Intramolecular Photodealkylation of 10-Alkylisoalloxazines, a Model Reaction for Riboflavine PhotodegradationA number of 10-alkyl- and 10-cycloalkylisoalloxazines have been synthesized. On irradiation either anaerobically or aerobically the alkyl groups (except methyl, ethyl and neopentyl) are split off in a synchronous cyclic mechanism yielding alloxazine and an alkene or cycloalkene, resp. This photodealkylation proceeds aerobically via the first excited singlet state, anaerobically via singlet and triplet state in a ratio of 1:8. By comparison of the reaction rates a plane six-membered ring of the N(1)-C(10a)-N(10)-C(1′)-C(2′)- H(2′)-group can be shown to be the transition state; the quantum yields are correlated to the probability of occupying this state. - The results are compared to the photodegradation of riboflavine.
    Notes: Eine Anzahl 10-Alkyl-und 10-Cycloalkylisoalloxazine wird synthetisiert. Sowohl bei anaerober als auch aerober Bestrahlung werden in einem cyclischen Synchronmechanismus die Alkylgruppen, außer Methyl, Äthyl und Neopentyl, als Alken bzw. Cycloalken abgespalten, wobei Alloxazin entsteht. Aerob verläuft diese Photodealkylierung über den ersten angeregten Singulett-Zustand, anaerob über Singulett- und Triplett-Zustand im Verhältnis 1:8. Durch Vergleich der Reaktionsgeschwindigkeiten kann für den Übergangszustand ein ebener Sechsring der N(1)-C(10a)-N(10)-C(1′)-C(2′)- H(2′)-Gruppierung wahrscheinlich gemacht werden; die Quantenausbeuten sind proportional der Wahrscheinlichkeit, mit der diese Konformation eingenommen werden kann. - Der Photoabbau des Riboflavins wird mit diesen Ergebnissen verglichen.

On the other hand, 3-methylisoalloxazine-10-propanoic (2a) and 3,β-dimethylisoalloxazine-10-propanoic acid (2c) undergo an intramolecular photodecarboxylation and cyclisation of the N(10)-side-chain to N(1) in the pH-range of 2-6 to yield the 1,2,5,6-tetrahydro-4H,7H-benz[g]imidazo-[1,2,3-ij]pteridine-4,6-dione system (8) in a triplet reaction, which again confirms the photoactivity of the N(1)-position. In the pH-range 〉 6 the N(10)-substituent is split off via the singlet and triplet state in an intramolecular reaction yielding 3-methylallo? azzine and acrylic or crotonic acid, resp. - The mass-spectroscopic and thermal behaviour of these isoalloxazine-10-alkanoic acids is analyzed (McLafferty rearrangement and γ-hydrogen abstraction) and is compared to the photochemical reactivity.
    Notes: Während 7,8-Dimethylisoalloxazin-10-essigsäure (3a) in einer intermolekularen Triplett-Photo-reaktion zu 7,8-Dimethylalloxazin (“Lumichrom”), Kohlendioxid und Formaldehyd reagiert, entsteht bei der thermischen Zersetzung 7,8,10-Trimethylisoalloxazin (“Lumiflavin”). Hingegen reagieren 3-Methylisoalloxazin-10-propionsäure (2a) und 3,β-Dimethylisoalloxazin-10-propionsäure (2c) im pH-Bereich 2-6 intramolekular unter Photodecarboxylierung und Cyclisierung der N(10)-Seitenkette nach N(1); in dieser Triplett-Reaktion, die erneut die Photoaktivität der N(1)-Position bestätigt, entsteht das 1,2,5,6-Tetrahydro-4H, 7H-benz[g]imidazo[1,2,3-ij]pteridin-4,6-dion-System (8). Im pH-Bereich 〉 6 findet eine intramolekulare, vom Singulett- und Triplett-Zustand aus verlaufende photochemische Abspaltung des N(10)-Substituenten statt, wobei 3-Methylalloxazin und Acryl- bzw. Crotonsäure entstehen. - Das massenspektrometrische und thermische Verhalten der Isoalloxazin-10-alkancarbonsäuren wird analysiert (McLafferty-Umlagerung und γ-Wasserstoff-Abstraktion) und mit der Photoreaktivität verglichen.

    Description / Table of Contents: The Photochemistry of 10-Phenylisoalloxazine: Intramolecular Singlet and Intermolecular Triplet ReactionsFrom the first excited triplet state of 10- phenylisoalloxazine (2a) photoreduction or photoalkylation occurs in a way analogous to lumiflavin ( = 7,8,10-trimethylisoalloxazine) to yield 1,5-dihydroisoalloxazine or 5-and 4a-alkylated dihydro-derivatives thereof. - The first excited singlet state, however, undergoes photocyclization in a proton-catalyzed reaction, which can be described as an intramolecular, electrophilic photosubstitution of the N(1)-atom on the phenyl-substituent, yielding the new heterocyclic system of 5H-benzimidazo[1,2,3-ij]-benzo[g]pteridine-6,8(7H-dione (11), which is also formed on photocylization of 1,3-diphenylalloxazine (6). The oxidative degradation of 11 has been studied.
    Notes: 10-Phenylisoalloxazin (2a) reagiert im ersten angeregten Triplett-Zustand bei der Umsetzung mit verschiedenen Substraten analog dem Lumiflavin ( = 7,8,10-Trimethylisoalloxazin) unter Bildung des 1,5- Dihydroisoalloxazins bzw. 5- oder 4a-alkylierter Dihydro-Derivate. - Der erste angeregte Singulett-Zustand reagiert hingegen unter Photocyclisierung in einer protonen-katalysierten, Reaktion, die als intramolekulare, elektrophile Photosubstitution des N(1)-Atoms am Phenyl-Substituenten beschrieben werden kann. Dabei entsteht das heterocyclische System des 5H-Benzimidazo[1,2,3-ij]benzo[g]pteridin-6,8(7H)-dions (11), das auch durch Photocyclisierung des 1,3-Diphenylalloxazins (6) zugänglich ist. Der oxidative Abbau von 11 wird untersucht.

    Description / Table of Contents: Syntheses and Photoreductions of NitroisoalloxazinesNitration of 3,10-dimethylisoalloxazine (1b) yields the 7-nitro derivative 2b. Nitration of 3,7,10-trimethylisoalloxazine (1c) gives the 6- and 9-mononitro compounds in a ratio of 2c:2d = 97:3, from 3,7,8,10-tetramethylisoalloxazine, the 6- and 9-nitro and the 6,9-dinitro derivatives are obtained in a ratio of 2g:2h:2i = 76:8:1. Riboflavine (1h) yields only the 6-mononitro derivative 2k. - 8-Nitroisoalloxazine 2n is obtained from the 8-amino derivative 3a via the diazonium salt 4a. Mild nitration of 1,3,7,8-tetramethylalloxazine (5b) yields the 6- and 9-mononitro derivatives 5d and 5f, respectively. - On photoreduction of the 6- and 8-nitroisoalloxazines in the presence of ethylenediaminetetraacetic acid, the corresponding hydroxylamino derivatives 7b and a, respectively, are formed which can be reoxidized by hexacyanoferrate(III). Continued photoreduction gives the amino derivatives. 9-Nitroisoalloxazines 2h is photoreduced only to the 1,5-dihydro-9-nitro compound 6a, which is reoxidized by oxygen.
    Notes: Die Nitrierung von 3,10-Dimethylisoalloxazin (1b) ergibt das 7-Nitroderivat 2b. Aus 3,7,10-Trimethylisoalloxazin (1c) entstehen dabei das 6- und 9-Mononitroderivat im Verhältnis von 2c:2d = 97:3, aus 3,7,8,10-Tetramethylisoalloxazin (1f) das 6-, 9-Nitro- und das 6,9-Dinitroderivat im Verhältnis von 2g:2h:2i = 76:8:1. Riboflavin (1h) ergibt ausschließlich die 6-Mononitroverbindung 2k. - Die Synthese des 8-Nitroisoalloxazins 2n ist aus der 8-Aminoverbindung 3a über das Diazoniumsalz 4a möglich. Vorsichtige Nitrierung von 1,3,7,8-Tetramethylalloxazin (5b) ergibt ein Gemisch des 6- und 9-Mononitroderivats 5d bzw. 5f. - Bei der Photoreduktion der 6- und 8-Nitroisoalloxazinderivate in Gegenwart von Ethylendiamin-tetraessigsäure entstehen zunächst die entsprechenden Hydroxylaminoverbindungen 7a und b, die durch Hexacyanoferrat(III) zu den Nitroderivaten reoxidiert werden; fortgesetzte Photoreduktion ergibt die Aminoverbindungen. Das 9-Nitroisoalloxazin 2h wird nur zur 1,5-Dihydro-9-nitro-Stufe 6a photoreduziert, die durch Sauerstoff zur Ausgangsverbindung reoxidierbar ist.

    Description / Table of Contents: Pteridines, LXXI1).  -  Synthesis and Photochemical Behaviour of 8-Substituted LumazinesThe 8-substituted lumazines 34-69 have been synthesized from 6-chloro-5-nitrouracil (1) and its 3-methyl derivative 2 via the corresponding 6-amino-5-nitrouracils 3-26, substituted at the amino nitrogen. The photochemical properties of the 8-substituted lumazines have been investigated. Various photoreactions were observed depending on the chemical nature of the N-8 side-chain and leading to an easy photodealkylation of the β-hydroxy-, β-methoxy-, and β-aminoethyl derivatives 34-44. Increasing donor properties of the β-substituent enhances the photolabilty which is expressed in a Norrish-type II cleavage reaction to the corresponding lumazines 70-75. 8-Cycloalkyllumazines 52-54 show photoreduction to 7,8-dihydrolumazines 76-78. The newly synthesized 8-substituted lumazine derivatives have been characterized by elementary analyses and UV spectra. Structural peculiarities due to the various substituents in position 6, 7, and 8 will be discussed.
    Notes: Ausgehend von 6-Chlor-5-nitrouracil (1) und seinem 3-Methyl-Derivat 2 warden über die entsprechenden am Aminostickstoff substituierten 6-Amino-5-nitrouracile 3-26 die 8-substituierten Lumazine 34-69 dargestellt. Ihr photochemisches Verhalten wird untersucht. In Abhängigkeit von der chemischen Natur der N-8-Seitenkette laufen unterschiedliche Photoreaktionen ab von denen vor allem die leichte Photodealkylierung der β-Hydroxy-, β-Methoxy- und β-Aminoethyl-Derivate 34-44 herausragt. Mit zunehmender Donatoreigenschaft des β-Substituenten steigt auch die Photolabilität, die sich in einer Norrish-Typ-II-Spaltung zu den entsprechenden Lumazinen 70-75 kundtut. Die 8-Cycloalkyllumazine 52-54 erleiden eine Photoreduktion zu den 7,8-Dihydrolumazinen 76-78. Sämtliche neuen 8-substituierten Lumazine werden durch Analysen und UV-Spektren charakterisiert und strukturelle Besonderheiten, bedingt durch die verschiedenen Substituenten in 6-, 7- und 8-Stellung, diskutiert.

    Description / Table of Contents: Reductive Photoalkylation of the Flavin Nucleus; Structure and Reactivity of the PhotoproductsOn the basis of the manifold similarities between flavin photochemistry and flavin biocatalysis, we have made an effort toward systematic investigation of flavin-dependent photodehydrogenation: 3-benzyllumiflavin (1) reacts photochemically with carboxylic acids, e. g. 3-indoleacetic acid, phenoxyacetic acid and (tert-buty1thio)acetic acid, to give 4a-alkylated 4a,5-dihydro-flavin derivatives 3a-c and COz. In the presence of thiodiacetic acid and dithiodiacetic acid the 5-carboxymethyl-l,5-dihydroflavin derivative 4a and the 4a-carboxymethylthio derivative 3d, respectively, are formed.- Alkyl sulfides are attacked in the cr-position yielding 5-substituted 1,5-dihydroflavin derivatives, which undergo trans-elimination if P-CHz-groups are present; in the case of the thiolane the adduct 4d is found to be stable because of sterical hindrance. With benzaldehyde the 5-benzoyl-l,5-dihydro derivative 4e is formed. - Alkenes are attacked in the allylic position yielding 4a-alkylated 4a,5-dihydro derivatives, 3e - g. In the presence of 1,4-cyclohexadiene the 1,5-dihydroflavin 4c is formed, as also in the case of cycloheptatriene, where the resulting tropylium cation reoxidizes 4c back to 1. These reactions, which start from the first excited triplet state, do not necessarily proceed via radical pairs; in many cases they may be interpreted in terms of an electrophilic attack by the excited flavin (3F1,*). Cleavage of R-H into carbenium ion and hydride ion does not occur.- With some reasonable exceptions, the flavin substituents can be removed easily, although they are attached via 0-bonds. The preferred site of the flavin substitution depends on the nature of the substituent: electrophilic groups occur mainly at NS, nucleophilic ones at C4a.
    Notes: Ausgehend von der vielfachen Ähnlichkeit im chemischen Verhalten von lichtangeregtem Flavin und Proteingebundenem Flavin haben wir die Flavinabhängige Photodehydrierung systematisch untersucht: 3-Benzyllumiflavin (1) reagiert photochemisch mit Carbonsäuren wie 3-Indolessigsäure, Phenoxyessigsäure oder (tert-Buty1thio)essigsäure unter Decarboxylierung und Bildung der 4a-alkylierten 4a,S-Dihydroflavinderivate 3a- c. In Gegenwart von Thiodiessigsäure oder Dithiodiessigsäure entsteht das 5-Carboxymethyl-1,5-dihydroflavin-derivat 4 a bzw. das 4a-(Carboxymethy1thio)derivat 3d. - Alkylsulfide werden in α-Stellung unter Ausbildung von 5-substituierten 1,5-Dihydroflavinderivaten angegriffen, die, wenn β-CH2-Gruppen vorhanden sind, einer trans-Eliminierung unterliegen; im Fall des Thiolans ist das Addukt 4d aufgrund sterischer Hinderung stabil. Mit Benzaldehyd entsteht das 5-Benzoyl-l,5-dihydroderivat 4e. - Alkene werden in Allylstellung angegriffen, wobei die 4a-alkylierten 4a,5-Dihydroderivate 3e- g gebildet werden. Mit 1,4-Cyclohexadien entsteht 1,5-Dihydroflavin 4c ebenso wie mit Cycloheptatrien, im letzteren Fall oxidiert das entstandene Tropyliumkation im Dunkeln 4 c zu 1 zurück. Alle diese Reaktionen verlaufen über den ersten angeregten Triplettzustand; sie erfordern nicht notwendigerweise Radikalpaare,sondern können viel eher als ein elektrophiler Angriff des angeregten Flavins (3FlOx*) verstanden werden. Eine Spaltung von R-H in Carbeniumion und Hydridion tritt also nicht auf. - Die Flavinsubstituenten sind bis auf verständliche Ausnahmen äußerst leicht wieder abzuspalten, obwohl hier σ-Bindungen vorliegen. Der Ort der Fixierung des Substrates am Flavin hängt von der Natur des Substituenten ab : Elektrophile Gruppen treten vorzugsweise an N5, nucleophile an C4a auf.
