    Notes: Summary Prostacyclin (PGI2) is a potent vasoactive prostanoid regulating vascular tone. We recently purified and cloned a PGI2-stimulating factor (PSF), which stimulates PGI2 production by vascular endothelial cells (ECs). Previous study demonstrated that PSF is predominantly located in vascular smooth muscle cells (SMCs) and present in serum. PSF may act on vascular ECs to regulate PGI2 synthesis for maintaining vessel wall homeostasis. Decreased PSF production in the vessel wall may result in an imbalance of prostanoid synthesis, leading to the development of vascular lesions such as diabetic angiopathy. In the present study, to investigate the regulatory mechanisms of PSF gene expression, we examined the effect of high glucose concentrations on PSF mRNA expression in cultured bovine aortic SMCs. Expression of PSF mRNA was significantly decreased to 66 ± 6 % of control value (p 〈 0.01), when the glucose level was raised from 5.5 to 27.8 mmol/l. We also examined the effect of osmolarity on PSF mRNA expression by addition of an appropriate dose of mannitol to the culture medium. We confirmed that high glucose concentration itself reduced the expression of PSF mRNA and glucose had much more effect than the osmolarity control. The expression of PSF mRNA was significantly decreased to 72 ± 5 % of control value (p 〈 0.05) by a protein kinase C (PKC) activator, phorbol-12-myristate-13-acetate (PMA). The decreased expression of PSF mRNA in the presence of high glucose or PMA was restored by co-incubation with a PKC-specific inhibitor (GF109203X). These results suggest that PSF gene expression in vascular SMCs may be decreased via a specific effect of high glucose concentrations. High glucose-induced activation of PKC is suggested to participate partly in the regulation of PSF gene expression. [Diabetologia (1998) 41: 134–140]

    Notes: Summary In a study of experimental autolysis of peripheral nerves of normal Wistar rats, crystalloid inclusions similar to Hirano bodies were observed. They occurred in the nuclei and cytoplasms of Schwann cells and in myelinated axons. They first developed after a 12-h autolysis and increased in number and size with time. Their fine structure, localization, and morphogenesis are discussed in relation to the autolysis of the cellular organelles.

    Notes: Abstract The incidence of ovigerous females in populations of two grapsid crabs, Hemigrapsus penicillatus (de Haan, 1835) and Sesarma (Parasesarma) pictum (de Haan, 1835) were followed from August 1975 to November 1976. H. penicillatus, which inhabits the lower intertidal region near the mouth of Tatara-Umi Estuary, breeds from March to November. S. pictum, which inhibits crevices and abandoned holes of other species and is abundant at and above the upper intertidal region of the estuary, breeds from May to September. H. penicillatus matures when the female reaches 6 to 7 mm carapace width, whereas S. pictum becomes mature when the carapace width is 12 to 13 mm, although the maximum size attained by females of both species is almost the same. H. penicillatus produces 5 to 6 broods, S. pictum 2 to 3 broods, during a breeding season. The size of a newly laid egg of H. penicillatus is smaller than that of S. pictum. The number of eggs produced by a female H. penicillatus is greater than that of S. pictum during a breeding season. In both species the peak of the breeding season is during summer, at which time the ovarian activity is also apparently accelerated. The major environmental factor which controls the breeding in these crabs appears to be temperature. H. penicillatus is submerged at every high tide, and is relatively inactive from late November to February when the ambient water and air temperatures are rather low. S. pictum is dormant in crevices or understones of the splash zone from November to March. The length of the breeding season of these crabs appears to be inversely proportional to the period of their winter dormancy.

    Notes: Abstract To make clear how then-hexane metabolism is modified by co-exposure with MEK, rats were exposed to various concentrations of MEK mixed with a fixed concentration ofn-hexane. Twenty-four male Wistar rats were divided into four equal groups. Each group was exposed for 8 h to 2000 ppmn-hexane, 2000 ppmn-hexane plus 200 ppm MEK, 2000 ppmn-hexane plus 630 ppm MEK and 2000 ppmn-hexane plus 2000 ppm MEK, respectively. Free metabolites and the sum of free and conjugated metabolites ofn-hexane were analyzed by gas chromatography. The main metabolite was 2-hexanol during the exposure and 2,5-hexanedione (2,5-HD) after the exposure in any group. The main metabolites, 2-hexanol and 2,5 HD, decreased in iverse proportion to the co-exposed MEK concentrations. The results suggest that augmentation ofn-hexane neurotoxicity by MEK co-exposure could not be explained only by 2,5-HD. In addition, 2,5-HD is recommended as an index for biological monitoring ofn-hexane exposure. However, one should be careful to evaluate the exposedn-hexane concentration by urinary 2,5-HD, becausen-hexane metabolism could be largely modified by co-exposure with MEK.

    Notes: Summary Flow studies using dynamic CT and xenon (Xe) CT were carried out in 25 patients with ischaemic stroke in the territory of the middle cerebral artery to define the clinical characteristics of cerebral ischaemia at a chronic stage. The parameter of peak height/mean transit time (PH/MTT) obtained from dynamic CT can provide an accurate index for blood circulation in the cerebral vascular bed. Xe CT measurements revealed various kinds of ischaemia around the infarction even in the chronic stages. In mild ischaemia of more than 30ml/100g/min, reduction of cerebral blood flow (CBF) was well correlated to the PH/MTT. However, in severe ischaemia between 20 and 30 ml/100 g/ min, changes of CBF were no longer correlated with the PH/MTT. There were cases showing severe reduction of CBF but which showed sufficient blood circulation (moderate value of PH/MTT). Mild reductions of CBF in parallel with decreased blood supply were often found in the peri-infarct area of infarctions in the centrum semiovale. On the other hand, infarctions in the cortico-subcortical region showed severe ischaemia, in even where blood circulation was relatively well sustained.

    Notes: Summary  Eight patients with ganglioglioma who received surgical treatment at our institute between January 1989 and January 1997 were reviewed to determine their clinical, CT and MRI characteristics. Tumours were located in the temporal lobe (four patients), trigone of the lateral ventricle (two patients), basal ganglia (one patient) and fronto-temporal lobe (one patient). On imaging, two types of tumours were seen, a solid mass in 5 patients (62.5%) and a cystic mass in three patients (37.5%). Six complete tumours (75%) and all of the solid components of the cystic tumours were enhanced by contrast medium. Seven tumours (88%) had no peripheral oedema. On CT, the tumours being studied appeared as iso-(62.5%) or low density (37.5%) intra-axial tumours. Four tumours (50%) contained calcification. On MRI, the tumours appeared as well-circumscribed, iso- (62.5%) or low intensity (37.5%) intra-axial tumours on T1 weighted images, and as high (75%) on T2 weighted images. Three underwent total resection, 2 subtotal resection and 3 partial resection. No patients had have any further treatment such as radiation therapy or chemotherapy. Postoperative sudies were conducted on all patients with an average follow-up period 56 months (range 4–147 months) after surgery. There was no evidence of recurrence of tumours or of growth of residual tumours. We observed gangliogliomas which were located in unusual regions such as the trigone in two of the patients. To our knowledge, our series is the first report to describe trigonal gangliogliomas. We conclude, therefore, that ganglioglioma should be included as a possibility in the differential diagnosis of intracranial masses, even when they are located in the trigone.
