    Notes: Abstract  Sequences of Aotus nancymaae immunoglobulin kappa light-chain rearrangements were analyzed after reverse transcription polymerase chain reaction. Among 22 in-frame rearrangements analyzed, 12 IGKV genes belonging to the families 1, 2, or 3 were identified. Aotus counterparts for all five human IGKJ genes were found. The identity of the deduced human and Aotus amino acid sequences was between 83% and 92% for junctional regions and 74% for the constant region. Sequence comparisons between rearrangements indicated that somatic mutations, the addition of nongermline-encoded nucleotides, and exonuclease trimming contribute to the generation of diversity of Aotus immunoglobulin kappa chains. The high identity of Aotus and human IGK genes is comparable to that of T-cell receptor genes and further supports the proposal to use the Aotus Plasmodium falciparum infection model for the evaluation of malaria vaccine candidates.

    Notes: Abstract. The cytotoxic effects of the herbicides paraquat (1,1′-dimethyl-4,4′-bipyridylium dichloride), dinoseb (2-sec-butyl-4,6-dinitrophenol) and 2,4-D (2,4-dichlorophenoxyacetic acid) on freshly isolated rat hepatocytes were investigated. Paraquat and 2,4-D (1 – 10 mM) caused a dose and time dependent cell death accompanied by depletion of intracellular glutathione (GSH) and mirroring increase of oxidized glutathione (GSSG). Dinoseb, the most effective cytotoxic compound under study (used in concentrations 1000 fold lower than paraquat and 2,4-D), exhibited moderate effects upon the level of GSH and GSSG. These limited effects are at variance with significant effects upon the adenine and pyridine nucleotide contents. ATP and NADH levels are rapidly depleted by herbicide metabolism. This depletion is observed in the millimolar range for paraquat and 2,4-D and in the micromolar range for dinoseb. 2,4-D completely depletes cellular ATP, with subsequent cell death, as detected by LDH leakage. Paraquat rapidly depletes NADH, according to the redox cycling of the herbicide metabolism. The most effective compound is dinoseb since it exerts similar effects as described for paraquat and 2,4-D at concentrations 1000 fold lower. Simultaneously with NADH and ATP depletion, the levels of ADP, AMP and NAD+ increase in hepatocytes incubated in the presence of the herbicides. In contrast to NADH, the time course and extent of ATP depletion and fall in energy charge correlate reasonably with the time of onset and rate of cell death. It is concluded that the herbicides, paraquat and 2,4-D are hepatotoxic and initiate the process of cell death by decreasing cellular GSH. As a consequence of this primary disturbance, alteration of adenine and pyridine nucleotides contents is a critical event in the induction of irreversible cell injury.

    Notes: Abstract Infections by Listeria monocytogenes are uncommon, with cerebritis being even rarer. We present three cases of cerebritis which occurred during an outbreak of listeriosis. The CT and MR findings at diagnosis and during follow-up are described. Predominant deep white matter lesions with nodular and ring enhancement were seen. The MR yielded a higher resolution of the lesions than CT.

    Notes: Abstract We report a 4-year-old boy, in acute renal failure, who had acute encephalopathy with very high serum aluminium levels (135 μg/l [5μmol/l]) after receiving vesical irrigations with alum. We believe that in situations of non-focal neurological deterioration with no apparent cause in patients with impaired renal function receiving aluminium-containing preparations, the possibility of acute aluminium poisoning should be considered.

    Notes: During the cooling of ceramic materials stresses arise that cause flows of material. Theseflows lead to residual stresses in the materials at the kiln exit. Laboratory tests were carried outusing a composition based on white-firing clays, which was fired according to cycles with differentcooling rates. Residual stresses were measured using the layer removal method. The internaltemperature profile during industrial cooling and the stress generation process were simulated. Theresults of the simulation fit with great accuracy the experimentally measured residual stress values

    Notes: We present a neutron scattering study on methyl group dynamics in glassy toluene. The spectra in the whole temperature range, covering the transition from quantum rotational tunneling to classical hopping, have been successfully analyzed in terms of a potential barrier distribution model. The average barrier in the glass is found to be notably higher than the unique barrier of the crystalline β-phase, whose short-range structure is known to be similar to that of the glass. Due to the mainly intermolecular origin of the interactions on the methyl groups in toluene, it is concluded that the rotational potentials are strongly affected by structural disorder. © 2001 American Institute of Physics.

    Notes: C-phycocyanin and allophycocyanin from the green alga Spirulina platensis were isolated and crystallized by gel-acupuncture techniques. A novel two-step chromatographic procedure was used for purification. Blue hexagonal crystals were obtained by diffusing magnesium chloride into the protein solution for a week, followed by diffusion of PEG 6000 in order to complete the reduction of the solubility of the protein in the capillary tube used as a growth cell. In the case of allophycocyanin, crystals with a size of 0.4 × 0.3 × 0.3 mm were characterized by X-ray diffraction. They belong to space group P6322 with unit-cell parameters a = b = 102.04, c = 131.22 Å. The crystals of C-phycocyanin belong to either space group P6 or P63 with unit-cell constants a = b = 182.38, c = 60.87 Å, α = β = 90, γ = 120°. The crystals diffract beyond 2.4 and 2.5 Å resolution, respectively, using a rotating anode as an X-ray source.
