    Notes: Abstract Many marine organisms inherently express the presence and function of a new defence mechanism, termed multixenobiotic resistance mechanism (MXRM) because of its similarity to multidrug resistance (MDR) found in tumor cell lines resistant to chemotherapeutic drugs. However, previously no information was available on a possible induction of the activity of MXRM in organisms living at polluted sites. The purpose of the present study was to demonstrate the inducibility of this defence mechanism in the marine snail Monodonta turbinata, an organism known to inhabit a wide range of environmental conditions existing along a pollution gradient. Specimens of M. turbinata used in the present study were collected from March 1993 through March 1994 near Rovinj, Croatia. The accumulation of generally labelled 3H-vincristine (3H-VCR) in the gills of snails living at an unpolluted site was 67% higher and very sensitive to verapamil, an inhibitor of P-glycoprotein activity in comparison to snails living at a polluted site. The accumulation of vincristine (VCR) in snails from a polluted site was reduced and insensitive to verapamil due to the induced state of P-glycoprotein, as was demonstrated immunochemically by the enhanced concentration of a 140 kDa protein cross-reacting with the polyclonal antibodies raised against mammalian P-glycoprotein. MXRM could be induced in snails from the unpolluted site to the level found in snails living at a polluted site either by transplantation for 3 d to a polluted site, or by exposure for 3 d to sea water polluted with Diesel-2 oil (hydrocarbon concentration equivalent to 50 ppb of Kuwait oil). Discovery of the inducibility of this new defence mechanism in aquatic organisms may have important implications in ecotoxicology, as was demonstrated by a 104% enhanced accumulation of VCR in the presence of the MXRM-inhibitor, the so-called “chemosensitizer”. Besides, a striking difference in the levels of MXRM-elements (the sensitivity of the accumulation rate of xenobiotics to verapamil, immunochemical expression of P-glycoprotein, and the sensitivity of the binding of xenobiotics on membrane vesicles to verapamil) found between specimens living at the polluted and unpolluted sites offers a new molecular biomarker for exposure to pollutants.

    Notes: Abstract A hydrolysis procedure along with a high-pressure liquid chromatographic procedure is given enabling simple and reliable thymine determinations in the nanogram range in different fractions of sea-water samples taken from three different locations in the Northern Adriatic Sea. The levels corresponded to 1–3 μg DNA per liter. From total polyanionic thymine, which had been precipitated as the cetyltrimethylammonium salt, the highest percentage was linked to the particulate fraction, with a definite subsurface minimum at 10 to 15 m. There was a corresponding maximum of a high molecular “non-particulate” thymine-containing fraction at the corresponding depth. From the bottom at 30 m upwards to about 20 m, a low molecular thymine-containing material has been found. Remarkably these basic features were common to all three locations, one of which was supposed to be in clean water, one near a thickly settled, touristic area, and the other in front of a large river delta coming from industrial hinterland.

    Notes: Abstract The siliceous spongeGeodia cydonium Jameson was used to study the influence of pollution in marine environments on selected parameters of the intracellular signal transduction pathway. The parameters chosen were: intracellular distribution of protein kinase C (PK-C),ras-gene expression and DNA polymerasealpha (DNA Polalpha) activity. Both PK-C andras-gene product (ras-protein) have previously been established to be key molecules in the intracellular signalling pathway in sponges; increased level ofras-protein mediates events following sponge cell-cell contact. Three unpolluted and three polluted sites in the off-shore seawater around Rovinj (Yugoslavia) were selected for the study in 1989. The state of pollution of these sites has been well-defined in a series of previous studies (1976 to 1989). Transplantation of regenerating sponge cubes ofG. cydonium to the polluted sites resulted in pronounced changes in the parameters chosen, compared to controls exposed to unpolluted environments. Expression ofras gene was increased by three- to five-fold after exposure of regenerating sponge tissue to the impacted sites. At the site with the highest pollutional load,ras mRNA level was about 50% of that at the reference sites. In parallel experiments it was established that, in response to pollution, a translocation of PK-C from the cytosolic to membrane fraction occurred. At the most impacted site, most of the enzyme activity was cytosolic. DNA Polalpha-activity, as a measure of sponge cell proliferation, decreased in a pollutioncorrelated manner. Our results indicate that the intracellular signalling system within sponge cells is activated in response to moderate pollution but is depressed in heavily polluted environments.

    Notes: Abstract Experiments were carried out with the marine sponge Suberites domuncula to determine whether sponges may express-like mammalian tumor cells-a multidrug-like transporter system. The results demonstrate that sponge cells possess such a protective system termed multixenobiotic resistance (MXR) pump or P-glycoprotein-like pump. The protein was identified by antisera for the mammalian P170 multidrug resistance protein as a 130 kDa molecule. Binding studies were performed with 3H-vincristine (3H-VCR) and membrane vesicles; this process is ATP-dependent and inhibited by verapamil, which is known to reverse the multidrug-resistance phenotype in mammalian systems. Accumulation experiments were performed to demonstrate that the uptake of 3H-VCR is time-dependent, and increases at elevated extracellular levels of 3H-VCR. Application of the dyeing technique with calcein-AM, a suitable functional assay for multidrug transporter systems in mammal cells, also revealed the existence of the MXR pump in S. domuncula plasma membranes. These data demonstrate that S. domuncula is provided with a multidrug-like transporter, the MXR pump, which might function as a protection system for sponges in polluted environments.

    Notes: Abstract Induction of benzo (a) pyrene monooxygenase (BPMO) activity occurred in Blennius pavo, a species with a restricted territorial range, in response to exposure to a Diesel 2 oil. A response delay of 14 days was found at a concentration of 170 ppb and of 3 days when the water was saturated with Diesel 2 oil. When induced fish were transferred to clean water, elevated BPMO activity was maintained at a high level for at least a month. A benthic protochordate, Microcosmos sulcatus, showed no increase in BPMO activity when exposed to these concentrations even after 30 days of exposure. Field observations revealed a great variation in the BPMO activity from B. pavo caught at different sites. Fish from contaminated sites had significantly elevated levels of BPMO activity. Sardine schools caught at different sites had different, low levels of BPMO activities. However, specimens from the same school had closely similar levels of enzyme activity. An oil pollution incident (New Year 1977 oil spill in Northern Adriatic Sea) caused an increase in the BPMO activity in the livers of Blenniideae, reaching a peak on the 23rd day (representing an 8.5-fold increase in the background level), followed by a decrease in activity until a new background level, 3 times that of the original background level, was reached on the 45th day. This new background level is constant (through May-5 months after the incident). Measurement of BPMO activity in the livers of nonmigrant fish could serve as a useful biochemical parameter for monitoring and evaluation of acute or long-term oil pollution at a given site.
