    Notes: Summary Isolated skeletal muscle from healthy individuals was used to evaluate the role of phosphoinositide 3-kinase (PI 3-kinase) in insulin signalling pathways regulating mitogen activated protein kinase (MAP-kinase) and protein kinase-B and to investigate whether MAP-kinase was involved in signalling pathways regulating glucose metabolism. Insulin stimulated glycogen synthase activity ( ≈ 1.7 fold), increased 3-o-methylglucose transport into human skeletal muscle strips ( ≈ 2 fold) and stimulated phosphorylation of the p42 ERK-2 isoform of MAP-kinase. This phosphorylation of p42 ERK2 was not blocked by the PI 3-kinase inhibitors LY294002 and wortmannin although it was blocked by the MAP-kinase kinase (MEK) inhibitor PD 98059. However, PD98059 (up to 20 μmol/l) did not block insulin activation of glycogen synthase or stimulation of 3-o-methylglucose transport. Wortmannin and LY294002 did block insulin stimulation of protein kinase-B (PKB) phosphorylation and stimulation of 3-o-methylglucose transport was inhibited by wortmannin (IC50≈ 100 nmol/l). These results indicate that MAP-kinase is activated by insulin in human skeletal muscle by a PI 3-kinase independent pathway. Furthermore this activation is not necessary for insulin stimulation of glucose transport or activation of glycogen synthase in this tissue. [Diabetologia (1997) 40: 1172–1177]

    Notes: Abstract Aims/hypothesis. We have previously reported that hyperglycaemia activates glucose transport in skeletal muscle by a Ca2+-dependent pathway, which is distinct from the insulin-signalling pathway. The aim of this study was to explain the signalling mechanism by which hyperglycaemia autoregulates glucose transport in skeletal muscle. Methods. Isolated rat soleus muscle was incubated in the presence of various concentrations of glucose or 3-O-methylglucose and protein kinase C and phospholipase C inhibitors. Glucose transport activity, cell surface glucose transporter 1 and glucose transporter 4 content and protein kinase C translocation was determined. Results. High concentrations of 3-O-methylglucose led to a concentration-dependent increase in [3H]-3-O-methylglucose transport in soleus muscle. Dantrolene, an inhibitor of Ca2+ released from the sarcoplasmic reticulum, decreased the Vmax and the Km of the concentration-response curve. Protein kinase C inhibitors (H-7 and GF109203X) inhibited the stimulatory effect of high glucose concentrations on hexose transport, whereas glucose transport stimulated by insulin was unchanged. Incubation of muscle with glucose (25 mmol/l) and 3-O-methylglucose (25 mmol/l) led to a three fold gain in protein kinase Cβ 2 in the total membrane fraction, whereas membrane content of protein kinase Cα, β 1, δ, ɛ and ϑ were unchanged. A short-term increase in the extracellular glucose concentration did not change cell surface recruitment of glucose transporter 1 or glucose transporter 4, as assessed by exofacial photolabelling with [3H]-ATB-BMPA bis-mannose. Conclusion/interpretation. Protein kinase Cβ 2 is involved in a glucose-sensitive, Ca2+-dependent signalling pathway, which is possibly involved in the regulation of glucose transport in skeletal muscle. This glucose-dependent increase in 3-0-methylglucose transport is independent of glucose transporter 4 and glucose transporter 1 translocation to the plasma membrane and may involve modifications of cell surface glucose transporter activity.     Notes: Abstract Photoluminescence (PL) experiments have been carried out in glasses from the ZnO-CdO-SiO2 system. The glasses exhibit broad bands in the PL spectra centered between 540 and 560 nm for emissions and at about 250 nm for excitations. Decay time curves for the different glass samples appear, within experimental errors, quite similar. The curves are not a simple exponential and have been discomposed as a sum of three exponentials with lifetime components of about 0.025, 0.1, and 0.52 ms. The results seem to indicate that luminescence is dominated by short ordering, nuclei and/or crystalline centers.

    Notes: The effect of two temperature regimes (daytime, 29 ± 2°C, night-time, 24 ± 3°C; and daytime, 23 ± 1°C, night-time, 18 ± 2°C) on the symptoms caused by tomato spotted wilt virus (TSWV), and the accumulation of TSWV virions, was compared in Datura stramonium, Nicotiana tabacum cv. White Burley and Physalis ixocarpa. Tobacco plants were more severely affected by TSWV at the high temperature regime, but the incidence (percent of plants with symptoms) was 100% for both regimes. In P. ixocarpa and D. stramonium the higher temperature caused an increase in both incidence and rate of development of symptoms. At high temperature, all three species showed both local and systemic symptoms; however, at low temperature only P. ixocarpa consistently developed systemic symptoms. In general, virus accumulation in the inoculated leaves (presumably the combined effect of virus replication and local movement) of all plants was higher at the lower temperature. Long distance movement in tobacco, leading to virion accumulation in other plant organs, was favoured by high temperature; but there was relatively little effect in P. ixocarpa and D. stramonium.

    Notes: Objective:  The aim of this study was to evaluate the influence of Emdogain® (EMD) on cultured gingival fibroblasts, periodontal ligament fibroblasts and dermal fibroblasts, using an in vitro model of wound healing.Background:  Enamel matrix derivative has been demonstrated to promote periodontal regeneration. However, the precise mechanisms by which this agent acts are still unclear.Methods:  The effect of EMD on proliferation of the cells was studied using subconfluent cultures of gingival fibroblasts and periodontal ligament fibroblasts. The cells were made quiescent overnight and then stimulated with various concentrations of EMD (10, 50, 100 and 150 μg/ml) for 24 h. Negative and positive controls were cells cultured in media containing 0.2% and 10% fetal calf serum (FCS). The DNA synthesis was measured by the cellular uptake of [3H]thymidine. For in vitro wounding the cells were cultured, wounded and stimulated with 0.2% FCS, 10% FCS and EMD at a concentration of 20 μg/ml. The percentage of wound fill after treatment was measured after d 1, 4, 6, 12 and 16. The proliferation of cells was also calculated by the extent of incorporation of crystal violet.Results:  The results demonstrated that cells in vitro fill an empty space by a combination of proliferation and cell migration. The most rapid closure of a wound area occurred where both proliferation and migration can occur as was seen when wounded cultures were maintained in 10% FCS or at a concentration of 20 μg/ml EMD which promoted proliferation.Conclusions:  Therefore, EMD appears to exert an influence on cells that is compatible with improved wound healing.

    Notes: Abstract The recycling of toxic goethite waste, originated in the hydrometallurgy of zinc ores, in glass-ceramic matrices has been studied. Oxide compositions suitable to form glasses were prepared by mixing the goethite waste with granite scraps and glass cullet, yielding the following oxide composition (wt%): SiO2, 44.6; Al2O3, 3.3; Fe2O3, 25.5; MgO, 1.6; CaO, 4.5; Na2O, 5.9; PbO, 3.1; ZnO, 6.5; K2O, 1.0; TiO2, 2.0; other 2.0. By proper addition of carbon powder, the initial Fe3+/Fe2+ ratio (12) of glasses melted in air at 1450°C was approximated to the stoichiometric value of magnetite (2) to obtain high nucleation and crystallization rates. The heat treatment of iron supersaturated goethite glasses above 630°C led to the formation of magnetite nuclei with a high tendency to grow and coalesce with time. The crystallization of pyroxene, occurring on the magnetite crystals above 800°C, was found to be influenced by the nucleation period, so that the highest crystalline volume fraction, Vf (0.80–0.85), was obtained for 90–120 min nucleation time at 670°C and 120 min crystallization at 860°C.
