    Notes: Summary An account is given of an experimental design and a computing procedure for in vivo measurement of the blood-tissue exchange of amino acids and the metabolic rate of tissue proteins with radioactively labelled amino acids. The method was used for evaluation of the exchange rates of tyrosine between the plasma and the brain and between the free and protein-bound tyrosine compartments in the brain of adult rats in experimental hyperphenylalanaemia and hypertyrosinaemia. Hyperphenylalanaemia inhibited the exchange of tyrosine between plasma and brain. In both hyperphenylalanaemic and hypertyrosinaemic rats the rate of synthesis of the cerebral proteins fell. Alterations in the intracerebral pool of free amino acids produced by excessive loading with phenylalanine or tyrosine are suggested as the cause of the impairment of cerebral protein synthesis.

    Notes: Summary The efflux and exchange of L-tryptophan (Trp) from rat cerebral cortex slices were studied in a superfusion system. The substrate specificity of Trp exchange was assessed by measuring the stimulation of [3H]Trp exit provoked by other extracellular amino acids. Large neutral amino acids were the most potent, but also glutamic acid, lysine and glycine had some effect. The stimulation caused by extracellular Trp and phenylalanine persisted also at 0 ° C though severalfold attenuated. Only intracellular histidine provoked slight inhibition of [3H]Trp efflux and intracellular Trp, phenylalanine and lysine had a small stimulatory effect. The results suggest an involvement of carrier-mediated processes in the exchange and efflux of Trp. The substrate specificities of the exchange and efflux are not apparently identical.

    Notes: Summary Brain slices from adult and newborn rats were incubated with [35S]taurine in Krebs-Ringer bicarbonate medium at pH 7.4. In both age groups electrical stimulation increased the oxygen consumption of the slices. The influx of taurine only increased in slices from adult rats. The influx conformed to Michaelis-Menten kinetics. In slices from adult rat Vmax increased and apparent Km remained unchanged during electrical stimulation. Both Km and Vmax were greater in adult than in newborn rats. The efflux of taurine from slices depended on the intracellular concentration of taurine and was not measurably influenced by electrical stimulation.

    Notes: Abstract: Depolarization-induced release of [3H]γ-aminobutyric acid (GABA) and [3H]noradrenaline (NA) from rat cerebral cortex slices was studied in two superfusion systems: one with stationary and the other one with continuously shaken slice compartments. Calcium-dependent depolarization-induced release of GABA and NA could be demonstrated only with shaken slices. GABA, but not NA, could also be released by high K + media and veratridine from stationary slices. Synaptic transmitterreleasing mechanisms are apparently damaged in stationary slices, possibly due to impaired energy metabolism.

    Notes: Abstract: Mouse brain slices take up hypotaurine (2-aminoethanesulphinic acid) from medium by means of two concentrative low- and high-affinity transport systems. [35S]Hypotaurine uptake by the slices was significantly reduced in the absence of external potassium, calcium, or magnesium ions. An excess of potassium ions also inhibited hypotaurine uptake by one-half. Uptake was almost completely abolished on removal of sodium ions. The Km constants for both low- and high-affinity transport components increased in a low-sodium medium, suggesting that sodium ions are required when hypotaurine is attached to its possible carrier sites in plasma membranes. Sodium ions also mimicked allosteric effectors of hypotaurine transport, showing positive cooperativity. More than two sodium ions may be involved in the transport of one hypotaurine molecule across the cell membrane. The calculated activation energies of transport were fairly similar in normal and sodium-deficient media and thus sodium ions may not participate in the activation mechanisms of the transport. With respect to cation dependence, hypotaurine transport in brain slices exhibits features characteristic of neurotransmitter amino acids.

    Notes: Abstract: Uptake of [35S]hypotaurine by brain slices prepared from adult and 8-day-old mice was studied at varying temperatures, under O2 and N2 atmospheres, and in the presence of metabolic inhibitors and varying concentrations of hypotaurine in the incubation medium. The tissue/medium concentration gradients generated were exceptionally high for an amino acid. Hypotaurine uptake was energy- and temperature-dependent, more strictly in adult mice. Uptake was saturable, containing a high-affinity and a low-affinity component. The estimated transport constants for the high-affinity uptake of hypotaurine (8-day-old mice, 17.2 μ mol/liter; adults, 35.3 μ mol/liter) were of the same order of magnitude as the reported transport constants of putative amino acid transmitters, but the total transport capacity appears to be greatest for hypotaurine.

    Notes: —The proteolytic activity of brain homogenates obtained from 1-, 5-, 14-, 60-, 150-, and 300-day-old rats was assayed with urea-denatured haemoglobin and casein, endogenous tissue proteins, Nα-benzoyl-dl-arginine 2-naphtylamide (BANA), Nα-benzoyl-dl-arginine methyl ester (BAME), Nα-toluene p-sulphonyl-dl-arginine methyl ester (TAME), Nα-benzoyl-dl-phenylalanine 2-naphthyl ester (BPANE), and Nα-acetyl-dl-tyrosine ethyl ester (ATEE) as substrates.Several peaks of activity were detected with all these substrates in different pH ranges. Activity was highest with protein substrates at pH 3·0-4·0, with smaller peaks of activity at pH 5·5-6·5 and 8·0-9·0. At pH 3·0 the activity with trypsin substrates, viz. BANA, BAME and TAME, was also relatively high, but much less with chymotrypsin substrates, ATEE or BPANE. With BAME, TAME, BPANE and ATEE the hydrolysis rate was highest at neutral or slightly alkaline pH. During postnatal development the hydrolysis of protein substrates increased three-fold at pH 3·0 and about two-fold at pH 6·5 and 8·5. The rate of hydrolysis of BANA, BAME and TAME generally increased during the first 2 postnatal weeks and thereafter decreased, whereas no marked increase in the rate of hydrolysis of BPANE and ATEE occurred until the age of about 2 weeks. The results were less consistent with synthetic substrates than with protein substrates, indicating the existence of non-uniform alterations during development in the activity of the individual hydrolytic enzymes participating in the breakdown of brain proteins.
