    Notizen: : The effect of angiotensin II on catecholamine release from bovine adrenal medulla has been investigated. In retrogradely perfused, isolated bovine adrenal glands, angiotensin II increased basal efflux of catecholamines, but the presence of angiotensin II did not increase the release of catecholamines evoked either by bolus injections of the secretogogue carbachol or by depolarization with a perfusing solution containing a raised concentration of K+. In chromaffin cells maintained in primary tissue culture, angiotensin II increased 3H release from cells preloaded with [3H]-noradrenaline but did not enhance the release evoked by carbachol or by depolarization with K+. The increase in 3H release evoked by angiotensin II from chromaffin cells in tissue culture was inhibited by its analogue antagonist Sar1,Ala8-angiotensin II (saralasin) and was entirely dependent on the presence of Ca2+ in the experimental medium. These findings suggest that, in the chromaffin cells of the bovine adrenal medulla, angiotensin II acts on specific receptors to cause a calcium-dependent catecholamine release but triggers no additional response that acts synergistically with depolarizing or nicotinic stimuli to augment catecholamine release.

    Notizen: Solutions are derived for the partial-differential equations of the rigid model of ion hose with and without an applied axial magnetic field. An upper bound on the amplitude of the beam response, derived from these exact solutions, is compared with the upper bound derived by means of an asymptotic saddle-point calculation. In the exact solutions, explicit boundary conditions are retained. The details of the boundary conditions are significant in understanding the driven ion-hose instability in the experiments. Consideration of the effects of a magnetic field is important because in the ion-hose experiments at Sandia National Laboratories the beam is immersed in an applied axial-magnetic field.

    Notizen: In this paper the concept of a stochastic electron beam is introduced. A stochastic electron beam is one for which the transverse dynamics is chaotic. Utilizing adiabatic orbits, the resonance overlap condition for transition to stochasticity in a particular electron beam system, the "triple-Bennett configuration,'' is derived. The tangent map is derived and used to state conditions for strong orbit instability and the resulting exponential decay of phase correlations. The dependence of the K entropy on the strength ε of the nonaxisymmetry of the triple-Bennett configuration is investigated numerically. Implications for the suppression of the ion-hose instability in the ion-focused regime are briefly discussed.

    Notizen: The linearized tracking force on an electron beam propagating slightly off axis in an Ohmic plasma channel is calculated. Electron inertial effects are included in Ohm's law. The channel of conductivity (σ), density (ne), and radius (a) is surrounded by a region of ambient plasma with conductivity (σ0), density (ne0), and radius (b). Enclosing the system is a highly conducting metallic tank. The electromagnetic fields are calculated using the frozen-field approximation of the Maxwell field equations. The fields are expanded into monopole and dipole moments linearized in the small parameter (d/a), where (d) is the beam displacement. The electron beam is assumed to be a rigid cylinder of charge propagating at the speed of light through the channel and is not allowed to respond dynamically to the force.

    Notizen: Summary Experiments to determine the effects of the catecholamine neuronal uptake blockers cocaine and desipramine, and of the cardiac glycoside, ouabain, upon 3H(noradrenaline) efflux have been performed with bovine adrenal medullary chromaffin cells in tissue culture. Both cocaine and desipramine reduced 3H-noradrenaline uptake into chromaffin cells. Inhibitable uptake was 80% of total accumulation over 60 min; this degree of inhibition was produced by cocaine (30 μmol/l) or desipramine (1 gmol/l). Cocaine (30 μmol/l) had no effect upon spontaneous 3H-efflux measured over 60 min, but reduced that evoked over the same period by carbachol (EC50), veratridine (EC50) and by ouabain (100 gmol/l). Cocaine did not reduce that efflux evoked by raised levels of K+ (28 mmol/l; EC50). Desipramine (1 gmol/l), like cocaine, had no effect upon spontaneous efflux of 3H, but reduced that efflux evoked by carbachol, veratridine and ouabain. Tetrodotoxin (TTX) inhibited veratridine-evoked 3H efflux (IC50 0.2 μmol/l). The degree of inhibition caused by TTX (0.2 μmol/l) was not increased by cocaine (30 μmol/l). TTX also inhibited ouabain-evoked 3H efflux: this was reduced by 55% by a concentration of TTX (1 μmol/l) sufficient to virtually abolish veratridine-evoked efflux. Cocaine (30 gmol/l) in the presence of TTX (1 μmol/l) did not further inhibit ouabain-evoked efflux. Cocaine (30 μmol/l) did not alter 86Rb+ uptake into chromaffin cells, nor did it alter that inhibition of 86Rb+ uptake produced by ouabain (100 gmol/l) indicating that cocaine has no effect upon Na,K-ATPase activity. The results are consistent with the suggestion that both cocaine and desipramine, besides inhibiting catecholamine uptake in bovine chromaffin cells, affect also the nicotinic receptor, or its associated ion-channel, and the Na+-channel opened by veratridine and sensitive to TTX. The data from the K+ experiment suggest that cocaine does not directly affect the voltage-sensitive calcium channel nor the exocytosis step. With respect to the mechanism of action of ouabain, the data show clearly that part of the efflux is brought about by a mechanism which involves the TTX-sensitive Na+ channel, and, that the remaining part is independent of the neurotransmitter uptake process.
