    Notes: The histopathology of biliary hyperplasia and renal calcinosis, associated with infections of two parasites, Myxidium incurvatum and Rhabdospora thelohani, is described in turbot, Scophthalmus maximus (L.), from Scottish marine fish farms and wild populations. Liver and kidney lesions were divided into four, and parasite infections into five grades of severity. A few of the fish examined were not parasitized, but every one showed some evidence of pathological change, even the apparently normal individuals from all populations. Although there is a positive association between the parasitic and pathological data, parasitism is not considered to be the principal cause of either the hepatic or renal lesion; the evidence suggests rather a secondary opportunist role for the parasites. The general severity of the condition in captive stocks suggests that the cause may lie in the fundamentals of present techniques of turbot husbandry, but no specific agent has been identified. A case is put forward for considering Rhabdospora to be a parasite rather than a specialized host cell.

    Notes: The lympho-reticular tissues in the plaice were investigated for their phagocytic properties on colloidal carbon after its intraperitoneal injection. Fish were killed at intervals ranging from 10 min to 25 days after injection. Although peritoneal macrophages constituted a large population of phagocytic cells, most of the carbon apparently gained access to the circulation as free particles and phagocytosis was performed predominantly by the ellipsoids of the spleen, the network of reticulo-endothelial (RE) cells throughout the haemopoietic tissue of the kidney, and by the RE cells occupying intermuscular spaces in the atrium of the heart. The cardiac macrophages rapidly emigrated from the organ while the carbon containing macrophages in the kidney and spleen formed aggregates in the lymphoid areas, either within or outwith pre-existing aggregates of melano-macrophages.The possible significance of phagocyte aggregations, including melano-macrophages, in association with lymphoid elements in the kidney and spleen is discussed in the context of immune mechanisms.

    Notes: The rate of healing of a surgical wound was studied in two teleost fish, one with a tropical, and the other with a temperate temperature range. Comparisons were made of both the rate and qualitative nature of wound healing within and between species at temperatures of 30,23,10 and 5° C. The rate of wound healing was found to be proportional to temperature and temperature stress had little effect on healing rates. The findings were related to reported rates of wound healing in man. In general the wounds studied healed at a rate comparable to those reported for the healing of superficial skin wounds in man and other mammals despite the fact that the fish wounds were not merely superficial but involved integument and muscle.

The quantitative dietary ascorbic acid requirements of fingerling Nile tilapia, Oreochromis niloticus L., were assessed by feeding seven diets containing graded supplements of this vitamin (0, 50, 75, 100, 125, 300 and 400mg/100g dry diet) to triplicate groups of fish for 12 weeks. Fish fed the diet devoid of ascorbic acid exhibited significantly (P 〈 0·01) poorer growth and higher condition factor. Food conversion and protein efficiency ratios were improved with increasing dietary ascorbic acid level up to 125mg/100g of dry diet. Protein utilization, haematocrit, haemoglobin level and tissue ascorbate concentrations were depressed in fish fed the ascorbic-acid-free diet. Tissue ascorbate concentrations, especially for liver, were highly correlated with dietary ascorbic acid level. Fish fed the ascorbic-acid-free diet exhibited deficiency signs including erratic and convulsive swimming, anorexia, lethargy, caudal fin erosion, skin haemorrhages and mortality. Based on the nutritional and pathological parameters investigated, the recommended dietary inclusion level is 125mg/100g dry diet, which is equivalent to a net requirement (after processing and storage) of 42mg/100g diet.

Ascorbic acid depleted Oreochromis niloticus fingerlings were fed on each of three diets providing nil, normal and luxus levels of the vitamin, after small surgical incisions had been made in the dursoluteral musculature. Fish from each group were sampled regularly over 16 days and histopathological evaluation of the lesion area carried out. The tissue ascorbate levels were measured in each group during the study. The results showed that the epithelial element of the healing process developed irrespective of the vitamin level but although fibroblast activity was marked in all three groups, coltagenization was very much slower in the deficient group, and in these the lesion was not mature, even at the termination of the experiment.

L-gulonolactone oxidase activity was assessed in kidney and liver tissues of 14 teleosts by use of a histochemical technique for qualitative identification of the presence of enzyme activity and a biochemical technique for quantification of activity of the enzyme. Nine tilapias, two cyprinids and three salmonids were investigated; enzyme activity was only detected in liver and kidney tissues of common carp, Cyprinus carpio L., and kidney tissue of Oreochromis spilurus (Günther) and O. aureus (Steindachner).

    Notes: Hatchery reared Atlantic salmon (Salmo salar L.) parr were tagged with the standard I.C.E.S./I.C.N.A.F. salmon tag at water temperatures of 15°C and 6°C. They were then subjected to swimming trials in a tunnel respirometer. Following this treatment the 15°C fish developed necrotic infarcts around the tag insertions. Provided they did not become infected these lesions healed, but if the fish were retested 9 days later, the wound enlarged, became extremely inflamed, and the fish died soon after. This effect was not seen in 6°C fish.

    Notes: Starvation resulted in considerably increased deposition of melano-macrophages within the spleen and kidney of Pleuronectes plalessa, Salmo gairdneri, Xiphophorus helleri, and Tilapia zillii also within the liver and spleen of Scyliorhinus canicula. These results suggest that catabolic tissue breakdown is a major factor contributing to the formation of the pigments within melano-macrophages.
