    Notes: Abstract Burimamide, a histamine H2-receptor antagonist, has been shown to cause pressor responses in pithed rats. The response can be prevented by prior removal of the adrenal glands or by pretreatment with the α-adrenoceptor antagonist, phentolamine, 5 mg/kg, suggesting that the pressor response to burimamide is due to release of catecholamines from the adrenal glands. The pressor activity of burimamide has been compared with that of metiamide and two close chemical analogues, methylburimamide and thiaburimamide, in order to identify which chemical features of the compounds are necessary for this activity. Methylburimamide was the most potent pressor agent, followed by burimamide, metiamide and thiaburimamide. The pressor effects (and presumably catecholamine-releasing activities) appear to be related to the basicities of the compounds. We conclude that the release of catecholamines by these histamine H2-receptor antagonists is probably due to their cationic (imidazolium) forms.

    Notes: Abstract Intra-arterial infusions of histamine cause vasodilatation of resistance vessels, extravasation of albumin and oedema in cat skeletal muscle. Treatment with either mepyramine or metiamide significantly reduced the extravasation of albumin and rate of oedema formation. The doses of mepyramine and metiamide used did not modify the vascular responses to bradykinin.

    Notes: Abstract The vascular changes due to thermal injury have been determined in rat hind paws. In anaesthetized rats one hind paw has been injured by immersion in water for 30 seconds; temperature range 49–65°C. Changes in blood flow, blood content and albumin content have been determined and compared with values from uninjured rats. The swelling caused by thermal injury was temperature-dependent, the higher temperatures caused the larger swellings. There were also temperature-dependent increases in blood flow to the injured paw and albumin content of the injured paw. Changes in blood content only occured at the highest temperatures. Swelling, increased blood flow and albumin content could be detected within 1 minute of injury. The swelling and albumin content of the tissue then continued to increase gradually. The increase in blood flow was greatest 1 minute after injury and then stabilized at a flow some 4–5 times higher than normal paw flow. The method described can be used to measure vascular changes due to other inflammatory stimuli and could be applied to measure changes in other discreet areas.

    Notes: Abstract S-[3-(N,N-Dimethylamino)propyl]isothiourea (dimaprit), has been shown to be a highly specific histamine H2-receptor agonist. Parallel line assays showed that in vitro at H2-receptors it had approximately 17.5% the activity of histamine on the rat uterus and 71% on the guinea-pig right atrium, with similar maximal responses; it had less than 0.0001% the activity of histamine on H1-receptors. Dimaprit stimulated gastric acid secretion in the rat, dog and cat in which it had, respectively, approximately 19, 58 and 400–500% the activity of histamine. In the dog and cat the maximum secretory response to dimaprit was significantly greater than that obtained to histamine. The H2-receptor specificity of dimaprit in causing depressor and vasodilator effects was also demonstrated in the cat, in which it had 18–20% of the H2-receptor activity of histamine. Dimaprit should prove to be a very useful tool in studies examining the role of histamine in physiology and pathology. The absence of marked cardiovascular effects at doses maximal for the stimulation of gastric acid secretion, as seen in the cat studies, could lead to this compound being of value as a diagnostic agent in the measurement of maximal acid secretory capacity.

    Notes: Abstract The pharmacology of histamine-induced increases in microvascular permeability has been studied in rat skin. Histamine caused dose-dependent increases in microvascular permeability, assessed as increases in extravascular albumin accumulation. The responses to histamine were inhibited in a dose-dependent manner by pretreatment with mepyramine and were not changed by cimetidine. 2-(2-Aminoethyl)pyridine also increased microvascular permeability whereas impromidine did not. These results suggest that H1-receptors and not H2-receptors are involved in the permeability response to histamine in rat skin. In contrast, dimaprit increased microvascular permeability and responses to dimaprit exceeded the maximum response to histamine. The response to dimaprit proved to be independent of H2 receptors and was consistent with an indirect response due to mast cell degranulation.

    Notes: Abstract We report a case of intraductal papillary adenocarcinoma of the distal pancreatic duct. Although a rare subgroup of exocrine pancreatic tumors, their diagnosis is an important one in view of their favorable prognosis with pancreatectomy. Because of the established behavior of villous tumors of the colon, to which they are similar histologically, these tumors should be resected, even if biopsy shows benign disease. Patients should be followed radiologically for recurrence or the development of new adenoma following resection.
