    Notes: Abstract The concentrations and chemical composition of water-extractable P were compared in four soil types from NE Scotland. All sites were sampled during the early establishment phase of a spring barley (Hordeum vulgare) crop. The quantity of total soluble P extracted ranged from 〈2.0 to 10 mg P kg soil-1, of which up to 50% was classified as being organically associated. Sample fractionation showed that both orthophosphate inorganic P and organic P were associated with a wide molecular-size range of organic material. A strong positive correlation was readily apparent between P and the sum of Fe + Al in the fractionated samples. The extent of enzymatic hydrolysis of organic P varied between soil samples and the type of enzyme. Phytase consistently produced the greatest degree of hydrolysis.

    Notes: Abstract Seeds from two varieties of spring barley (Prisma and Camorgue) were grown axenically in water. After 14 days, the culture solutions contained organic P substances (about 4 μg P per plant) derived from root exudation, representing about 3% of the total P found in the seed. Gel filtration, separated the organic P into two well defined peaks, one with a high molecular weight (〉45000 daltons) and the other with a low molecular weight (〈500 daltons). The bioavailability of the soluble organic P released was assessed enzymatically and chemically. At the optimum pH of 5.0, phytase and acid phosphatase hydrolysed about 80% and 65%, respectively of the organic P in the exudate after 24 h whereas at the optimum pH of 9.8, alkaline phosphatase hydrolysed up to 40% P after the same length of time. In a pH 5.0 buffer, up to 10% of the organic P was hydrolysed compared with up to 45% in a pH 9.8 buffer. The high molecular weight organic P fraction recovered from the G-75 Sephadex behaved similarly.

    Notes: Abstract Optical energy balance in plasmas produced by irradiating solid targets by highpower laser pulses, has been experimentally determined. The total light scattered has been measured as a function of the laser intensity and the irratiated spot diameter. It has been found that the net light absorption does not depend on the laser intensity alone, but is also affected by the irradiated spot area. It has also been observed that diffuse back reflection becomes a dominant loss mechanism at high incident flux and small irradiated spot area.

    Notes: Interaction of elliptically polarized laser radiation with plasma produced from a solid target generates magnetic fields simultaneously along axial and transverse directions. The axial magnetic field induces the plane of polarization of the incident laser beam to rotate due to the Faraday effect. Numerically the Faraday rotation has been calculated for interaction of Nd glass laser (λ=1.06 μm) in the subdense plasma in which the plasma density, temperature, and self-generated magnetic field vary in the axial direction.

    Notes: Abstract: Cyclin-dependent kinase, Cdk5, has been identified in neural tissue in connection with neurofilament and τ protein phosphorylation. This report describes the characterization of a 62-kDa protein that copurifies with Cdk5 from rat spinal cord homogenates. Dissociation of the protein from neural Cdk5 is concomitant with a reversible loss in kinase activity. Amino acid sequence information from tryptic peptide fragments was used to clone the complementary DNA from rat brain. A single full-length cDNA was characterized coding for a 67.5-kDa protein (p67). Exogenously expressed p67 stimulated Cdk5 kinase activity in vitro in a dose-dependent manner and when presented as an affinity matrix, selectively adsorbed Cdk5 from a cleared rat brain homogenate. In situ hybridization analysis of E18 rat embryos and adult rat brain demonstrated that p67 transcript expression is restricted to neural tissue. Immunohistochemical staining with an amino-terminal peptide-specific antibody further indicated that p67 is exclusively expressed in neurons. Localization in vivo and in cultured rat hippocampal neurons showed that p67 is highly enriched in axons. We propose that p67, by virtue of its regulation of Cdk5, participates in the dynamics of axonal architecture through the modulation of phosphorylation of cytoskeletal components.

    Notes: Abstract: The squid giant axon and extruded axoplasm from the giant axon were used to study the capacity of axoplasm for phospholipid synthesis. Extruded axoplasm, suspended in chemically defined media, catalyzed the synthesis of phospholipids from all of the precursors tested. 32P-Labeled inorganic phosphate and γ-labeled ATP were actively incorporated into phosphatidylinositol phosphate, while [2-3H]myo-inositol and l-[3H(G)]serine were actively incorporated into phosphatidylinositol and phosphatidylserine, respectively. Though less well utilized, [2-3H]glycerol was incorporated into phosphatide acid, phosphatidylinositol, and triglyceride, and [methyl-3H]choline and [1-3H]ethanolamine were incorporated into phosphatidylcholine and phosphatidylethanolamine, respectively. Isolated squid giant axons were incubated in artificial seawater containing the above precursors. The axoplasm was extruded following the incubations. Although most of the product lipids were recovered in the sheath (composed of cortical axoplasm, axolemma, and surrounding satellite cells), significant amounts (4–20%) were present in the extruded axoplasm. With tritiated choline and myo-inositol, the major labeled phospholipids found in both the extruded axoplasm and the sheath were phosphatidylcholine and phosphatidylinositol, respectively. With both glycerol and phosphate, phosphatidylethanolamine was a major labeled lipid in both axoplasm and sheath. These findings demonstrate that all classes of phospholipids are formed by endogenous synthetic enzymes in axoplasm. In addition, we feel that the different patterns of incorporation by intact axons and extruded axoplasm indicate that surrounding sheath cells contribute lipids to axoplasm. A comprehensive picture of axonal lipid metabolism should include axoplasmic synthesis and glial-axon transfer as pathways complementing the axonal transport of perikaryally formed lipids.
