    Notes: Summary The effect of 2-deoxy-d-glucose on growth of Schizosaccharomyces pombe 972h−, and on its cell wall composition, structure and susceptibility to enzymatic degradation has been investigated. The growth rate was not markedly affected at below 80 μg/ml of the inhibitor, but the increased frequency of appearance of aberrant forms and the reduction in the final population attained varied directly with increased inhibitor concentration. These abnormal cells show localized lesions and, at these points, extrusion of cell contents still enclosed by an inner cell wall layer which is not sensitive to osmotic shock. Cells grown normally yield “prosphaeroplasts” on treatment with snail digestive enzymes; thiol pretreatment was not necessary but degradation proceeded more rapidly when MgSO4 was used in place of sorbitol as osmotic stabilizer. Deoxyglucose-grown organisms rapidly yield true osmotically-sensitive sphaeroplasts on similar treatment. Analysis of isolated walls from mechanically disrupted cells showed marked changes in composition; an increase in the total lipid was accompanied by decreased content of both glucose and glucosamine.

    Notes: Abstract Thiobacillus tepidarius was shown to contain cytochrome(s) c with absorption maxima at 421, 522 and 552 nm in room temperature reduced minus oxidized difference spectra, present at 1.1–1.2 nmol per mg dry wt and present in both membrane and soluble fractions of the cell. The membrane-bound cytochrome c (1.75 nmol per mg membrane protein) had a midpoint potential (Em, pH 7.0) of 337 mV, while the soluble fractions appeared to contain cytochrome(s) c with Em (pH 7.0) values of about 270 and 360 mV. The organism also contained three distinct membrane-bound b-type cytochromes (totalling 0.33 nmol per mg membrane protein), each with absorption maxima in reduced minus oxidized difference spectra at about 428, 532 and 561 nm. The Em (pH 7.0) values for the three cytochromes b were 8 mV (47.8% of total), 182 mV (13.7%) and 322 mV (38.5%). No a- or d-type cytochromes were detectable spectrophotometrically in the intact organism or its membrane and soluble fractions. Evidence is presented for both CO-binding and CO-unreactive cytochromes b or o, and CO-binding cytochrome(s) c. From redox effects observed with CO it is proposed that a cytochrome c donates electrons to a cytochrome b, and that a high potential cytochrome b or o may be acting as the terminal oxidase in substrate oxidation. This may be the ‘445 nm pigment’, a photodissociable CO-binding membrane haemoprotein. Substrate oxidation was relatively insensitive to CO-inhibition, but strongly inhibited by cyanide and azide. Thiosulphate oxidation couples directly to cytochrome c reduction, but tetrathionate oxidation is linked (probably via ubiquinone Q-8) to reduction of a cytochrome b of lower potential than the cytochrome c. The nature of possible electron transport pathways in Thiobacillus tepidarius is discussed. One speculative sequence is: c− → b8 → b182 → c270 → c337 → b322/c360 → O2

    Notes: The bid for Safeway announced by Morrisons in January 2003 ended months of speculation as to the next victim in the battle for power in the UK multiple food market. The Morrisons bid provided the catalyst for other major food retailers to express their interest in the Safeway company and its stores (also joined by a number of non-food retailers). There is little doubt that this battle raises important geographical issues. First is the spatial fit of each of the bidders and the Safeway stores. The key question is who will gain most new regional market share from the purchase of the Safeway stores? The second is the implication for local spatial monopolies in different parts of the UK depending on the likely winner. This will almost certainly be investigated by the UK Competition Commission. The aim of this paper is to examine both these geographical issues and present some what-if scenarios concerning future amalgamations of store networks.

    Notes: Cytochrome c552 is the terminal component of the formate-dependent nitrite reduction pathway of Escherichia coli. In addition to four ‘typical’ haem-binding motifs, CXXCH-, characteristic of c-type cytochromes, the N-terminal region of NrfA includes a motif, CWSCK. Peptides generated by digesting the cytochrome from wild-type bacteria with cyanogen bromide followed by trypsin were analysed by on-line HPLC MS/MS in parent scanning mode. A strong signal at mass 619, corresponding to haem, was generated by fragmentation of a peptide of mass 1312 that included the sequence CWSCK. Neither this signal nor the haem-containing peptide of mass 1312 was detected in parallel experiments with cytochrome that had been purified from a transformant unable to synthesize NrfE, NrfF and NrfG: this is consistent with our previous report that NrfE and NrfG (but not NrfF) are essential for formate-dependent nitrite reduction. Redox titrations clearly revealed the presence of high and low mid-point potential redox centres. The best fit to the experimental data is for three n = 1 components with mid-point redox potentials (pH 7.0) of +45 mV (21% of the total absorbance change), −90 mV (36% of the total) and −210 mV (43% of the total). Plasmids in which the lysine codon of the cysteine–lysine motif, AAA, was changed to the histidine codon CAT (to create a fifth ‘typical’ haem c-binding motif), or to the isoleucine and leucine codons, ATT and CTT, were unable to transform a Nrf− deletion mutant to Nrf+ or to restore formate-dependent nitrite reduction to the transformants. The presence of a 50 kDa periplasmic c-type cytochrome was confirmed by staining proteins separated by SDS–PAGE for covalently bound haem, but the methyl-viologen-dependent nitrite reductase activities associated with the mutated proteins, although still detectable, were far lower than that of the native protein. The combined data establish not only that there is a haem group bound covalently to the cysteine–lysine motif of cytochrome c552 but also that one or more products of the last three genes of the nrf operon are essential for the haem ligation to this motif.
