Summary In 3,979 men and women working in various occupations who on the basis of clinical examination were considered healthy, activities of serum transaminases (GOT, GPT) were measured in the UV-test. New combinations of reagents offering optimal substrate concentrations were utilised for the determinations, since the test-kits obtainable on the market do not provide adequate concentrations of the required substrates. Statistical estimation of normal ranges yielded the following results: 1. The form of distribution of both transaminases can be best described by lognormal distribution; for this reason the normal range cannot be calculated on the basis of arithmetic mean values and standard deviations. 2. Transaminase values are significantly higher in men than in women; furthermore, in the substrate optimum GPT values are always conspicously higher than GOT values in both sexes. No evidence could be supplied for a possible influence of the menstrual cycle or the duration of menopause on measured values. 3. In both sexes GOT- and GPT activities depend significantly on age, body weight and stature. 4. Since the upper normal limits are of major interest from the clinical point of view, the upper tolerance limits were defined and determined for both sexes taking into consideration the influence of age and body weight. 5. On the basis of the results obtained with transaminases, the definition of normality in general and the difficulty of obtaining and utilizing clinical-chemical normal values are discussed.

Zusammenfassung Bei 3979 Werktätigen beiderlei Geschlechts und verschiedener Berufe, die auf Grund einer klinischen Untersuchung als gesund anzusehen waren, wurden die Aktivitäten der Serumtransaminasen (GOT, GPT) im UV-Test gemessen. Für die Messungen wurden neue, substrat-optimierte Reagentienzusammenstellungen verwendet, da in den handelsüblichen Test-Kits die notwendigen Substrate in zu geringen Konzentrationen vorliegen. Bei der statistischen Berechnung der Normalbereiche wurden folgende Ergebnisse erhalten: 1. Die Verteilungsform beider Transaminasen kann am besten durch eine Log-Normalverteilung beschrieben werden; der Normalbereich kann deshalb nicht auf der Basis arithmetischer Mittelwerte und Standardabweichungen errechnet werden. 2. Männer haben signifikant höhere Transaminasewerte als Frauen; im Substratoptimum liegen außerdem die GPT-Werte bei beiden Geschlechtern stets eindeutig über denen der GOT. Die Beeinflussung der Meßwerte durch Menstruationscyclus oder Klimakteriumsdauer ließ sich nicht sichern. 3. Bei beiden Geschlechtern besteht eine signifikante Abhängigkeit der GOT- und GPT-Aktivitäten vom Alter, vom Körpergewicht und von der Körpergröße. 4. Da für klinische Belange hauptsächlich die oberen Normalgrenzen von Interesse sind, wurden für beide Geschlechter unter Einbeziehung der Alters- und Gewichtsfunktionen obere Toleranzgrenzen definiert und errechnet. 5. Anhand der Ergebnisse werden der Normalitätsbegriff im allgemeinen und die Problematik der Aufstellung und des Gebrauchs klinisch-chemischer Normalwerte am Beispiel der Transaminasen diskutiert.
