF fish. Serum protein did not differ at week 2, but the level at week 4 of the NF fish (35.91 mg mL−1) was significantly lower than that of the FD fish (41.77 mg mL−1). Significantly lower (P 〈 0.05) blood glucose (39.5 and 40.3 mg dL−1) and liver glycogen (1.7 and 1.8 mg g−1) were seen in the NF fish at weeks 2 and 4, respectively, as compared with blood glucose and liver glycogen levels of FD fish (67.5 and 92.8 mg dL−1 and 46.5 and 52.6 mg g−1 at weeks 2 and 4, respectively) and FEOD (82.8 and 85.5 mg dL−1 and 45.1 and 51.4 mg g−1 at weeks 2 and 4, respectively). Mortality in the NF fish caused by F. columnare (78%) was significantly higher (P 〈 0.05) than mortality in the FD and FEOD treatments (0.0 and 1.7%, respectively). Blood glucose and liver glycogen showed the same trend of low values for NF fish following challenge (week 6). Blood glucose, liver glycogen, GI and HSI are sensitive indicators for channel catfish deprived of feed (NF) for 4 weeks. Blood glucose and liver glycogen levels around 40 mg dL−1 and 2 mg g−1, respectively, are indicative of starvation in juvenile channel catfish. Moreover, NF fish were susceptible to F. columnare infection. Thus, it is suggested that in the absence of natural food, juvenile channel catfish should be fed at least once every other day to apparent satiation to maintain normal physiological function and improve resistance to F. columnare.

    Notes: Streptococcus agalactiae was isolated from cultured gilthead seabream, Sparus auratus L., and diseased wild Klunzinger's mullet, Liza klunzingeri (Day), in Kuwait Bay, Arabian Gulf. Isolates were catalase negative, β-haemolytic, Gram-positive cocci and serogroup B. Experimental infectivity trials with mullet and seabream brain isolates in Nile tilapia, Oreochromis niloticus L., caused 100 and 90% mortality, respectively, within 7 days post-inoculation indicating virulent S. agalactiae as the bacterial pathogen responsible for the epizootic in Kuwait Bay.
