    Description / Table of Contents: Summary In addition to an increase of the erythropoesis and myelopoesis and an increase of the reticulum cells a 5-year-old boy with erythroleukemia showed immunological phenomena: positive Coombs' test, hypergammaglobulinemia, nucleophagocytosis and normoblasts with a reticulum cell showing central storage which were arranged in a rosette-like manner. Genuine LE-cells could not be demonstrated. These findings correspond to the “syndrome resembling lupus-erythematosus.” Chromosome analysis of 250 bone-marrow cells in the final stage of hemoblastosis which could not be affected by treatment shows a mosaic of at least 3 cell lines: 46 XY/46, XY, C-, r+/47, XY, C-, 2r. The authors discuss the cytogenetic findings of the cases with erythroleukemias hitherto examined. Hypodiploidias are predominant.
    Notes: Zusammenfassung Bei einem 5jährigen Jungen mit Erythroleukämie fanden sich neben der gesteigerten Erythro- und Myelopoese sowie einer Vermehrung von Retikulumzellen immunologische Phänomene: positiver Coombs-Test, Hypergammaglobulinämie, Nukleophagozytose und rosettenartig angeordnete Normoblasten mit zentral speichernder Retikulumzelle. Echte LE-Zellen waren nicht nachzuweisen. Die Befunde entsprechen dem “Lupus-erythematodes-ähnlichen syndrom”. Die Chromosomenanalyse von 250 Knochenmarkzellen im Finalstadium der therapeutisch unbeeinflußbaren Hämoblastose zeigte ein Mosaik von mindestens 3 Zellinien: 46 XY/46, XY, C-, r+/47, XY, C-, 2 r. Die zytogenetischen Befunde der bisher untersuchten Fälle von Erythroleukämien serden diskutiert. Hypodiploidien überwiegen.

    Description / Table of Contents: Zusammenfassung Die Versuche, Metaphasechromosomen und Zellkerne mit den Acridinderivaten Acranil und Atebrin zu fluorochromieren, haben gezeigt, daß sich das Y-Chromosom durch seine intensive Fluorescenz von allen anderen Chromosomen abhebt. In Interphasekernen zeigt ein leuchtender Punkt („F-body“) das Vorhandensein eines Y-Chromosoms an. Er kann auch als Doppelpunkt erscheinen. Der F-body wird in 80% der Epithelkerne (Mundepithelien, Amnionzellen) — Färbung vorzugsweise mit Acranil — und in 98% der Leukocytenkerne (Granulocyten, Lymphocyten) — Färbung mit Atebrin — festgestellt.
    Notes: Summary Staining of the human metaphase chromsomes and interphase nuclei with “Atebrin” as well as “Acranil” have revealed intensive fluorescence of the Y chromosome. A brightly fluorescent spot (“F-body”) in interphase nuclei suggests the presence of one Y chromosome. The spot may be doubled. The F-body is seen in 80% of epithelial cells (from buccal smears or amniotic fluid)-the preferable stain being Acranil-, and in 98% of leucocytes,-the preferable stain being Atebrin.

    Notes: Summary Chromosomal analysis in a child with ambiguous sex showed mosaicism of at least two cell lines with one or more marker chromosomes or none at all. They were shown to be derived from the Y chromosome by fluorescent in situ hybridisation (FISH) using different DNA probes that cover parts of the long and the short arm.

    Notes: Abstract This paper reports observations on 6 new patients with the Dubowitz syndrome which was first defined by Grosse et al., in 1971 and which is a recessively inherited, pleiotropic malformation syndrome including variable degrees of intrauterine growth retardation and primordial shortness of stature, microcephaly, mental retardation, eczema, and a characteristic appearance, voice and combination of minor anomalies. Data in the present report show that eczema can be absent, and patients can be of normal height, and of normal intelligence in spite of a head circumference which has so far always fallen below the third percentile. So far 11 patients (8 females and 3 males) are known with the Dubowitz syndrome; in one family the parents were first cousins.

    Description / Table of Contents: Zusammenfassung Neben dem klassischen Klinefelter-Syndrom mit der Chromosomenkonstellation 47,XXY stehen heute eine Reihe von Varianten, deren häufigste 46,XX, 48,XXYY, 48,XXXY und 49,XXXXY sind. Auf Grund seiner unverkennbaren Symptomatologie nimmt das XXXXY-Syndrom unter ihnen eine Sonderstellung ein. Mindestens 79 Fälle dieses Karyotyps wurden bis Ende 1972 in der Literatur mitgeteilt. 7 neue Fälle dieses seltenen Syndroms werden beschrieben. Häufige Symptome sind die intrauterine Dystrophie, die erhebliche psychomotorische Retardierung, das hypoplastische Genitale, eine eigenartige Facies und Skeletanomalien, die vor allem Unterarm und Hand betreffen (radioulnare Synostose, Klinodaktylie oder Brachyphalangie der 5. Finger, Pseudoepiphysen der Mittelhandknochen). Seltenere Symptome sind Augenfehler und angeborene Vitien, insbesondere ein offener Ductus Botalli. Mit Hilfe der Xg-Blutgruppenanalyse war es mehrfach möglich, den Nachweis zu erbringen, daß alle 4 X-Chromosomen mütterlicher Herkunft sein müssen. Als Ursache der Polysomie des X-Chromosoms wird daher in diesen Fällen eine sukzessive Non-disjunction diskutiert.
    Notes: Abstract In addition to 47,XXY Klinefelter's syndrome, several variants have been described. The anomalies most frequently found have the karyotypes 46,XX, 48,XXYY, 48,XXXY, and 49,XXXXY. Because of its unequivocal symptomatology the 49,XXXXY chromosome aberration has a prominent place among them. Seven new cases of this rare syndrome are described and 79 cases published up to 1972 are reviewed. Frequent symptoms are prenatal dystrophy, marked psychomotor retardation, hypoplastic genitals, the peculiar facial features, and skeletal malformations affecting particularly the forearms and hands (radioulnarsynostosis, clinodactyly or brachyphalangy of the fifth fingers, metacarpal pseudoepiphyses). Rare symptoms are ocular malformations and congenital heart defects, especially an open arterial duct. Xg blood group analysis made it possible to demonstrate the maternal origin of all four X chromosomes in some cases. In these cases successive non-disjunction of maternal X chromosomes is discussed.

    Description / Table of Contents: Zusammenfassung Bei einem geistig und statisch retardierten Kind mit Skaphocephalie und Hypospadie wurde eine Translokation zwischen den Chromosomen D2 und G1 festgestellt. Die gleiche Aberration und zusätzlich ein kleines akrozentrisches Extrachromosom fand sich bei der gesunden Mutter des Kindes. Daraus wird geschlossen, daß bei der Mutter eine genetisch balancierte reziproke Translokation D2/G1 vorliegt und deshalb bei dem Kind eine Defizienz beider Chromosomen vorhanden sein muß. Die Symptomatik wird mit ähnlichen Beobachtungen von Defizienzen akrozentrischer Chromosomen verglichen.
    Notes: Summary Translocation between one chromosome D2 and one G1 has been found in a child with psychomotor retardation, scaphocephalus and hypospadias. The child's healthy mother showed the same aberration and an additional small acrocentric chromosome. It is concluded that the mother has a genetically balanced reciprocal translocation D2/G1 causing deficiency of both chromosomes in the child. The symptomatology is compared with similar reports concerning acrocentric deficiencies of chromosomes.

    Description / Table of Contents: Zusammenfassung Die Diagnose der Neurofibromatose im Kindesalter wird erschwert durch die Tatsache, daß meist nur Pigmentflecken, nicht aber die charakteristischen Haut-und Nerventumoren vorhanden sind. Von 16 zwischen 1951 und 1966 in der Universität-Kinderklinik Münster beobachteten Patienten wurden 4 mit einer ungewöhnlichen Symptomatik ausgewählt. Bei einem Mädchen lag eine auf den Urogenitaltrakt beschränkte NF mit Klitorisvergrößerung vor. Ein Junge litt an einem fixierten renalen Hypertonus infolge Nierenarterienstenose, die von zahlreichen weiteren Gefäßveränderungen in der Niere begleitet war (vasculäre NF n. Feyrter). Die große Variabilität der Erscheinungen der NF wurde am Beispiel eineiiger Zwillinge demonstriert. Nur bei einem Zwilling fanden sich Nerven-und Hauttumoren im Kopf-und Gesichtsbereich. Ein Rankenneurom mit Hypertrophie einer Gesichtshälfte und Buphthalmus trat bei einem anderen Mädchen bereits sehr früh in Erscheinung. Die pathognomonischen Café au lait-Flecken wurden in keinem Fall vermißt. Sie sind das verläßlichste Hinweiszeichen der NF im Kindesalter, das stets eine gründliche klinische Untersuchung veranlassen sollte.
    Notes: Summary Diagnosis of neurofibromatosis in children is difficult since most often only pigment spots are present instead of the typical cutaneous and nervous tumors. Four out of 16 patients of the Children's Clinic in Münster observed during the period of 1950 to 1966 were selected for unusual manifestations. In one girl neurofibromatosis was limited to a hypertrophy of the clitoris and to the urogenital tract. A boy suffered from renal hypertension caused by stenosis of the renal artery and multiple vascular abnormalities of the kidney. The great variability of symptology is demonstrated by monozygotic twins. One twin only exhibited craniofacial tumors of the skin and nerves. In another girl plexiform neurinoma with facial hypertrophy and buphthalmos was observed in early infancy. Since in all cases the typical café-au-lait spots were present. They can be considered to be the most reliable symptoms which always demand a thorough clinical examination of the patient.
