7-Deaza Isosters of 2′-Deoxyxanthosine and 2′-Deoxyspongosine - Synthesis via Glycosylation of 2,4-Dichloro-7H-pyrrolo[2,3-d]pyrimidine7-Deaza-2′-deoxyxanthosine (1a) and 7-deaza-2′-deoxyspongosine (2), two new pyrrolo[2,3-d]-pyrimidine deoxynucleosides, have been prepared by nucleophilic displacement of the chloro substituents of the intermediate 2,4-dichloro-7-[2-deoxy-3,5-di-O-(p-toluoyl)-β-D-erythro-pentofuranosyl]-7H-pyrrolo[2,3-d]pyrimidine (5a). Compound 5a was obtained by glycosylation of 2,4-dichloro-7H-pyrrolo[2,3-d]pyrimidine (3b) with the halogenose 4. Due to the reactivity of the chromophore halogens removal of the toluoyl residues by sodium methoxide resulted in rapid displacement of the chloro substituents which prevented the isolation of the free deoxyriboside of 3b and led to the 2,4-dimethoxynucleoside 7. As shown by the conversion of 5a into 2, regioselective displacement of the halogen in position 4 can be accomplished under conditions which maintain the protecting groups of the sugar moiety.

7-Desaza-2′-desoxyxanthosin (1a) und 7-Desaza-2′-desoxyspongosin (2), zwei neue Pyrrolo[2,3-d]-pyrimidin-Desoxynucleoside, wurden aus der gemeinsamen Vorstufe 2,4-Dichlor-7-[2-desoxy-3,5-di-O-(p-toluoyl)-β-D-erythro-pentofuranosyl]-7H-pyrrolo[2,3-d]pyrimidin (5a) durch nucleophile Substitution erhalten. 5a entstand durch Glycosylierung von 2,4-Dichlor-7H-pyrrolo-[2,3-d]pyrimidin (3b) mit der Halogenose 4. Mit Natriummethoxid erfolgte der Austausch der Chromophorhalogene unter Abspaltung der Toluoylreste, was eine Isolierung des freien Desoxyribosids der Base 3b verhindert und statt dessen zum 2,4-Dimethoxy-Nucleosid 7 führte. Wie die Umwandlung von 5a in 2 zeigt, kann das Halogen in Stellung 4 regioselektiv ausgetauscht werden, wenn für den Erhalt der glyconischen Schutzgruppen gesorgt wird.
