    Notes: Summary The absolute bioavailability (f) and pharmacokinetics of transnasal butorphanol were evaluated in patients experiencing rhinitis. In an open three-way crossover study, a single 2-mg dose of butorphanol tartrate was administered by intravenous bolus injection (Treatment A), by the transnasal route (Treatment B), or by the transnasal route with pretreatment of the vasoconstrictor, oxymetazoline (Treatment C). Plasma concentrations of butorphanol were determined using a drug specific radioimmunoassay. The pharmacokinetic parameters were derived using the noncompartmental methods. Butorphanol was rapidly absorbed after transnasal administration. The mean maximum concentrations (Cmax) for the transnasal treatment with and without pretreatment of oxymetazoline were 1.61 and 3.01 ng·ml−1, respectively. The corresponding mean absorption times (MAT) were 1.34 and 0.23 h. The mean half-life values were 5.95, 6.28, and 5.77 h, for treatments A, B, and C, respectively. The resulting mean area under the plasma concentration curve (AUC) values were 11.9, 8.6, and 8.07 ng·h·ml−1 for treatments A, B, and C, respectively. The estimates for absolute bioavailability (f) of transnasal butorphanol were 69% and 72% when administered with and without oxymetazoline, respectively. The mean CLT and Vss were 121 l·h−1 and 791 l, respectively, for the intravenous treatment. The pretreatment of oxymetazoline significantly lowered the Cmax and prolonged the absorption time of butorphanol. Although the rate of absorption of transnasal butorphanol was affected by oxymetazoline, the absolute bioavailability in rhinitis patients (72%) was similar to that found with the pretreatment of oxymetazoline (69%) and those reported in healthy volunteers. Dosage regimen of transnasal butorphanol does not need modification in patients experiencing rhinitis even when they are pretreated with oxymetazoline.

    Notes: The photoluminescence (PL) at λ≤850 nm of boron-doped porous Si films anodically etched, passively etched, annealed, and reactive ion etched (RIE) under systematically varied conditions is described and discussed. As previously observed, the PL yield η of films etched from 7 to 20 Ω cm wafers in HF/H2O/C2H5OH solutions rapidly degraded during 40 mW illumination in air at 488 nm. In addition, it was totally quenched by O2 annealing or RIE but not by H2 RIE. However, the yield of films etched in HF/H2O only decreased by less than 10% following similar illumination for 7 h, and O2 annealing or RIE reduced it by 0%–50% only. The instability under illumination, the effects of oxygen annealing and RIE, Auger line shape measurements, and surface charge buildup during Auger analysis are consistent with a significantly higher density of weak Si—Si bonds in films etched in ethanol-containing solutions. These bonds are apparently broken by photoinduced oxidation, oxygen annealing, or O2 RIE, but the oxygen does not efficiently passivate the newly generated dangling bonds. While the peak emission wavelength λmax of the films etched from these wafers shifted with annealing and etching conditions, it invariably peaked in the ∼7600–8600 A(ring) range; η reached ∼1.7%. H2 RIE had no effect on η, but λmax was blueshifted. This behavior is attributed to similar breaking of weak Si—Si bonds followed by efficient passivation of the newly generated dangling bonds.The blueshift results from the higher H/Si ratio; the invariant yield is believed to be limited by the density of dangling bonds at the SiOxHy/Si interface which is apparently unaffected by the treatments. It is therefore believed that the PL is at least partially due to SiHx or SiOxHy complexes on the crystallite and column surfaces, which are similar to those found at the surface of some H-rich a-Si:H. Size effects, however, cannot be ruled out. In contrast to films etched from 7 to 20 Ω cm wafers, λmax of those prepared from 1 to 2 Ω cm substrates was consistently ∼7000 A(ring) after a brief exposure to air. While η exhibited a complex dependence on the O2 RIE power, λmax was unchanged by either O2 or H2 plasmas. The emission from heavily doped ∼0.02 Ω cm films was usually undetectable.     Notes: The effects of chronic administration of clorgyline and pargyline on rat brain monoamine metabolism have been examined. The inhibitory selectivity of these drugs towards serotonin deamina-tion (MAO type A) and phenylethylamine deamination (MAO type B) can be maintained over a 21-day period by proper selection of low doses of these drugs (0.5-1.0 mg/kg/24h). The results are consistent with MAO type A catalyzing the deamination of serotonin and norepinephrine and with MAO type B having little effect on these monoamines. Dopamine appears to be dcaminated in vivo principally by MAO type A. Clorgyline administration during a 3-week period was accompanied by persistent elevations in brain norepinephrine concentrations; serotonin levels were also increased during the first 2 weeks, but returned towards control levels by the third week of treatment. Low doses of pargyline did not increase brain monoamine concentrations, but treatment with higher doses for 3 weeks led to elevations in brain norepinephrine and 5-hydroxytryptamine; at this time significant MAO-A inhibition had developed. The changes in monoamine metabolism seen at the end of the chronic clorgyline regimen are not due to alterations in tryptophan hydroxylase activity. At this time tyrosine hydroxylase activity was also unaffected.

    Notes: Using the technique of in situ hybridization, we have attempted to identify messenger RNA for tumour necrosis factor-alpha (TNFα) in cells infiltrating allergen-induced late phase reaetion (LPR) of the skin and the nose ofatopic subjects. We have also compared the number of TNFα mRNA positive cells in bronchoalveolar lavage (BAL) from atopic asthmatics and normal controls. Twenty-four hours after local allergen challenge, 12/14 skin biopsies and 9/10 nasal biopsies had positive hybridization signals for TNFα mRNA whereas only 4/14 and 2/10 biopsies were positive in the relevant diluent controls. Compared with diluent sites significantly increased numbers of cells expressing mRNA for TNFα were observed in the LPR of skin (P 〈0.004) and nose(P 〈0.006). All BAL from asthmatics (n= 10) and from normal volunteers (n= 10) had cells showing positive hybridization signals for TNFα mRN A but these were at increased frequency in asthmatics (P〈 0.001). These results suggest that TNFα may be an important cytokine in atopic allergic inflammation.
