    Notes: Summary A new aldose reductase inhibitor, (E)-3-carboxy-methyl-5-[(2E-methyl-3-phenylpropenylidene]rhodanine (ONO-2235) was administered orally to streptozotocin-diabetic rats (60 mg/kg IV) for 14 days and its effect on motor nerve conduction velocity studied. The compound significantly improved motor nerve conduction velocity of diabetic rats at a minimal dose of 10 mg · kg-1 · day-1 (treated 28.8±0.5 versus untreated 23.2±4.7 m/s, p 〈 0.01). The sorbitol content of the sciatic nerve and red blood cells measured after 2 weeks was concomitantly reduced in ONO-2235-treated rats (sciatic nerve: 120±13 versus 595±146 nmol/g wet weight; red blood cell: 91±21 versus 165±39 nmol/g haemoglobin; p 〈 0.01 in both 20 mg · kg-1 · day-1-treated versus untreated animals). These results suggest that sorbitol accumulation might contribute to the development of peripheral nerve dysfunction in acutely diabetic animals and the new aldose reductase inhibitor could be a potential drug for therapy of diabetic neuropathy.

Objective and Design: To investigate the effect of ER-34122, a novel pyrazole derivative, on 5-lipoxygenase (LOX) and cyclooxygenase (COX) metabolite production in vitro, ex vivo and in vivo.¶Material: In vitro, lysate of rat basophilic leukemia cells, the microsome fraction of sheep seminal vesicles, human polymorphonuclear leukocytes, human synovial cells, and human monocytes. Ex vivo and in vivo, male Balb/c mice or SD rats.¶Treatment: In ex vivo study, ER-34122 (0.03-1 mg/kg) was orally administered 1 h before withdrawal of blood samples. In carrageenin-induced paw edema, ER-34122 (3–100 mg/kg) and indomethacin (1–10 mg/kg) were orally administered 1 h before carrageenin injection. In arachidonic acid-induced ear inflammation, ER-34122 (0.3–10 mg/kg), zileuton (10–100 mg/kg) and indomethacin (0.3-3 mg/kg) were orally administered 1 h before arachidonic acid application.¶Methods: 5-Hydroxyeicosatetraenoic acid and other eicosanoids were determined by using an HPLC method and enzyme immunoassay, respectively. Rat hind paw edema and mouse ear edema were assessed by measuring paw volume and ear thickness, respectively. Myeloperoxidase (MPO) activity and eicosanoid content of the ear tissue were also determined.¶Results: ER-34122 inhibited both LOX and COX product generation in vitro, and ex vivo. ER-34122 and indomethacin inhibited carrageenin-induced paw edema formation. In the arachidonic acid-induced ear inflammation, ER-34122 inhibited inflammatory responses (edema formation and MPO accumulation) as well as eicosanoids (LTB4, LTC4 and PGE2) generation. A representative LOX inhibitor, zileuton, also inhibited these inflammatory responses, while a COX inhibitor, indomethacin, did not suppress them though it completely inhibited PGE2 generation.¶Conclusions: The anti-inflammatory characteristics of ER-34122 are considered to be superior to those of COX inhibitors such as indomethacin, because in addition to its inhibitory activity on the COX pathway, ER-34122 inhibits LOX products generation, as revealed by the inhibition of edema formation or polymorphonuclear leukocyte infiltration in the arachidonic acid-induced ear inflammation model.

    Notes: Abstract The sensitivity to some chemical agents was examined comparatively at sperm, fertilization, cleavage, blastula, gastrula, pluteus and metamorphosis stages of a sand dollar from Japanese waters (Peronella japonica) and a sea urchin from the Pacific coast of Australia (Heliocidaris erythrogramma). These agents included Cu sulphate, ABS and NH3 chloride. Responses observed included departures from control rates of fertilization and developmental reduction at the attainment of first cleavage, gastrula, pluteus or metamorphosis. Developmental anomalies were noted at the fertilization, 2-cell, gastrula, pluteus and metamorphosis stages. Using minimum effective concentrations of the 3 chemicals at various developmental stages of P. japonica, it was found that sensitivity to chemicals varies from fertilization to metamorphosis. It seems that sperm activity is the most sensitive, and that fertilization and gastrulation are more sensitive than first cleavage, blastulation and pluteus formation. H. erythrogramma seems to show nearly the same responses to Cu, but is more sensitive at metamorphosis.

    Notes: Abstract Production of t-PA by human embryonic lung fibroblasts, IMR-90 cells, is regulated by negative feedback control. The increase in the concentration of the extracellular t-PA lead to a reduction of the production. Therefore, we investigated the application of t-PA adsorption column to ceramic bed reactor to promote t-PA production. Amberlite XAD-8 was selected out as an adsorbent, because it is autoclavable and can adsorb 32,000 IU of t-PA per g wet gel. The t-PA adsorption column was located in the medium recirculation line to the vessel. On the other hand, medium was recirculated between the ceramic bed reactor and the vessel using another flow line. The bioreactor system with the adsorption column was about 2.5 times higher with the resulting cumulative t-PA than that without the adsorption column.

    Notes: Abstract Physicians have long wished for a preparation for intravenous administration which has the functions of native γ-globulin molecules but which has minimal untoward effects. S-sulfonated γ-globulin (GGS) is a preparation which satisfies these requirements, and its intravenous administration results in all of the expected Fc activities. Anticomplementary activity of GGS in the sera of various patients in vitro has indicated an extremely low level of 2–11%. With this in mind, a comparison of the clinical adverse effects of GGS was made in three groups: group A with infections having no underlying disease; group B with malignancies; and group C with immunodeficiencies. GGS was administered 1,076 times to a total of 567 patients. The incidence of side effects was 0.8% in patients of group A; 1.5% in the group B patients; and 14.6% in group C patients. In group C the rate of adverse reactions to the number of administrations was 3.9%. The adverse reactions were mainly minor, i.e., vasomotor symptoms, transient shock-like symptoms, and pyrexia appeared in some cases. In most cases there were no side reactions even in patients given repeated doses of GGS, and the number of cases classified as highly susceptible to adverse reactions was very small. The incidence of side effects was in inverse proportion to age, and the most hazardous symptoms appeared in patients with severe combined immuno-deficiencies. The side reactions appeared within 30 min of GGS administration, with the exception of the febrile responses. The above results indicated that the incidence of side reactions to GGS was not greater than that observed with conventional intravenous preparations of γ-globulin. However, precautions must be taken in the use of GGS for replacement therapy in patients with immunodeficiencies.

    Notes: Abstract S-sulfonated γ-globulin (GGS), derived from human γ-globulin by selective sulfonation, retains the dimensional structure and antibody activity of 7S γ-globulin. GGS does not bind with complement, and is therefore suitable for intravenous administration. Fc activity is recovered through in vivo conversion to intact globulin following IV administration. A total of 414 administrations of GGS was made to 48 patients with primary immunodeficiency syndrome. The frequency of episodes of fever, cough, and infections suffered by these patients who had been treated with other globulin preparations for about one year before the administration of GGS was compared with that following replacement GGS therapy for about one year. Symptomatic relief on administration of GGS was confirmed to be excellent. The administration of 100 mg/kg/BW of GGS resulted in the IgG level rising to 220 mg/dl, with a decrease to 100 mg/dl after one week. The IgG level at two weeks was 70 mg/dl. It is possible to maintain an IgG level of 200 mg/dl by administration of 100 mg/kg/BW of GGS once every 3–4 weeks.

    Notes: Summary Flow studies using dynamic CT and xenon (Xe) CT were carried out in 25 patients with ischaemic stroke in the territory of the middle cerebral artery to define the clinical characteristics of cerebral ischaemia at a chronic stage. The parameter of peak height/mean transit time (PH/MTT) obtained from dynamic CT can provide an accurate index for blood circulation in the cerebral vascular bed. Xe CT measurements revealed various kinds of ischaemia around the infarction even in the chronic stages. In mild ischaemia of more than 30ml/100g/min, reduction of cerebral blood flow (CBF) was well correlated to the PH/MTT. However, in severe ischaemia between 20 and 30 ml/100 g/ min, changes of CBF were no longer correlated with the PH/MTT. There were cases showing severe reduction of CBF but which showed sufficient blood circulation (moderate value of PH/MTT). Mild reductions of CBF in parallel with decreased blood supply were often found in the peri-infarct area of infarctions in the centrum semiovale. On the other hand, infarctions in the cortico-subcortical region showed severe ischaemia, in even where blood circulation was relatively well sustained.

    Notes: We report that entirely end-bonded multi-walled carbon nanotubes (MWNTs) withinTomonaga-Luttinger liquid (TLL) states at high temperatures can show superconductivity withthe transition temperature Tc as high as 12K that is approximately 40-times larger than those inropes of single-walled nanotubes. We find that emergence of this superconductivity is verysensitive to junction structures of Au electrode/MWNTs. This indicates that MWNTs withoptimal numbers of electrically activated shells can allow the superconductivity that overcomesthe TLL sates, due to intershell effects
