    Notes: Abstract. Infestation in soft tissue by Echinococcus granulosus is not a common disease, and its diagnosis is based on clinical, laboratory data and radiological findings. The aim of our retrospective study is to give an overview of the different signs and patterns shown by MRI that can be useful in characterizing soft tissue hydatid disease. The MRI images obtained in seven patients with soft tissue and subcutaneous hydatidosis were reviewed. Typical signs of hydatidosis were multivesicular lesions with or without hypointense peripheral ring (“rim sign”). Related to the presence and absence, respectively, of viable scolices in the microscopic exam, daughter cysts were presented either as high signal intensity or low signal intensity on T2-weighted images. Low-intensity detached layers within the cyst and peripheral enhancement with gadolinium-DTPA were also presented. Atypical signs were presented in an infected muscular cyst, a subcutaneous unilocular cyst and several unilocular cysts. Knowledge of the different patterns in MRI of soft tissue hydatid disease can be useful in diagnosing this entity. We observed that the “rim sign” is not as common as in other locations, and in addition, MRI seems to be of assistance when evaluating the vitality of the cysts.

    Notes: Abstract Based on conserved amino-acid regions predicted for the chitin synthases (Chs) of Saccharomyces cerevisiae, two different primer sets were synthesized and used in polymerase chain reactions (PCRs) to amplify 614-bp and 366-bp sequences from genomic DNA of the zoopathogenic fungus Phialophora verrucosa. DNA-sequencing and Southern-blotting analyses of the 614-bp DNA amplification products suggested that portions of two distinct P. verrucosa chitin synthase genes (PvCHS1, PvCHS2), coding for two different zymogenic-type PvChs isozymes, had been identified. The deduced amino-acid sequence of each fell into different Chs classes, namely class I and class II. In addition, the 366-bp DNA segment was shown to code for a conserved region having homology with the CSD2/CAL1 gene of S. cerevisiae, which encodes a nonzymogenic-type enzyme, Chs3, in that fungus. The amino-acid sequence derived from PvCHS3 exhibits 88.2% similarity and 78.4% identity to the same amino-acid region of the S. cerevisiae enzyme. These results provide a critical first step toward investigating the molecular and pathogenic importance of CHS gene regulation in this fungus and for exploring steps leading to Chs function as potential targets for the design of new therapeutic agents.

    Notes: Samples of finely ground cowpea flour containing 7.5%, 19.4% and 25.5% moisture were heated in sealed tubes at 100° 125° and 150°C for periods of 0.5 to 120 min. First order rate constants for losses of trypsin inhibitor activity and nitrogen solubility ranged from 1 × 10−2 to 18 min−1 and from 4 × 10−2 to 8 min−1 respectively. In vitro protein digestibility (IVD) increased, then decreased with heating as described by sequential first order kinetics. Rate constants for increase of IVD varied from 0.13 to 12 min−1, while for decrease in IVD the range was 5 × 10−5 to 3 × 10−2 min−1. Activation energies and relationships between In k and water activity were computed.

    Notes: Background The inflammatory functions of complement component 5 (C5) are mediated by its receptor, C5R1, which is expressed on bronchial, epithelial, vascular endothelial and smooth muscle cells. A susceptibility locus for murine allergen-induced airway hyper-responsiveness was identified in a region syntenic to human chromosome 19q13, where linkage to asthma has been demonstrated and where the gene encoding C5R1 is localized.Objective The aim of this study was to screen for novel polymorphisms in the C5R1 gene and to determine whether any identified polymorphisms are associated with asthma and/or atopy and whether they are functional.Methods Single-nucleotide polymorphism (SNP) detection in the gene encoding C5R1 was performed by direct sequencing. Genotyping was performed in three populations characterized for asthma and/or atopy: (1) 823 German children from The Multicenter Allergy Study; (2) 146 individuals from Tangier Island, Virginia, a Caucasian isolate; and (3) asthma case–parent trios selected from 134 families (N=783) in Barbados. Functional studies were performed to evaluate differences between the wild-type and the variant alleles.Results We identified a novel SNP in the promoter region of C5R1 at position −245 (T/C). Frequency of the −245C allele was similar in the German (31.5%) and Tangier Island (36.3%) populations, but higher in the Afro-Caribbean population (53.0%; P=0.0039 to 〈0.0001). We observed no significant associations between the −245 polymorphism and asthma or atopy phenotypes. Upon examination of the functional consequences of the −245T/C polymorphism, we did not observe any change in promoter activity.Conclusion This new marker may provide a valuable tool to assess the risk for C5a-associated disorders, but it does not appear to be associated with asthma and/or atopy.

    Notes: Abstract The present experiment explored the anorectic and adipsic effects of fluprazine hydrochloride, a phenylpiperazine compound. Thirty-eight albino rats were randomly assigned either to a control saline group (six rats) or to groups (eight subjects each) receiving an IP dose of fluprazine in saline (1.25, 2.5, 5 or 10 mg/kg). No anorectic effect of the drug doses was observed 30, 60, 90, 120, 180 and 240 min, and 24 h after drug injection. However, water drinking was significantly decreased 30 min after drug administration, with 5 and 10 mg/kg, compared to saline.

    Notes: Summary Crosses between common bean genotypes from different centres of origin can result in F1 plants with dwarfing that is severe enough to prevent formation of F2 seed. This hybrid dwarfness is controlled by two complementary genes, Dl-1 and Dl-2. The dominant alleles at both loci must be present to cause dwarfing, and an increased dose of either allele increases the severity of the dwarfing. Since dwarfing restricts crossing among gene pools, it holds interests for bean breeding and studies of bean evolution and physiology. Results of grafting studies are described which confirm the previously reported dependance of dwarfing on independent root and shoot effects, and further show that grafting may be used to characterize genotypes for presence of the Dl-1 and Dl-2 alleles and to produce normal shoots in plants which otherwise would have been dwarfed. In the first experiment, two carriers each of Dl-1 and Dl-2 and a non-carrier were grafted reciprocally; of the 25 combinations, only the four combinations of Dl-2-carrying shoots with Dl-1-carrying roots produced dwarfing. In a second experiment, 11 carriers of Dl-1 and 9 of Dl-2 were grafted to Dl-1 roots and Dl-2 shoots. Although most graft combinations where Dl-1 roots were combined with Dl-2 shoots resulted in dwarfed plants, 10 genotypes showed mixed responses or otherwise did not respond as expected. Further experiments confirmed the occurrence of materials giving mixed responses or showing dwarfing irrespective of the graft combination. While grafting has utility as a screening tool, results should be interpreted with caution. There remains a need to determine whether the unexplained results reflect effects of additional alleles or genes affecting the dwarfing response. Grafting was used successfully to obtain normal F1 shoots, and ultimately, to produce F2 seed after two cycles of backcrossing (BC2) for four crosses which otherwise would have given dwarfed plants. Segregation of dwarfed and normal plants in progeny rows of F2 plants derived from the BC2 populations showed 1:3 ratios, this being consistent with the reported inheritance.
