    Description / Table of Contents: Summary With bone marrow from rats as a model, a new method is evolved for relative measurements of the DNA-synthesis rate in vitro. For that purpose the increase of the14C-thymidine incorporation with the labelling period into the DNA of suspended cells is determined by means of liquid scintillation counting. Amethopterin added to the incubation medium in a final concentration of 1.3×10−4moles/l is able to block the formation of thymidylate by methylation. On these terms, bone marrow cells can cover their requirement of DNA-thymidine from exogenous14C-thymidine only. A concentration of 10−5moles14C-thymidine/l makes an undisturbed DNA-synthesis possible. Suspension of the cells in medium 199 or rat serum does not influence the DNA-synthesis rate measured under these conditions. By comparing the14C-thymidine incorporation with and without amethopterin in the incubation medium, the extent of participation of methylation for thymidylate formation is estimated. With a thymidine concentration of 10−5moles/l it amounts to 30%, of 10−4moles/l to 17%.
    Notes: Zusammenfassung Am Modell des Rattenknochenmarks wird ein neues Verfahren entwickelt, die DNS-Synthesegeschwindigkeit in vitro vergleichend zu messen. Dazu wird der14C-Thymidin-Einbauzuwachs mit der, Inkubationszeit in die DNS suspendierter Zellen durch Flüssigkeitsszintillation bestimmt. Das dem Inkubationsmedium zugesetzte Amethopterin in einer Konzentration von 1,3×10−4Mol/l ist in der Lage, die Bildung von Thymidylat durch Methylierung zu verhindern. Die Knochenmarkzellen können dann ihren Bedarf an DNS-Thymidin nur aus dem exogenen14C-Thymidin decken. Eine14C-Thymidinkonzentration ab 10−5Mol/l erlaubt eine ungestörte DNS-Synthese. Die so gemessene relative Maßzahl der DNS-Synthesegeschwindigkeit ist unabhängig davon, ob die Zellen in Medium 199 oder in Rattenserum suspendiert werden. Durch Vergleich der14C-Thymidininkorporation mit und ohne Amethopterin im Inkubationsmedium wird die Beteiligung der Methylierung an der Thymidylatbildung bestimmt. Sie beträgt bei einer Thymidinkonzentration von 10−5Mol/l 30%, von 10−4Mol/l 17%.
