    Description / Table of Contents: Zusammenfassung Bei einem 36jährigen Patienten mit einer kompletten Agammaglobulinämie und seinen Verwandten wurden humorale und celluläre Immunität untersucht. Im Knochenmark des Patienten fehlten die Plasmazellen. Sowohl die positiven Hautreaktionen gegenüber PPD und Tetanustoxoid als auch die Stimulation der peripheren Lymphocyten mit PHA deuten auf ein intaktes T-Zellsystem hin. Der Anteil der B-Lymphocyten im peripheren Blut war normal. Dieser Befund korreliert mit einer normalen Stimulationsrate mit Anti-IgG-Serum. Die Befunde sprechen für eine Reifungsstörung der B-Lymphocyten zu Plasmazellen.
    Notes: Summary Humoral and cell-mediated immune reactions of a 36-year old patient with a late onset of an agammaglobulinaemia and of his immediate relatives have been investigated. The immunoglobulins gamma-G, gamma-A, gamma-M, gamma-E and gamma-D, and plasma cells in the bone marrow could not be detected in the patient's serum. Immune reactions with PPD and tetanus toxoid as well as the stimulation of the lymphocytes with PHA indicate that the T-cell system was intact. The examination of the peripheral blood lymphocytes by immunofluorescence techniques gave a normal number of B-lymphocytes. These findings could be corroborated by the results of lymphocyte stimulations with anti-IgG. The data suggest that the maturation of the B-lymphocytes to plasma cells may be disturbed.

    Description / Table of Contents: Zusammenfassung Die cellulären Immunreaktionen gegenüber homologen leberspezifischen Proteinen (HLP) und dem Hepatitis-assoziierten Antigen (HAA) wurden bei Patienten mit akuter HAA-positiver Hepatitis unter Verwendung des Leukocytenmigrationstestes untersucht. Die Migration war bei 30% der Patienten inhibiert, wenn HLP als Antigen verwandt wurde. Mit HAA zeigten 64% der Patienten, die später im Serum HAA-negative wurden, einen Migrationsindex unter 0,7 und 15% eine Stimulation. Die Häufigkeit der Migrationsinhibition betrug nur 22% bei den Patienten, die persistierende HAA-Antigenträger waren. Nur bei 20% der Fälle war der Test gegenüber beiden Antigenen anormal. Aus den Ergebnissen kann geschlossen werden, daß 1. eine celluläre Immunität gegenüber HAA öfter bei Patienten auftritt, die das Antigen verlieren. 2. die verzögerte Immunität gegenüber den beiden getesteten Antigenen nicht miteinander in Beziehung steht. 3. die Toleranz gegenüber homologen leberspezifischen Antigenen bei der akuten HAA-positiven Hepatitis gebrochen werden kann. Dieser Befund weist eindeutig auf den Einfluß der Virusinfektion für die Entstehung einer Autoimmunität bei Lebererkrankungen hin.
    Notes: Summary The cell-mediated immune responses towards homologous, liver-specific antigens (HLP) and hepatitis-associated antigen (HAA) were studied in patients with acute HAA-positive hepatitis using the leukocyte migration test. The migration was inhibited in 30% of the patients when HLP was used. With HAA 64% of the patients—who later became negative for HAA in their serum-exhibited migration indices below 0.7 and 15% of them a stimulation. The incidence of migration inhibition was as low as 22% in patients who were persistently positive for HAA in their serum. In 20% of the cases only was the test abnormal for both antigens. It can be concluded from our results that 1. cellular immune reactions towards HAA occur more often in patients who clear the antigen. 2. the delayed type immunity against the two antigens tested seems not to be correlated. 3. the tolerance against homologous liver specific antigens can be broken in acute HAA-positive hepatitis. This finding clearly points out the influence of viral infection for the development of autoimmunity in liver diseases.

    Description / Table of Contents: Summary The Dane particle is — because of its characteristics — believed to be the complete hepatitis-Bvirus. Sera containing high numbers of Dane particles were shown to be highly infectious. In the present study we related the HBeAg-, the HBsAg- and the anti-HBc titer to the level of DNA polymerase activity measured in 20 fold Dane particle concentrates. The data obtained indicate that the HBeAg concentration gives a semiquantitative estimate on the number of circulating Dane particles. Mean DNA polymerase activity was found to increase with HBsAg concentration and is therefore also of value-if determined in a HBeAg positive serum- for quantitation of Dane particles. The anti-HBc titer was found to be unrelated to the number of circulating Dane particles.
    Notes: Zusammenfassung Das Dane Partikel wird als das komplette Hepatitis B Virus angesehen. Seren mit einer großen Zahl von Dane Partikeln gelten als hoch infektiös. In der vorliegenden Studie haben wir die Höhe der DNA Polymerase Aktivität — gemessen in 20fach Dane Partikel Konzentraten — mit dem HBsAg, dem HBeAg und dem anti-HBc Titer verglichen. Unsere Untersuchungen zeigen, daß der HBeAg Titer ein semiquantitatives Maß für die Zahl zirkulierender Dane Partikel darstellt. Die HBsAg Konzentration steigt ebenfalls mit der Zahl im Serum zirkulierender Dane Partikel an und stellt ebenso — wenn in HBeAg positiven Seren gemessen — ein semiquantitatives Maß für die Zahl zirkulierender Dane Partikel dar. Zwischen dem anti-HBc Titer und dem Serum Dane Partikel Gehalt besteht keine Beziehung.

    Notes: The ganglioside GD3 has a variety of biological functions. These include stimulatory effects is on proliferation, natural killer activity and cytokine production by freshly isolated peripheral T cells. In this study we have characterized anti-GD3 antibody (MoAb Z21) mediated effects on T cell clones. Our data indicate that α/β TCR CD4+ and CD8+ as well as γ/δ TCR positive T cells can be stimulated resulting in proliferation and cytokine production. This effect could be blocked by cyclosporin A and did not involve the LFA-3 or CD4 molecule. Apart from IFN-γ and IL-2 production by T helper I and T helper 0 cells we have observed production of IL-4 and IL-10 by T helper 2 cells indicating that the GD3 molecule is not a marker for a certain functional T cell subset. In contrast to anti-CD3 mediated activation, the responsiveness of T cells to stimulation via GD3 was dependent on the cell surface expression of the molecule and could be enhanced by costimulation via CD2, CD3, CD26 or CD28. In addition, anti-GD3 antibodies delivered a potent costimulatory signal for antigen-induced proliferation of CD4+ T lymphocytes. In summary, our experiments illuminate the mechanisms of anti-GD3 antibody induced T cell activation.
