    Notes: Determination of HER-2 status is an essential prerequisite in considering a patient's eligibility for anti-HER-2 therapy; few studies have focused on its evaluation on cytological material. We present a new method of assessing HER-2 status on cytological samples, by fluorescence in situ hybridization (FISH) with an automated detection system, using fresh frozen (FF) and May–Grünwald–Giemsa (MGG) stained smears, and we evaluate the reliability of HER-2 determination on fine needle aspiration cytology (FNAC). The pre-treatment protocol for FF smears is easier and faster than for MGG stained slides. However, with the described procedure, FISH is also feasible on archival MGG stained slides. We conclude that with this method, cytological samples obtained by FNAC, either FF or MGG, are a reliable option for assessing HER-2 status.

    Notes: Objective:  Searching for multiple molecular markers in thyroid aspirates appears to be a promising approach for establishing a preoperative diagnosis of papillary thyroid carcinoma (PTC).Methods:  Based on this hypothesis, a total of 63 samples from 55 patients, were collected at random. RNA was extracted from the residue cells inside the needle used for fine needle aspiration cytology (FNAC) and thereafter molecular analysis was carried out both for RET rearrangements (type 1, 2, 3) and BRAF codon 599 mutation molecule. Results were compared with the cytological and histopathological diagnoses in 24 patients submitted to surgery.Results:  58% PTCs presented a genetic alteration either RET/PTC rearrangement, BRAF V599E mutation or both: three cases of PTCs (25%) presented a RET/PTC rearrangement; three cases of PTCs (25%) presented a BRAF V599E mutation and in one case (8%) both alterations were identified.Conclusions:  The present results suggest that searching for multiple molecular markers in thyroid aspirates may enhance the accuracy of FNAC and refine preoperative diagnosis of PTC.

    Notes: Ria de Alvor is a small estuary between Lagos and Portimão on the south coast of Portugal, 37°7′–37°10′N, 8°35′–8°38′W.Samples were collected monthly in the period April 1985–March 1986 in an ichthyoplankton and ichthyofauna survey of the estuary. A WP-2 plankton net was used for catching ichthyoplankton, and a beam trawl and a beach seine were used for catching ichthyofauna.Scaldfish, Arnoglossus laterna (Walbaum, 1792), even though the most abundant species in ichthyoplankton samples, was never found in ichthyofauna samples.From the results obtained, it is concluded that a great part of the fish species are temporary marine visitors using the Ria as a nursery ground, although this system appears to provide a spawning area for species with benthic eggs. No evidence was found of pelagic spawning.

    Notes: Lichen sclerosus et atrophicus (LSA) is a chronic, benign, depigmenting disease of the skin and mucous membranes most frequently affecting the female genitalia. Involvement of the oral mucosa without concurrent genital or skin lesions has been reported only occasionally in the literature. In view of the rarity of reported cases, one lesion affecting only the labial mucocutaneous area is presented along with a description of the disease's clinical and histopathological findings.

    Notes: Abstract This paper presents the results from a sedimentary survey undertaken in July 1992 in the Sado estuary, western coast of Portugal. Sediment samples were taken at 14 sites, corresponding to four organic enrichment gradients, three of which are located close to harbour facilities or to industrial or urban effluent outfalls. The fourth site is located away from direct sources of anthropogenic disturbance. An integrated approach was conducted in order to study the quality status of the superficial sediments, and included 1. the analysis of sediment descriptors and contaminants (grain-size, total organic matter, heavy metals, PCBs), 2. the evaluation of toxicity, using bioluminescent bacteria and the Microtox® solid-phase protocol and 3. the study of macrofaunal communities structure and composition. Most of the sediments showed low IC50 values (high toxicity), while benthic macrofauna did not indicate community disturbance in most of the sites, nor the contaminants analysed provided explanation for the potential toxicity observed. Most of the sediments showed high fines content. A statistical significant relation between fines and IC50 values was found, indicating the sensitivity of the toxicity assessment method for granulometry. This sensitivity might explain the lack of agreement between the IC 50 values and the sediment chemistry-infaunal communities structure. The potential grain-size sensitivity was further analysed using another test sediment from which artificial samples with different sand∶fines ratios were prepared. It is concluded that extreme caution should be used when applying the solid-phase Microtox® protocol to assess the toxicity of natural sediments, covering a range of granulometry and contamination situations.
