    Notes: Abstract A strictly anaerobic bacterium dechlorinating tetrachloroethene (perchloroethylene, PCE) via trichloroethene (TCE) to cis-1,2-dichloroethene (DCE) was isolated from activated sludge with pyruvate plus PCE as energy substrates. The organism, called Dehalospirillum multivorans, is a gram-negative spirillum that does not form spores. The G+C content of the DNA was 41.5 mol%. According to 16S rRNA gene sequence analysis, D. multivorans represents a new genus and a new species belonging to the epsilon subdivision of Proteobacteria. Quinones, cytochromes b and c, and corrinoids were extracted from the cells. D. multivorans grew in defined medium with PCE and H2 as sole energy sources and acetate as carbon source; the growth yield under these conditions was 1.4 g of cell protein per mol chloride released. Alternatively to PCE, fumarate and nitrate could serve as electron acceptors; sulfate could not replace fumarate, nitrate, or PCE in this respect. In addition to H2, the organism utilized a variety of electron donors for dechlorination (pyruvate, lactate, ethanol, formate, glycerol). Upon growth on pyruvate plus PCE, the main fermentation products formed were acetate, lactate, DCE, and H2. At optimal pH (7.3–7.6) and temperature (30°C), and in the presence of pyruvate (20 mM) and PCE (160 μM), a dechlorination rate of about 50 nmol min–1 (mg cell protein)–1 and a doubling time of about 2.5 h were obtained with growing cultures. The ability to reduce PCE to DCE appears to be constitutive under the experimental conditions applied since cultures growing in the absence of PCE for several generations immediately started dechlorination when transferred to a medium containing PCE. The organism may be useful for bioremediation of environments polluted with tetrachloroethene.

    Notes: Abstract A new bacterial strain isolated from soil consumed nitric oxide (NO) under oxic conditions by oxidation to nitrate. Phenotypic and phylogenetic characterization of the new strain PS88 showed that it represents a previously unknown species of the genus Pseudomonas, closely related to Pseudomonas fluorescens and Pseudomonas putida. The heterotrophic, obligately aerobic strain PS88 was not able to denitrify or nitrify; however, strain PS88 oxidized NO to nitrate. NO was not reduced to nitrous oxide (N2O). Nitrogen dioxide (NO2) and nitrite (NO2 –) as possible intermediates of NO oxidation to nitrate (NO3 –) could not be detected. NO oxidation was inhibited under anoxic conditions and by high osmolarity, but not by nitrite. NO oxidation activity was inhibited by addition of formaldehyde, HgCl2, and antimycin, and by autoclaving or disintegrating the cells, indicating that the process was enzyme-mediated. However, the mechanism remains unclear. A stepwise oxidation at a metalloenzyme and a radical mechanism are discussed. NO oxidation in strain PS88 seems to be a detoxification or a co-oxidation mechanism, rather than an energy-yielding process.

    Notes: Summary. In the last few years there has been considerable research on numerical methods for differential algebraic equations (DAEs) $f(x', x, t) = 0$ where $f_{x'}$ is identically singular. The index provides one measure of the singularity of a DAE. Most of the numerical analysis literature on DAEs to date has dealt with DAEs with indices no larger than three. Even in this case, the systems were often assumed to have a special structure. Recently a numerical method was proposed that could, in principle, be used to integrate general unstructured higher index solvable DAEs. However, that method did not preserve constraints. This paper will discuss a modification of that approach which can be used to design constraint preserving integrators for general nonlinear higher index DAEs.

    Notes: Abstract Cell-free extracts obtained from nitrogen-fixing cells of organisms of the family Azotobacteriaceae were analyzed for superoxide dismutases (SOD). For all the representative organisms examined, unusually high specific activities for SOD were recorded. Single Fe-containing SOD were found in Azotobacter vinelandii (5 strains), A. chroococcum (2 strains), A. beijerinckii (1 strain), Azomonas macrocytogenes (2 strains), and Derxia gummosa (2 strains). Highly active, single Mn-containing SOD were found in all of 6 Beijerinckia strains examined (B. indica, B. lacticogenes, B. mobilis, B. fluminensis, B. derxii and B. venezuelae). Multiple SOD were found for Azomonas agilis (Mn-SOD and 2 Fe-SOD), A. insignis (Mn-SOD, Fe-SOD) and Azospirillum lipoferum (2 Fe-SOD) and Azospirillum brasilense (Mn-SOD and 2 Fe-SOD).

    Notes: An archaeon was detected in the leaching solution from a commercial copper production plant and in copper sulfide ores leached with the solution. The leaching solution in this plant contains a high concentration of sulfate salts. Analysis of the microbial population by polymerase chain reaction-amplification of archaeal 16S rDNAs indicated the presence of a single sequence type. Comparison of the nucleotide sequence of the polymerase chain reaction product with available reference sequences suggested that this archaeon corresponds to a new species of a novel genus and family within the order Thermoplasmales. This archaeon grows in synthetic media but it has not been possible to obtain isolates free of chemolithotrophic bacteria.

    Notes: The nearly complete, PCR-amplified, 16S rRNA gene sequences have been determined from the representative type strains of eight xanthomonad phena, including six validly described species of the genus Xanthomonas and Stenotrophomonas maltophilia. Pairwise sequence comparisons and phylogenetic analysis demonstrated that the xanthomonads comprise a monophyletic lineage within the γ-subclass of the Proteobacteria. Although the genus Xanthomonas was observed to comprise a cluster of very closely related species, the observed species-specific primary sequence differences were confirmed through sequencing additional strains belonging to the respective species.
