    Notes: Human T lymphocytes, upon phytohaemagglutinin stimulation, are able to form colonies in semisolid media. Peripheral T cells bearing Fc receptors (TG) were studied for their possible regulatory activity on lymphoid colony development. Although no substantial differences were observed between the cloning efficiency of unfractionated T cells (thus including TG cells) and T cells depleted of TG, a sharp suppression of colony formation occurred when positively selected TG cells were readded to TG-depleted suspensions. Therefore, TG suppressor activity seems to be strictly dependent upon cell interaction with ox IgG immune complexes used for TG cell isolation. Different experimental approaches failed to demonstrate, although did not exclude, that suppression in this system is mediated by soluble factors, γ-irradiation of TG cells abrogated their suppressor capacity.

    Notes: Summary Dose-response curves were obtained for trypsin X-irradiated in aqueous solution (4.6×10−6M), alone or after incubation with haematoporphyrin (Hp). When trypsin was irradiated alone (exposure doses ranging from 10 to 25 kR) the exponential dose-response curve showed a D37 equal to 17200 R. When trypsin was incubated with Hp before irradiation a D37 equal to 61900 R was obtained, with a dose reduction factor of 3.6. Therefore in our experimental conditions Hp shows a highly significant radioprotective action.

    Notes: Summary Hofbauer cells are a major cell type of the human placental villous core and they are particularly numerous at the beginning of pregnancy. In the present study we describe a method suitable to obtain HC suspensions in a highly purified form. These suspensions have been analyzed for surface markers using a battery of monoclonal antibodies. Of all the surface markers used, Hofbauer cells were only positive for 4F2, LeuM2 and LeuM3 monoclonals which mainly detect cells of the monocyte-macrophage lineage. Hofbauer cells were consistently negative for HLA-DR antigens, C3bR and T- or B-cell markers. Hofbauer cells appeared capable of phagocytosing latex beads, adhering to and spreading over plastic surface and secreting lysozyme. In contrast, they failed to originate an efficient respiratory burst in response to appropriate stimulation. Hofbauer cells were positive for ANAE with a perinuclear localization of the enzyme activity, but consistently negative for peroxidase. These observations suggest that they share a number of features with cells of the monocyte-macrophage lineage and yet have some distinctive properties.

    Notes: Summary The administration of anti-lymphocyte globulin in a patient with pure red cell aplasia completely abrogated the PHA-induced T colony forming capacity of peripheral blood mononuclear cells. The possible mechanisms of the ALG-induced suppression are discussed. It is proposed that T-colony assays may represent an useful tool for monitoring some effects of immunosuppressive drugs on T cells.

    Notes: Abstract Cell lines were established from purified large granular lymphocites (LGL) isolated from the peripheral blood of seven patients with phenotypically homogeneous LGL expansions. LGL were stimulated with phytohemagglutinin (PHA) or recombinant interleukin-2 (rIL-2) and further expandedin vitro in IL-2-containing media. The surface phenotype of LGL, as assessed by monoclonal antibody staining, was T3+ T8+ in five patients, T3− T8− in one, and T3+ T8− in another patient. The cells also expressed Leu 7, Leu 11, and/or OKM 1 markers in various proportions and were identifiable as LGL by their morphological and cytochemical features. The original surface phenotype of the unstimulated LGL was retained in the IL-2-dependent cell lines from each individual patient, i.e., T3+ T8+ cells originated T3+ T8+ cell lines and T3− T8− cells originated T3− T8− cell lines. Other markers, such as Leu 11 and OKM 1, were generally lost in culture. LGL proliferated in response to rIL-2 but did not express detectable IL-2 receptors, even after prolonged periods of culture. All cell lines from each individual patient had the same surface phenotype, and within the single lines, all of the cells expressed the same markers. Cell lines from two patients consistently displayed chromosomal abnormalities. Although different in the two patients, the abnormalities were identical in all of the lines from the same patient and detectable in most of the cells examined, suggesting a clonal origin for the abnormally expanded LGL populations. Freshly isolated LGL did not exert NK activity. However, the IL-2-dependent LGL lines acquired the ability to kill K562 target cells and to produce gamma interferon (γ-IFN). No direct correlation was observed between these two properties.
