    Notes: Abstract Measurements of microbial community size, including total cell counts and specific degrader enumerations, were conducted on subsurface soil samples from both petroleum-contaminated and pristine aquifers. Samples were collected from both uncontaminated and contaminated areas of the petroleum-contaminated sites. In pristine and uncontaminated samples, total cell counts (acridine orange direct counts) were related to depth. The deeper samples contained smaller total microbial populations. However, indices of microbial activity varied considerably from sample to sample and probably reflect soil and site heterogeneity. Exposure to petroleum contamination apparently altered the microbial community structure. In samples exposed to low levels of contaminants as vapors and/or dissolved phases (ppb concentrations), and not free product, the toluene-specific degrader populations were larger at greater depths, and the numbers of amino acid-specific degraders were highly correlated to the numbers of decane-specific degraders, indicating that petroleum-adapted microbial communities were present in the contaminated samples. In highly contaminated samples, total microbial population densities decreased with increasing depth; however, microbial activity tended to increase with depth. These results indicate that petroleum contaminants exert toxic effects on the active microbial community at high exposures and enrich specific degraders at ppb levels of dissolved contaminants.

    Notes: Abstract The National Institute of Standards and Technology (NIST) has developed several Standard Reference Materials (SRMs) based on human serum. NIST SRM 909b, Human Serum, is a lyophilized human serum material with concentrations for seven organic and six inorganic analytes at two levels certified solely by definitive methods (DMs). This material provides the vehicle by which high precision, high accuracy measurements made with DMs at NIST can be transferred through the measurement hierarchy to other laboratories. Isotope dilution gas chromatographic-mass spectrometric (GC-IDMS) methods were applied to measure cholesterol, creatinine, glucose, urea, uric acid, triglycerides, and total glycerides. Thermal ionization isotope dilution mass spectrometry (TI-IDMS) was used for determination of lithium, magnesium, potassium, calcium, and chloride. In addition, chloride was determined by coulometry, providing a comparison between two DMs. Sodium, which lacks a stable isotope that would permit isotope dilution mass spectrometric (IDMS) measurement, was determined by gravimetry. SRM 909b includes certified values for total glycerides and triglycerides, which were not certified in the previous lot of this material (SRM 909a). Improvement in uniformity of vial fill weight in the production of SRM 909b resulted in smaller certified uncertainties over previous freeze-dried serum SRMs. Uncertainties at the 99% level of confidence for relative expanded uncertainty (%) for certification of the organic analytes on a mmol/L/g basis ranged from 0.44% for urea (level II) to 5.04% for glucose (level II). (In-house studies have shown glucose to be a relatively unstable analyte in similar lyophilized serum materials, degrading at about 1% per year.) Relative expanded uncertainties (99% C.I.) for certification of inorganic analytes on a mmol/L/g basis ranged from 0.25% for chloride (level I) to 0.49% for magnesium (level II).

    Notes: Summary The ceramides and steryl-sulfate components from desquamated and cohesive human stratum corneum were examined using a combination of chemical and chromatographic means. Six structurally distinct series of ceramides were identified, and the relative amounts of these species, as measured by quantitative thin-layer chromatography, did not differ in cohesive and desquamated stratum corneum. In contrast, the level of cholesteryl sulfate was significantly reduced in the desquamated material. The results are in accord with the hypothesis that cholesteryl sulfate serves in cell-to-cell cohesion within the stratum corneum, and its hydrolysis may be necessary to permit shedding of cells from the surface.

    Notes: Summary Low-temperature time-resolved luminescence experiments with picosecond resolution have been performed on excitons in a microcavity containing InGaAs quantum wells. The decay curves are very dependent on the energy difference between the cavity mode and the exciton. They agree qualitatively with a model which takes into account a reservoir at the exciton energy and two splitted radiant states.

    Notes: The cornified cell envelope (CE) formed by transglutaminase-mediated ε-(γ-glutamyl)lysine cross-linking of specialized corneocyte proteins is the most insoluble component of the terminally differentiated keratinocyte. Under normal Nomarski optics, two types of CE are readily distinguishable: an irregularly shaped, readily deformed ‘fragile’ envelope (CEf), which predominates in the deepest layers of the stratum corneum, and a polygonal “resilient” or ‘rigid’ envelope (CEr), which represents over 80% of the CE population in the superficial layers. This distinct spatial distribution indicates a maturation of the CE from the fragile to the resilient morphology during stratum corneum maturation. In this study, we have examined morphological and physical changes occurring in the CE during the terminal differentiation. The proportion of CEf and CEr present in superficial samples of stratum corneum were readily distinguishable following staining with Tetrarhodamine isothiocyanate (TRITC) and showed significant body site variation. The percentage of CEf was highest on samples recovered from exposed body sites (back of hand 〉 cheek 〉 inner arm [bicep region]) suggesting innate body site differences or that photodamage and other environmental trauma can reduce or delay normal CE maturation. Soap-induced dryness resulted in a significant decrease in CE maturation coincidental with reduced corneodesmosomal hydrolysis. Effective moisturization of winter-induced dry skin enhanced CE maturation (33% increase in TRITC fluorescence, n = 14 following 4-week treatment). Using a novel micromanipulation instrument, the force required (µN) to maximally deform individual CEf and CEr was compared. CEf recovered from deep stratum corneum were significantly softer and weaker than CEr recovered from superficial layers. These studies indicate that the normal process of CE maturation is associated with an actual strengthening of this insoluble protective structure and that the impairment of this process is associated with poor quality of the stratum corneum.

    Notes: We report 6 cases of an immature variant of trichoepithelioma which histologically appears to show differentiation toward the primitive hair germ. The lesions presented in mature adults (mean age 44 years). Four occurred in men and 2 in women. Four lesions occurred on extremities, an unusual location for trichoepitheliomas. Histologically, the lesions were characterized by well-circumscribed, but unencapsulated, dermal collections of small tumor lobules composed of basaloid cells with invaginations resembling primitive dermal hair papillae. There was no adenoidal growth pattern or horn cyst formation. The separation of the immature lesions from those of classical trichoepithelioma and basal cell carcinoma can be made if multiple morphological features are considered; no one particular finding is diagnostic. The major differential features between the immature trichoepithelioma and basal cell carcinoma are circumscription, tumor lobule uniformity, occasional immature hair germs, and lack of retraction artifact of the tumor lobules from the stroma. The differential features between the immature and classical trichoepithelioma are less conspicuous. The immature form typically exhibits no horn cysts, displays fewer primitive hair structures, and lacks the adenoidal growth patterns of the tumor lobules which are usually present in the classical trichoepitheliomas.
