    Notes: Abstract The anti-inflammatory properties of 7 non-steroidal drugs have been examined in two animal models and one in vitro system. The models used were (1) carrageenin-induced paw oedema in rats previously depleted of polymorphonuclear cells, (2) carrageenin-induced rat pleurisy and (3) migration of rat peritoneal leucocytes from glass capillary tubes in vitro. All of the drugs suppressed oedema formation in rats depleted of polymorphonuclear cells. None affected the migration of polymorphonuclear cells into the inflamed pleural cavity. Four drugs suppressed the migration of mononuclear cells in the latter model. Of these, only phenylbutazone and indomethacin suppressed the random migration of leucocytes in vitro. It was concluded that the suppression of mononuclear cell migration is not a general property of non-steroidal anti-inflammatory durgs.

    Notes: Abstract Cathepsin B and L activity was studied histochemically in arthritic rat ankle joints using specific synthetic substrates in a post coupling method on unfixed and undecalcified cryostat sections of rat ankle joints. Activity was strongly increased in chondrocytes and cells of the inflamed synovium with the development of arthritis induced by the synthetic adjuvant CP20961. Activity reached a maximum 20 days after induction of arthritis and decreased as the rats entered natural remission. Cathepsin B and L were at their highest level when macrophages were present in the joint space, as shown by using monoclonal antibody markers for rat macrophages (ED1 and ED2) in a biotin-avidin immunoperoxidase assay. This suggests that the macrophage infiltrate may have stimulated proteinase production in chondrocytes through cytokine release. The profile of appearance of cysteine proteinases suggests their involvement in the breakdown of cartilage and bone in the arthritic joint.

    Notes: Summary A method has been developed to cut unfixed and undecalcified sections of rat paws from animals with adjuvant arthritis and to stain them by a biotin-avidin immunoperoxidase technique. Good tissue integrity and morphology throughout the immunohistochemical procedure were retained if the sections were first mounted on transparent sellotape. The method is illustrated with two monoclonal antibodies (mAb) and is generally applicable with any mAb or polyclonal antibody and with joints from other small animals. For rats with adjuvant arthritis it was found important to block endogenous peroxidase before immunostaining. Complete inhibition of this enzyme without loss of antigenicity was best achieved after application of the mAb and biotinylated anti-IgG conjugate to the unfixed tissue sections.
