Notes: Summary The concentrations of organic C and total N in five different particle-size fractions were studied under different mineral and organic fertilizer regimens by examining soil samples from the 34-year-old soil-formation pot experiment Hu 3 in Rostock. The C and N concentrations were generally highest in the clay fraction and decreased in the order medium silt 〉fine silt 〉coarse silt and sand. For nearly all years and size fractions the following order was obtained for C and N concentrations under the various fertilizer regimens: Compost 〉farmyard manure 〉straw + mineral fertilizer 〉mineral fertilizer. The various particle-size fractions and fertilizer regimens differed in the development of soil organic matter levels. Consequently, characteristic redistributions were found in the proportions of C and N in the various particle-size fractions, particularly after organic fertilizer was no longer applied (years 20–34). This experimental phase was characterized by decreased organic C and increased total N concentrations, and increased proportions of C and N in the clay-size at the expense of the sand fractions.

Notes: Abstract The dominant suppressor Su(var)b 101 and the dominant enhancer En(var)c 101 were found to affect significantly white variegation in a strongly variegating line of the w m4 chromosome (w m4h ) which has been used as standard rearrangement for a genetic dissection of position-effect variegation (Reuter and Wolff, 1981). Both mutations were also shown to affect position-effect heterochromatisation in T(1;4)w m258-21 and variegation in all the rearrangements tested (white, brown, scute and bobbed variegation). These results suggest that the genes identified encode functions essential for the manifestation of gene inactivation in position-effect rearrangements. It seems also reasonable to assume that in all the rearrangements tested identical heterochromatisation processes lead to inactivation of the genes whose phenotype is variegated.

Notes: Abstract In position-effect variegation euchromatic genes are brought into the vicinity of heterochromatic sequences as a result of chromosomal rearrangements. This results in the inactivation of these genes in a proportion of cells causing a variegated phenotype. Tartof et al. (1984) have shown that the flanking heterochromatin in the w m4 variegating rearrangement in Drosophila melanogaster is homologous to the Type I inserts found in some portions of the rDNA repeats. We have studied the functional properties of these sequences using 51 revertant chromosomes, several variant lines of w m4 , strong enhancer mutations of position-effect variegation and X heterochromatin deletions. Our results suggest an array of tandemly repeated sequences showing additive effects and probably subject to magnification and reduction in number. Since only 3 of the 51 revertants isolated do not show variegation if strong enhancer mutations are introduced, only a very short sequence must be essential for the induction of white gene inactivation in w m4 . This suggests that the heterochromatic junction itself is sufficient to initiate the variegation of an adjacent gene. Parental source as well as paternal effects on the activity of these sequences have been detected. Revertant chromosomes of w m4 can be found after P-directed mutagenesis in hybrid dysgenic crosses suggesting mobile genetic elements at the breakpoints of inversion w m4 . These results are discussed with respect to the structural basis of positioneffect variegation as well as the function of certain heterochromatic sequences.

Notes: Abstract Two ornithine carbamoyltransferases (OCT 1 and OCT 2) were isolated from Pseudomonas syringae pv. phaseolicola and purified by precipitation with ammonium sulfate, heat denaturation, chromatography on DEAE-Sephadex A-50 and Sephadex G-200. Molecular weights of both enzymes: 110,000; optimal activity: pH 8.5 to 9.5 (OCT 1), pH 8.4 (OCT 2); apparent K m for ornithine: 7·10-4 (both enzymes); apparent K m for carbamoylphosphate: 7·10-4 (OCT 1), 2.8·10-3 (OCT 2). Both enzymes possess only an anabolic function. OCT 1 is highly inhibited by low concentrations of phaseolotoxin and Orn-P(O)(NH2)-NH-SO3H, OCT 2 is insensitive to both compounds. The inhibition of OCT 1 is reversible.

Notes: Abstract Lipopolysaccharides (LPS) from a number of purple nonsulfur bacteria and of phylogenetically related species were analyzed for the presence of sialic acid by gas chromatography/mass spectrometry. Species and strains of the genera Rhodobacter, Rhodopseudomonas, Rhodomicrobium, Rhodospirillum, Rhodocyclus and Rhodopila were investigated, sialic acid, however, was found only in the genus Rhodobacter. It occurs in strains of Rhodobacter capsulatus, R. sphaeroides, R. sulfidophilus and R. veldkampii. All these species belong to the α-3 subgroup of purple bacteria as defined by 16S rRNA catalogues. Approximately equimolar ratios of sialic acid and of 2-keto-3-deoxy-octonate (KDO) were found in isolated LPSs. Sodium deoxycholate gel electrophoresis of these LPS-samples also suggested a location of sialic acid in the LPS “core” region. Sialic acid was present only in those LPSs, which exhibited a “complete core region”.

Notes: A method is described for the growth from aqueous solution of Li1 - xHxIO3 single crystals. Unit-cell parameter, low-frequency resistivity and the temperature dependence of the α - β transition for several x values were determined. With an increase in x there is an increase in a0 and a decrease in C0 compared with the lattice constants of LiIO3. The maximum increase in a0 is 0.082 Å and the maximum decrease of C0 is 0.219 Å.
