    Notes: The search for effective means of selectively delivering high therapeutic doses of anti-cancer agents to tumors has explored a variety of systems in the last decade. The ability of intravenously injected clostridial spores to infiltrate and thence selectively germinate in the hypoxic regions of solid tumors is exquisitely specific, making this system an interesting addition to the anti-cancer therapy arsenal. To increase the number of therapeutic proteins potentially useful for cancer treatment we have tested the possibility of Clostridium acetobutylicum to secrete rat interleukin-2 (rIL2). Therefore, rIL2 cDNA was placed under the control of the endo-β-1,4-glucanase promoter and signal sequence of C. saccharobutylicum. Recombinant C. acetobutylicum containing the relevant construct secreted up to 800 μg l−1 biologically active rIL2. The obtained yield should be sufficient to provoke in vivo effects.

    Notes: After the discovery of C60, a large family of fullerene molecules was also identified. Among them, elongated fullerenes are formed by the tubular assembly of carbon atoms. The van der Waals bonds between fullerene molecules are due to the correlations between fluctuating charge densities inside the molecules. The interaction is then dominated by collective excitations which are sensitive to the shape of the molecules. Therefore, van der Waals attraction is expected to be modified when considering successively spherical C60, C70 and more elongated fullerenes (tubules). This paper presents self-consistent computations of the van der Waals interaction between a (111) diamond probe tip and various fullerene molecules adsorbed on a gold surface. Relative to spherical C60, the dependence law of the force experienced by the probe tip as a function of the tip-sample distance decreased when approaching fullerene tubules. Simulations of scanning force microscope scans of carbon tubules next to C60 molecules show that the shape of the molecules affects the interpretation of scanning force microscopy imaging. Particularly, information about the height of the various molecules deposited on the surface must be considered with some care since carbon tubules with the same radius as C60 interact more strongly with the probe tip. © 1994 American Institute of Physics.

    Notes: Previous studies have demonstrated the feasibility of using apathogenic clostridia as a promising strategy for hypoxia-specific tumour targeting. The present study shows that the use of the vascular targeting compound combretastatin A-4 phosphate could significantly (P〈0.001) increase the number of Clostridium vegetative cells in rat rhabdomyosarcomas with sizes between 0.2 cm2 and 3 cm2. Furthermore, this study showed that administration of metronidazole for a 9-day period was sufficient to eliminate systemically administered Clostridium from the tumour. Moreover, previous Clostridium spore administration did not effect tumour colonisation, regardless of the immune response status of the host.
