    Notes: Summary An in vitro method for detecting IgE insulin antibody using the principle of the radioallergosorbent test (RAST) is described. In six patients with insulin allergy, the RAST values were higher than in normal persons or insulin-treated diabetics without insulin-allergy. No differences were observed between normal persons and insulin-treated diabetics without insulin-allergy. Moreover, it was observed that in one patient treated with highlypurified insulin, there was a gradual decrease of RAST value parallel to the radioinsulin binding activity and clinical allergic symptoms. The RAST value of insulin is slightly inhibited by non-IgE antibodies and is, therefore, a semiquantitative value. However, the RAST is simple to perform and reproducible; It therefore is very useful in the detection of IgE insulin antibodies.

    Notes: Summary A competitive radioimmunoassay for the quantitative determination of glycated haemoglobin was developed. The antiserum, obtained by immunizing guinea pigs with reduced glycated human albumin, was capable of identifying and quantitating the glucitollysine residues of glycated Hb after reduction with sodium borohydride. To simplify the sample preparation we introduced trichloroacetic acid precipitation to remove unreacted sodium borohydride instead of using dialysis or gel filtration. Using this procedure, our radioimmunoassay became relatively simple and provided satisfactory within- and between-run (1.3–2.8% and 1.9–5.4% coefficient of variation, respectively). The radioimmunoassay method was compared to the measurement of HbA 1c by high performance liquid chromatography which is the most widely used method for quantitating glycated Hb. For this purpose glycated Hb was measured in normal glucose tolerance, impaired glucose tolerance, and diabetes mellitus groups based on WHO criteria. Both assays were able to discriminate between the normal and diabetic groups. In addition, while the determination of glycated Hb by the radioimmunoassay method was able to clearly discriminate between the normal and impaired glucose tolerance groups, the determination of HbA 1c by the high performance liquid chromatography method failed to discriminate between these two groups. Moreover, 15 of the 20 impaired glucose tolerance patients exceeded the upper normal range (mean normal values + 2 SD) in radioimmunoassay. But all 20 patients with impaired glucose tolerance were within the upper normal range in HbA 1c values. These results demonstrate that the measurement of glycated Hb by radioimmunoassay is more sensitive than the measurement of HbA 1c by high performance liquid chromatography since it can discriminate between the normal and impaired glucose tolerance groups.

    Notes: Summary The clinical and immunological findings are described in a female newborn infant who presented with hypoglycemia associated with a high titre of an insulin-binding protein in her serum. The insulin-binding protein belonged to IgG, the light chain of which consisted solely of Kappa-type. The other properties of the insulin-binding protein were very similar to those of insulin antibodies produced by insulin injection, except for a stronger affinity to human insulin as compared with bovine insulin. The patient's mother had a similar protein in her serum despite no previous history of insulin treatment. It is assumed that the hypoglycemia of this patient might have been due to the presence of the antibody produced in her mother's body against endogenous insulin and transferred to the patient transplacentally.

    Notes: Abstract The pathophysiology of the striatum and cerebral cortex were studied from the pharmacological aspect. Investigation of the dopamine content in the cerebral cortex revealed that the premotor and motor area showed the highest level (61±6.2 ng/g). Intravenous injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) at a dose of 10 mg/kg reduced the dopamine content in the caudate nucleus and putamen to 2–3% of the control level in common marmosets, while it fell to 60% in the nucleus accumbens. There was no alteration of the dopamine content in the cerebral cortex. Immunohistochemical staining for tyrosine hydroxylase in the midbrains of MPTP-treated marmosets showed almost complete disappearance of dopaminergic cells from the substantia nigra and good preservation of cells in the ventrotegmental area. Dopaminergic cells projecting to the caudate/putamen, nucleus accumbens, and cerebral cortex showed marked, moderate, and no vulnerability to MPTP, respectively. After systemic administration of MPTP, dopaminergic neurons projecting to the caudate nucleus and putamen were damaged equally. However, the compensatory increase of dopamine turnover was more prominent in the putamen than in the caudate nucleus. Thus, nigroputaminal dopaminergic neurons may have a higher level of activity than neuron in the caudate. The neural connections and functions of the caudate nucleus and putamen have already been differentiated anatomically or physiologically. This compensatory increase of the dopamine turnover rate is another aspect of functional differences between the caudate nucleus and putamen. Investigation of the dopamine content in the heat, body, and tail of the caudate nucleus showed no differences in the concentration of dopamine. However, a study of the metabolic rate of dopamine using α-methyl-p-tyrosine, a tyrosine hydoxylase inhibitor, showed higher metabolism of dopamine in the head of the caudate nucleus in common marmosets. Thus, dopaminergic neurons projecting to the caudate nucleus may show topographical differences in their firing rates. A microdialysis study indicated an increase in the metabolism of adenosine in the striatum of MPTP-treated animals. Cholinergic neurons are interneurons and are one of the main sources of adenosine in the striatum. Dopaminergic input from the substantia nigra acting on cholinergic neurons was decreased following MPTP treatment. The increase of adenosine metabolism suggested that cholinergic neurons in the striatum receive inhibitory inputs from nigrostriatal dopaminergic neurons.

    Notes: Summary Contact sensitivity (CS) induced by hapten has been thought to be analogous to delayed-type hypersensitivity, such as the Mantoux reaction, because of outstanding similarities between the two phenomena. It can be suggested that animals with CS respond also to intradermal injection of the conjugate of hapten and protein as well as to epicutaneous application of hapten. However, evidence against this has been reported. In the present experiments, delayed-type skin reaction (DSR) was successfully obtained in JY1 strain guinea pigs sensitized by painting the skin with 2,4-dinitrochlorobenzene using in vitro dinitrophenylated epidermal cell suspension (DNP-EC) as antigen for a delayed intradermal test. The experiment using anti-Ia alloantiserum and complement showed that the elicitation of DSR is due to the presence of Ia-positive cells (presumably Langerhans cells) among DNP-ECs. The delayed intradermal test with the conjugates such as haptenated ECs in the animals with CS is considered to be an experimentally useful way of analysing the antigen in the sensitivity.

    Notes: Summary The percutaneous administration of in vitro haptenated epidermal cells (EC) has become established as a procedure to produce contact sensitivity (CS) in experimental animals for routine use. The cells have also been found to elicit a significant delayed-type skin reaction by intradermal test in the animals sensitized by painting the skin with the hapten. The fate of 2,4-dinitrophenylated (DNP) isogeneic epidermal cell suspensions (EC) injected intradermally was investigated histologically in intact or 2,4-dinitrochlorobenzene (DNCB)-sensitized strain 13 guinea pigs to study the role of the cells in CS. DNP-EC were found to proliferate actively in the dermis and formed EC nests with central keratinization and then elicited inflammatory reaction associated with necrosis of the epidermal structures 7 days after injection in the intact animals. DNP-EC injected intradermally into the animals which had received and reacted against DNCB underwent a suppression of EC proliferation. These findings are discussed in relation to the role of the haptenated EC in CS.

    Notes: Summary After a monocular injection of [3H]amino acid into the vitreous chamber of the eye, the distribution of retinal terminations in the pulvinar nucleus of the crab-eating monkey and pigtail macaque was studied by autoradiography. Two groups of labeled terminals were found in the bilateral inferior pulvinar nuclei: one small, dense group was located in the most rostral part of the nuclei and the other, composed of a few small clusters of the labeled terminals, was observed over the medial zone of the middle portion. The terminals were slightly predominant in the contralateral nucleus. A small amount of silver grains showing labeled retinofugal fibers was found in the dorsal surface of the thalamus just medial to the stria terminalis contralateral to the injected site, but termination of these fibers could not be traced in this study.

    Notes: Abstract The influence of plasma lipids and the degree of adiposity on red blood cell (RBC) tocopherol levels was investigated in children. In the first study of nonobese children with a wide range of plasma lipid values, the correlation coefficient between RBC tocopheral levels and plasma tocopherol level was 0.39 (P〈0.001, n=195) but when RBC tocopherol was compared with plasma tocopherol based on plasma total lipids, the coefficient increased to 0.69. From this finding it is assumed that RBC tocopherol levels are governed by the tocopherol concentration in plasma lipids. In the second study of obese children, RBC tocopherol levels decreased in the children with 30% or more excess weight, as compared with those of control children and those with less than 30% obesity, while plasma tocopherol levels were unrelated to obesity grades. Because of no significant difference in plasma total lipid levels among these three groups of children, decreased RBC tocopherol in children with 30% or more obesity is probably attributable to their adiposity.

    Notes: Abstract: Recently, decreased interferon - γ (IFN-γ) and increased interleukin (IL)-4 production have been reported in measurements of the content of the cytokines in culture supernatants of peripheral blood mononuclear cells (PBMC) obtained from patients with atopic dermatitis (AD). These data suggest deviation of PBMC into Th2-reactive cells in AD. In the present study, we examined the frequency of IL-2-, IL-4-, and IFN-γ- producing cells in PBMC with flow cytometry. PBMC from 16 patients with AD and 18 healthy controls were stimulated for 2 days with anti-CD3 and IL-2, and further cultured for 4 days with a maintenance dose of IL- 2. Thereafter these cells were restimulated with phorbol 12-myristate 13- acetate (PMA) and ionomycin for 4 h in the presence of monensin. After fixation of the cells, the cell membranes were made permeable and intracellular cytokines were stained with anti-IL-2, anti-IL-4 or anti-IFN-γ antibody. Cytokine-producing cells were analyzed by gating CD4+ - or CD8+ -subsets. Thus counted frequency of IFN-γ-producing cells was significantly decreased in CD4+ subsets of AD patients (9.9±7.4%) when compared with that in the controls (20.0±6.7%). There was no significant difference either in the frequency of IL-2- or IL-4-producing CD4+ subsets or in that of CD8+ subsets. Furthermore, in the enzyme-linked immunosorbent assay, we also found a decreased production of IFN-γ in the culture of PBMC from AD patients, when compared with those from healthy controls, although it was only at a marginally significant level (P= 0.07). Again there was no increase in IL-4 concentration in AD patients. In addition, we found a weak negative relationship between the disease severity and the frequency of these cells. These results suggest that the decreased frequency of IFN-γ-producing CD4+ cells with subsequently decreased production of IFN-γ play a crucial role in the pathophysiology of AD.
