    Notes: Abstract  Butterfly wings are colored late in development, when pigments are synthesized in specialized wing scale cells in a fixed developmental succession. In this succession, colored pigments are deposited first and the remaining areas are later melanized black or brown. Here we studied the developmental changes underlying two wing pattern mutants, firstly melanic mutants of the swallowtail Papilio glaucus, in which the yellow background is turned black, and secondly a Spotty mutant of the satyrid Bicyclus anynana, which carries two additional eyespots. Despite the very different pattern changes in these two mutants, they are both associated with changes in rates of scale development and correspondingly, the final color pattern. In the melanic swallowtail, background scales originally destined to become yellow (normally developing early and synthesizing papiliochrome) show delayed development, fail to make papiliochrome, and subsequently melanize at the same time as scales in the wild-type black pattern. In the B. anynana eyespot, scale maturation begins with the central white focus, then progresses to the surrounding gold ring and later finishes with melanization of the black center. Mutants showing additional eyespots display accelerated rates of scale development (corresponding to new eyespots) in wing cells not normally occupied by eyespots. Thus by either delaying or accelerating rates of scale development, the final color, or position, of a wing pattern element can be changed. We propose that this heterochrony of scale development is a basic mechanism of color pattern formation on which developmental mutants act to change lepidopteran color patterns.

    Notes: Summary A set of stage specific proteins of approximally 86 to 90 kDal are synthesized by isolated wings ofPrecis coenia on day 5 of the pupal stage. They are named “B proteins". They are synthesized in presumptive black wing areas in higher amounts than in presumptive white wing areas and are the major proteins synthesized on day 5. Wings from 5 days old pupae, which were incubated with35S-methionine for 2 or 4 hours, incorporate radiolabel into presumptive black pattern elements. This is probably due to the localized synthesis of the above mentioned proteins. Injection of35S-methionine into whole pupae on day 5 resulted in the labelling of the mature black and grey scales but not white scales. This radiolabel incorporation pattern corresponds exactly to the incorporation of the melanin precursor14C-tyrosine into the scales. The results indicate that the “B proteins” are specifically related to the formation of black and grey portions of thePrecis wing pattern. Injection of35S-methionine into whole pupae on day 6 resulted in the labelling of the mature red scales probably due to labelling of “R proteins”, which may be involved in the formation of red pattern elements.

    Description / Table of Contents: Abstract. We report on a 23-year-old man with hidrotic ectodermal dysplasia of the Clouston type showing a severe expression of his gene defect. As a prominent feature of this case, we describe the presence of verruciform keratoderma of hands and feet, and we visualize these skin changes by means of a new imprinting technique.
    Notes: Zusammenfassung. Ein 23jähriger Patient leidet seit Geburt an einer hidrotischen ektodermalen Dysplasie vom Typ Clouston in besonders schwerer Ausprägung. Als ein eindrucksvolles Symptom bestand bei diesem Patienten eine ausgeprägte palmoplantare Keratodermie mit Ausbildung warzenähnlicher Strukturen, die mittels einer neuen Abformtechnik durchlichtmikroskopisch dargestellt werden.

    Notes: Abstract Cytological investigations and cytophotometric DNA determinations were carried out on Feulgen stained squash preparations of male germ cells of Carausius morosus. During zytogene an extra DNA synthesis begins in all spermatocytes. In part of the cysts a complete reduplication of the nuclear DNA takes place. Following this “zygoreduplication phase” the chromosomes of the spermatocytes are present in “tetrapachytene” form and spermatogenesis then proceeds with twice the somatic number of chromosomes. In the other cysts DNA synthesis is blocked after a ten per cent increase. This DNA could be traced through succeeding stages (pachytene, diffuse stage, and diplotene), but neither its structural basis nor its fate is known. The two types of spermatogenesis are described up to and including diplotene.

    Notes: We report on a patient, seen in 1993, who developed sensitization to eosin from topical preparations. The patient had a positive patch test reaction to pure tetrabromofiuorescein but not to its impurities separated by thin-layer chromatography. Our findings suggest that the allergen was eosin itself.

    Notes: We investigated whether patients with contact allergy differed from non-contact-allergic, non-atopic controls with regard to genotype and phenotype of the polymorphic enzyme N-acetyltransferase 2 (NAT2). 55 contact-allergic patients recruited from the Information Network of Departments of Dermatology (IVDK) were compared to 85 controls from among local health care personnel. NAT2 activity was calculated from HPLC analysis of the ratio of the caffeine metabolites 5-acetylamino-6-formylamino-3-methyluracil (AFMU) and 1-methylxanthine (1MX) in the urine. NAT2 genotype was determined by polymerase chain reaction (PCR). A statistically significantly increased proportion of rapid acetylators was found in contact-allergic patients. This may have 2 possible implications: acetylation may enhance contact sensitization; or NAT2 status may be a genetic marker for contact sensitizability.

    Notes: Contact allergy to methyldibromo glutaronitrile (MDBGN), often combined with phenoxyethanol (PE) (e.g., Euxyl K 400®), increased throughout the 1990s in Europe. Consequently, in 2003, the European Commission banned its use in leave-on products, where its use concentration was considered too high and the non-sensitizing use concentration as yet unknown. The 2 objectives of the study are (a) to find a maximum non-eliciting concentration in a leave-on product in MDBGN/PE-sensitized patients, which could possibly also be considered safe regarding induction and (b) to find the best patch test concentration for MDBGN. We, therefore, performed a use-related test (ROAT) in patients sensitized to MDBGN/PE (n = 39) with 3 concentrations of MDBGN/PE (50, 100 and 250 p.p.m. MDBGN, respectively). A subset of these patients (n = 24) was later patch-tested with various concentrations (0.1, 0.2, 0.3 and 0.5% MDBGN, respectively). 15 patients (38%, 95% confidence interval (CI) = 23–55%) had a negative and 24 (62%; 95% CI = 45–77%) a positive overall repeated open application test (ROAT) result. 13 reacted to the lowest (50 p.p.m.), 8 to the middle (100 p.p.m.) and 3 to the highest concentration (250 p.p.m.) only. In those 13 reacting to the lowest ROAT concentration, dermatitis developed within a few days (1–7). The strength of the initial and the confirmatory patch test result, respectively, and the outcome of the ROAT were positively associated. Of the 24 patients with a use and confirmatory patch test, 15 reacted to 0.1% MDBGN, 16 to 0.2%, 17 to 0.3% and 22 to 0.5%. With the patch test concentration of 0.5%, the number of ROAT-negative patients but patch-test-positive patients increases considerably, particularly due to + reactions. A maximum sensitivity of 94% (95% CI = 70–100%) is reached with a patch test concentration of 0.2%, and is not further improved by increasing the concentration. However, the specificity decreases dramatically from 88 (95% CI = 47–100%) with 0.2% to a mere 12.5% (95% CI = 0–53%) with 0.5%. It can be concluded (a) that for MDBGN 0.2% is very likely the best patch test concentration and (b) that 50 p.p.m. in a leave-on product can elicit contact dermatitis in sensitized persons. We were, therefore, unable to find a safe, still microbicidal, concentration for leave-on products. By contrast, with other contact allergens, dose–response use tests may be able to identify a non-eliciting concentration, which could give valuable clues to a non-inducing (i.e., safe) concentration in products.

    Notes: 4 patients developed an erythema-multiforme-like reaction following acute contact dermatitis from 2 different bufexamac-containing topical preparations. Histologically, the lesions did not show changes that were typical of erythema multiforme. Patch testing gave positive reactions to bufexamac at 1, 2, 3 and 4 days. The histological picture of 2 positive test reactions was suggestive of a drug reaction in the 1st, and of an acute eczematous reaction in the 2nd, case. A systemic allergic reaction with spread of lesions could have been involved, following percutaneous absorption of the allergen.
