    Notes: Summary An in vitro method for detecting IgE insulin antibody using the principle of the radioallergosorbent test (RAST) is described. In six patients with insulin allergy, the RAST values were higher than in normal persons or insulin-treated diabetics without insulin-allergy. No differences were observed between normal persons and insulin-treated diabetics without insulin-allergy. Moreover, it was observed that in one patient treated with highlypurified insulin, there was a gradual decrease of RAST value parallel to the radioinsulin binding activity and clinical allergic symptoms. The RAST value of insulin is slightly inhibited by non-IgE antibodies and is, therefore, a semiquantitative value. However, the RAST is simple to perform and reproducible; It therefore is very useful in the detection of IgE insulin antibodies.

    Notes: Summary A competitive radioimmunoassay for the quantitative determination of glycated haemoglobin was developed. The antiserum, obtained by immunizing guinea pigs with reduced glycated human albumin, was capable of identifying and quantitating the glucitollysine residues of glycated Hb after reduction with sodium borohydride. To simplify the sample preparation we introduced trichloroacetic acid precipitation to remove unreacted sodium borohydride instead of using dialysis or gel filtration. Using this procedure, our radioimmunoassay became relatively simple and provided satisfactory within- and between-run (1.3–2.8% and 1.9–5.4% coefficient of variation, respectively). The radioimmunoassay method was compared to the measurement of HbA 1c by high performance liquid chromatography which is the most widely used method for quantitating glycated Hb. For this purpose glycated Hb was measured in normal glucose tolerance, impaired glucose tolerance, and diabetes mellitus groups based on WHO criteria. Both assays were able to discriminate between the normal and diabetic groups. In addition, while the determination of glycated Hb by the radioimmunoassay method was able to clearly discriminate between the normal and impaired glucose tolerance groups, the determination of HbA 1c by the high performance liquid chromatography method failed to discriminate between these two groups. Moreover, 15 of the 20 impaired glucose tolerance patients exceeded the upper normal range (mean normal values + 2 SD) in radioimmunoassay. But all 20 patients with impaired glucose tolerance were within the upper normal range in HbA 1c values. These results demonstrate that the measurement of glycated Hb by radioimmunoassay is more sensitive than the measurement of HbA 1c by high performance liquid chromatography since it can discriminate between the normal and impaired glucose tolerance groups.

    Notes: Summary Immunohistochemical localization of the Ca2+-binding protein parvalbumin (PV) was investigated in the adult human central nervous system (CNS). The antiserum against purified rat skeletal muscle PV specifically recognized certain neuronal populations and their processes. Strongly positive were Purkinje, basket and stellate cells of the cerebellum, cerebral cortical nonpyramidal cells, and neurons in the thalamic reticular and ventrolateral nuclei, subthalamic nucleus, lateral and medial geniculate bodies, vestibular and cochlear nuclei, spinal trigeminal nucleus, cuneate and gracile nuclei, and dorsal nucleus of Clarke. Negative were cortical pyramidal neurons, neurons of the autonomic nerves, and neurons in the caudate nucleus, putamen, dentate nucleus, inferior olive, and substantia gelatinosa. The number and size of PV-immunoreactive neurons were significantly decreased in Alzheimer's disease. However, the decrease was not disease specific.

    Notes: Abstract The distribution of amyloid beta precursor protein (APP) in canine brain was investigated. By immunoblot analysis, APP-positive bands corresponding to proteins of 105–120 kilodalton were recognized in all canine brains regardless of the individual age of the dogs. Bands of similar molecular mass were also detected in the meninges, cerebrospinal fluid, and several visceral organs. Immunohistochemical studies were performed using cryostat and paraffin-embedded sections pretreated with formic acid or by the hydrated autoclave method. In the normal canine brain, APP was found to be distributed in the neurons and vascular system. In the brains with SP, obvious accumulation of APP was observed in swollen neurites within amyloid plaques, although the relationship between APP and diffuse plaques was unclear. APP accumulation in swollen axons was also seen around necrotic foci in the brain of one dog with necrotizing purulent encephalitis. These studies revealed the distribution of APP in canine tissues, especially in the brain, and the accumulation of APP in swollen neurites or axons.

    Notes: Summary The early morphological consequences of recirculation following middle cerebral artery (MCA) occlusion were studied in two rat models. The proximal MCA was occluded for 1 h by either a surgical clip or platelet thrombus; subsequently, 1 h of recirculation was facilitated. Following clip occlusion and recirculation, mild astrocytic swelling, especially around blood vessels, was detected in reperfused cortical and striatal areas. Neuronal changes included slight chromatin clumping and dilation of the rough endoplasmic reticulum. In contrast, severe structural abnormalities were detected following recanalization of the thrombosed MCA segment. Marked astrocytic swelling of cell bodies and perivascular processes with neuropil vacuolation were commonly seen. A heterogeneous pattern of neuronal alterations, including a high frequency of dense shrunken neurons surrounded by swollen astrocytic processes was documented in cortical and striatal regions. Severe neuronal changes were documented in brain regions exhibiting a wellperfused microcirculation. Vascular endothelial cells contained large numbers of pinocytotic vesicles associated with luminal and abluminal surfaces. Pronounced and rapid morphological changes evolve with reperfusion when thrombotic and ischemic events occur simultaneously. The basis for these rapid parenchymal changes following vascular thrombosis may involve acute alterations in cerebral microvascular permeability which exacerbate ischemic consequences.

    Notes: SiC fiber reinforced SiC matrix (SiC/SiC) composites are one of the most promisingmaterials for high temperature structural applications such as power generation and propulsionsystems. SiC/SiC composites are, however, susceptible to accelerated attacks in water vaporenvironments through oxidation and volatilization reaction. For protection from such attacks,Environmental Barrier Coatings (EBCs) are indispensable.We have investigated some oxides and rare-earth silicates as topcoat candidate materials forEBCs. Topcoat materials must be stable in the high-water-vapor pressurized environments at hightemperatures. Also, it is important that the thermal expansion coefficient of topcoat materials issimilar to that of the SiC/SiC composites.In this study, first, zirconium oxides, lutetium silicates and yttrium silicates were selected astopcoat candidate materials. They were exposed in a water-containing atmosphere at a temperature of1673 K for 100 h under a total pressure 0.96 MPa. Mass changes, structure of crystals andmicrostructures were investigated after the exposure experiments in order to evaluate the thermalstability of these materials. After their estimation, lutetium silicates were considered to be promisingfor topcoat materials.Then, lutetium silicates were coated as the topcoat of an EBC system on SiC/SiC composites, andtheir fracture toughness and microstructures were investigated after exposure to an oxidizingatmosphere. The evaluation results of the topcoat materials are reported in this paper

    Notes: 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH) is an azo compound which has been used as a radical chain initiator. The purpose of this study was to confirm contact sensitivity to AAPH in individuals who were engaged in the production of AAPH, and presented with prolonged eczematous eruptions mainly on exposed areas. Patch testing was carried out with AAPH (1 and 5% aq.) on 8 patients and 6 healthy volunteers who had never been exposed to this chemical. All patients showed a strong positive patch test reaction to this agent, while all control subjects showed negative results. Because this chemical has recently been used for studies on the oxidation of biological materials, not only production workers in the chemical industry but also medical researchers should avoid prolonged exposure to this agent.
