    Notes: Summary An enzyme immunoassay for mouse epidermal growth factor (EGF) involving a liquid phase double-antibody system was developed. The EGF-β-galactosidase conjugate prepared was stable for at least 8 months. By this method, EGF was detectable at a concentration as low as 20 pg per tube. The concentrations of EGF in various tissues of mice are also presented.

    Notes: Summary Experimental cerebral vasospasm was produced in a “two-hemorrhage” canine model and examined by immunohistochemistry for leukotriene C4 (LTC4). The immunostain for LTC4 showed a strong positivity in intima and adventitia and a scattered reaction in media of normal basilar artery. The immunoreactivity after subarachnoid hemorrhage (SAH) was little changed in intima and media. Inflammatory cells which were characterized histochemically as neutrophils and macrophages, were shown to infiltrate from the adventitia of basilar artery to the periphery of blood clot after SAH and were markedly immunoreactive for LTC4. Also the neutrophils increased in number with the lapse of time after SAH. Thus, it would be reasonable to conclude that the LTC4 responsible for the development of vasospasm would most likely be produced from the infiltrating neutrophils and macrophages. In addition, neurons in hypothalamus, median eminence, and pons, as well as ependymal and arachnoid cells were immunoreactive for LTC4 both in the control and after SAH, whereas astrocytes and oligodendrocytes were not immunoreactive for LTC4 in either case.

    Notes: Summary Clone MC3T3-E1 cells isolated from newborn mouse calvaria had the same type of alkaline phosphatase (ALP) as that found in adult mouse calvaria (the liver-bone-kidney type), as judged by polyacrylamide gel electrophoresis as well as by heat lability and amino acid inactivation. The effects of prostaglandin E2 (PGE2), parathyroid hormone (PTH), 1,25 dihydroxycholecalciferol [1,25(OH)2D3], and adenosine-3′, 5′-cyclic monophosphate (cAMP) analogs on ALP were investigated. PGE2 and 1,25(OH)2D3 increased ALP activity in dose-related manner with a maximal effect at concentrations of 10 ng/ml and 40 pg/ml, respectively. N6,O2-dibutyryl adenosine-3′, 5′-cyclic monophosphate (DBcAMP) also induced an increase in ALP activity in a dose-related fashion with a maximal effect at a concentration of 0.5 mM which was 2.2-fold over that of the controls. Induced ALP was of the “liver-bone-kidney” type. Antinomycin D and cycloheximide inhibited the increase in ALP activity induced by DBcAMP. The level of ALP was elevated by 8-bromo-adenosine-3′,5′-cyclic monophosphate and theophylline, but not by N6,O2-dibutyryl guanosine-3′,5′-cyclic monophosphate and sodium butyrate. Moreover, PGE2 dramatically increased the level of cAMP in the cells with a maximal effect at a concentration of 10 ng/ml, indicating that PGE2 and DBcAMP induce an increase of ALP activity in clone MC3T3-E1 cells by increasing the cAMP level; PTH did not affect enzyme activity and cAMP, level in the cells. These results suggest that PGE2, DBcAMP, and 1,25(OH)2D3 are involved in bone formationin vivo as well.

    Notes: The diffraction profile for a small crystallite has been obtained as the orientational average of the diffraction intensity given by Ino & Minami [Acta Cryst. (1979), A35, 163-170]. The formula obtained is a type of Debye interference function modified by a function {cal V}(r) (the self-convolution of a crystal shape function) and is expressed as a sum over all the atomic distance vectors in the crystal structure. Since the set of the vectors has Laue symmetry (the order of the group: L), the summation can be simplified to a sum over a reduced range corresponding to 1/L of the original range, while the {cal V}(r) is changed to {\bar v}(r) = ΣLp = 1= {cal V}(Rpr)/(LVt) (Vt: volume of the crystal; R1, . . . , RL: element of Laue symmetry group). Once the {\bar v} function is determined, the profile for a complicated crystal of any size and any crystal system can be systematically and efficiently calculated.

    Notes: A general method of calculating the intensity of X-ray diffraction from small crystalline particles whose boundary is defined by a shape function is discussed. The intensity formula which is generally given by a double sum over the reciprocal-lattice points is simplified into the form of a single sum, using 'the random-shift treatment' which assumes that the position of the boundary relative to the crystal lattice varies at random from crystal to crystal. By the use of Fourier theorems, the intensity formulas are also converted into a single sum over the direct lattice. Although the electron distribution in the particle has been defined in various ways by the shape function, a more reasonable expression of the electron density appropriate to small crystals is introduced. The intensity formulas derived on the basis of the new form of the electron density are compared with other intensity formulas which have so far been proposed.

    Notes: The exact intensity formula of the h-line profile from small crystallites has been obtained as the orientational average of the diffraction intensity of the h plane given by Ino & Minami [Acta Cryst. (1979), A35, 163-170]. While the formula is expressed as a triple integral including a sine Fourier integral, it can be expressed asymptotically with respect to the crystal size according to the theorem of asymptotic expansion of a Fourier integral. Hence the profile can be estimated by the sum of terms of single-integral type. The first term is of the same type as Wilson's formula but it has been shown that second and third terms improve considerably the accuracy of the asymptotic estimation especially for a very small crystal. The h-line profile can be successfully calculated for quite a small crystal of any shape and any crystal system by the asymptotic formula, which can be computed as easily as Wilson's formula.
