    Notes: Abstract Various murine tumor lines were screened by FACS analysis for the surface antigens ThB and Ly-6.2. Positive cell lines were used for immunoprecipitation studies. A monoclonal ThB-specific antibody immunoprecipitated a unique acidic protein of approximately 16 000 daltons from several positive tumors and from concanavalin A (Con-A) and LPS activated splenic lymphocytes. Monoclonal Ly-6.2-specific antibody was used to immunoprecipitate a 33 500 dalton protein that was shown to exist in four similarly sized forms with different basic charges. In the course of these studies, the apparent molecular weight of the surface antigen T 30, immunoprecipitated with a monoclonal T 30-specific antibody from the cell line EL4, was found to be approximately 25 000 daltons.

    Notes: Abstract Ly-5 (CD45) glycoproteins of the mouse, expressed by all or most hematopoietic cell lineages and specified by a single Ly-5 gene, range in size from isoform T200 of T cells (the smallest), in which exons 4, 5, and 6 are not represented, to isoform B220 of B cells (the largest), in which all three of these optional exons are represented. The main purpose of the present study, utilizing the polymerase chain reaction (PCR), was to ascertain whether known isoforms of intermediate size are generated by single or dual usage of optional exons 4, 5, and 6. Transcripts representing all eight isoforms predictable from varied use of three exons were observed among a diverse panel of nine B-cell tumors in culture, but there was no evident concordance with known contrasting differential features that distinguish members of the B-cell tumor panel. No two B tumors exhibited the same variety of transcripts and the relative quantities of transcripts expressed varied greatly from tumor to tumor. Cloning of B-cell tumors did not alter their distinctive transcript patterns. Separation methods (sodium dodecyl sulfate polyacrylamide gel electrophoresis; SDS-PAGE) did not suffice to segregate all corresponding expressed isoforms but did establish that transcripts representing usage of a single optional exon and of two optional exons were actually translated, which supports a provisional inference that all eight isoforms exist. The considerable diversity of B-cell transcript phenotypes was not seen among seven T-cell leukemias, two cytolytic T-cell lines, and three Th1 helper T-cell lines, all of which displayed a uniform phenotype comprising major expression of the T200 transcript (no optional exon) and minor expression of a transcript employing exon 5. However, a panel of five cloned Th2 T-cell lines, which represents a second and functionality different branch of the helper/inducer T-cell category, exhibited a characteristic transcript pattern which distinguished them from a panel of three Th1 T-cell lines. The major transcript in the Th2 lines was also T200, but the Th2 lines showed higher representation of transcript containing optional exons. A single Th2 clone expressed an unusual transcript suggesting a potential isoform not compounded simply by varied inclusion of the three identified optional exons. After activation of the helper T-cell lines with concanavalin A (Con A), expression of transcript containing optional exons appeared to decrease.

    Notes: Abstract Candida parapsilosis grows oxidatively under aerobic conditions and fermentatively in micro-aerobic (0.2 μM oxygen) continuous culture. The amount of unsaturated fatty acids and sterol in cells and mitochondria, and the aerobic cytochromes are decreased in micro-aerobic cultures. In aerobic cells mitochondrial protein synthesis accounts for the formation of more than one-third of the proteins of the organelles; in micro-aerobic cultures, this declines to 5%. At the same time, minor but significant differences in the nature of the protein products formed by the mitochondrial system are evident in micro-aerobic and aerobic cells.

    Notes: Abstract Saccharomyces cerevisiae andCandida parapsilosis were grown aerobically and micro-aerobically in continuous culture and the energetic efficiency (Y ATP) and the maintenance energy requirements compared. The bioenergetic parameters were determined using a gas-balance technique, and from cell yield decrements at low dilution rates. The data show that for aerobic cultures, cell yield related to ATP generation (Y ATP) for the two yeasts is similar and close to the value reported originally by Bauchop and Elsden (1960). However, the efficiency of substrate utilization is greater in the case ofC. parapsilosis since the apparent P/O value under aerobic conditions is 1.8 as compared to about 1 forS. cerevisiae. The maintenance energy coefficients based on ATP requirement for aerobic cultures of both yeast were greater than the corresponding values for micro-aerobic cultures; similarly cultures of a petite mutant ofS. cerevisiae had a lower maintenance energy requirement than comparable aerobic wild-type cultures. It is suggested that these differences may reflect the energy requirement for the upkeep of a greater degree of functional complexity in aerobic, wild-type cells, as compared with micro-aerobic or respiratory deficient cells.

    Notes: Abstract The independent effects of oxygen tension and growth rate on bacteriorhodopsin synthesis in Halobacterium halobium have been studied in chemostat cultures. Bacteriorhodopsin synthesis occurs only at low growth rates and is stimulated by low oxygen tension. Fast growth rates override the stimulatory effects of oxygen tension, with the result that bacteriorhodopsin can scarcely be detected. Illumination of cultures maintained at low growth rate and low oxygen tension significantly increases the steady state cell yield. This finding suggests that under these conditions the purple membrane proton pump is coupled to energy transduction.

    Notes: Abstract A newborn infant was diagnosed as having renal vein thrombosis due to heterozygous protein C deficiency and no other predisposing factor for thrombosis. He responded adequately to treatment with streptokinase, fresh frozen plasma and subsequent anticoagulation with heparin and warfarin (Coumadin). Four years from diagnosis he remains well with no recurrence of thrombosis and with normal renal function.

    Notes: Abstract A male Caucasian infant presented at 6 weeks of age with failure to thrive, diarrhoea, macrocytic anaemia, and decreased IgG. He had normal serum B12 and folate levels. Serum cobalamin binding capacity showed no detectable transcobalamin II. Both parents showed levels consistent with a heterozygous state. The literature is extensively reviewed, and the importance of early diagnosis to prevent neurological dysfunction is stressed.

    Notes: Summary This study examines and compares the hemodynamic and sympathoadrenal response to bicycle exercise in hypertensive subjects during two weeks' treatment with a cardio-selective (metoprolol) and nonselective (propranolol) beta-blocker. The increase in plasma norepinephrine and epinephrine concentration following exercise was augmented to a similar degree with each beta-blocker. Pre-exercise blood pressure and heart rate were similar for the two drugs. However immediately after exercise and particularly after resting for 20 min post exercise, diastolic blood pressure was lower during metoprolol treatment. Systolic blood pressure was also lower 20 min post exercise during metoprolol treatment. These observations indicate that cardio-selective beta-blockers offer advantages in blood pressure control during exercise through intact vascular β2-adrenoceptors opposing sympathetically mediated vasoconstriction.
