    Notizen: Using cloud point determinations, the phase boundaries (binodals) for metastable liquid–liquid (L–L) separation in supersaturated hen egg white lysozyme solutions with 3%, 5%, and 7% (w/v) NaCl at pH=4.5 and protein concentrations c between 40 and 400 mg/ml were determined. The critical temperature for the binodal increased approximately linearly with salt concentration. The coexisting liquid phases both remained supersaturated but differed widely in protein concentration. No salt repartitioning was observed between the initial and the two separated liquid phases. After the L–L separation, due to the presence of the high protein concentration phase, crystallization occurred much more rapidly than in the initial solution. At high initial protein concentrations, a metastable gel phase formed at temperatures above the liquid binodal. Both crystal nucleation and gel formation were accelerated in samples that had been cycled through the binodal. Solutions in the gel and L–L regions yielded various types of precipitates. Based on theoretical considerations, previous observations with other proteins, and our experimental results with lysozyme, a generic phase diagram for globular proteins is put forth. A limited region in the (T,c) plane favorable for the growth of protein single crystals is delineated. © 1997 American Institute of Physics.

    Notizen: Abstract: The effects of both (–)- and (+)-nicotine isomers were examined on in vitro uptake and release of [3H]dopamine in rat striatum. Both isomers inhibited uptake of [3H]dopamine in chopped tissue at concentrations well below those necessary for promoting release of preloaded [3H]dopamine. (–)-Nicotine was more potent than (+)-nicotine both at inhibiting uptake and at promoting release. Unlike other dopamine uptake inhibitors, however, nicotine inhibited only 50% of the total uptake. In the presence of 1 nM nicotine, the residual [3H]dopamine uptake was less sensitive to inhibition by cocaine than uptake in the absence of nicotine. Nicotine did not compete against the binding of [3H]GBR 12935, a selective dopamine uptake inhibitor. The nicotinic receptor agonists carbachol and 1,1-dimethyl-4-phenylpiperazinium iodide also inhibited uptake, whereas the nicotinic antagonists chlorisondamine and mecamylamine blocked nicotine's effect. Thus, the effect of nicotine on dopamine uptake appears to be mediated by a receptor similar to the nicotinic acetylcholine receptor. These receptors do not seem to be on the terminals that are accumulating dopamine, however, since tetrodotoxin prevented the effect of nicotine on [3H]dopamine uptake and nicotine had no effect on uptake in a synaptosomal preparation.

    Notizen: Several empirical studies have estimated the value of agricultural land as open space to local residents. An important goup of individuals that may be affected by the loss of agricultural land are visitors to a region. The value of ranchland to tourists visiting a resort town in the Rocky Mountains is estimated through a travel cost model that combines information on observed behavior data from actual trips with contingent behavior data on intended current visitation if the resource were converted to urban and resort uses. The value of ranch open space to tourists is the gain or loss in consumer surplus derived from a visit to the study area attributable to the resource. A random effects Poisson regression model is estimated because of the panel nature of the data, accounting for the correlation of the multiple responses from heterogeneous individuals. Twenty-five percent of the sample would reduce visitation and 23 percent of the sample would increase visitation if ranch open space were converted to urban and resort uses. The overall effect of converting ranch open space to resort and urban uses is no net change in average consumer surplus per trip for summer tourists in general.

    Notizen: A novel version of the capillary method for viscosity measurements of liquids is presented. Viscosity data can be deduced in a straightforward way from mass transfer data obtained by differential weighing during the gravity-induced flow of the liquid between two cylindrical chambers. Tests of this technique with water, carbon tetrachloride, and ethanol suggest that this arrangement provides an accuracy of about ±1%. The technique facilitates operation under sealed, isothermal conditions and, thus can readily be applied to reactive and/or high vapor pressure liquids.

This cross talk has been investigated in subwavelength-periodically patterned thin-film structures using a reflection-mode near-field optical microscope.     Notizen: Theoretical and experimental results for using the photothermal deflection technique to measure vapor species concentration, while minimizing the disturbance of the transport (material) parameters due to vapor heating, are developed and described. In contrast to common practice, the above constraints require using a pump-beam duty cycle of less than 50%. The theoretical description of the shortened heating time is based on a step-function formulation of the pumping cycle. The results are obtained as closed-form solutions of the energy equation for many chopping cycles until steady state is reached, by use of a Green's-function method. The Euler formulation of the Fermat principle is used to calculate the deflection angle. The equations are expanded to include the effects of vapor velocity on both the temperature and temperature gradient profiles. The effects of finite (unfocused) pump and probe beams and thermal (Soret) diffusion are also accounted for. Excellent agreement between theory and experiment is obtained.

    Notizen: We demonstrate a novel concept for a mode selection mechanism in surface emitting laser diodes. This concept is based on a strong coupling process between the laser mode and a surface mode on top of the laser diode. The mode selection mechanism is the result of a strong feedback from the surface mode into the laser diode. Nearly single-modelike emission spectra are achieved from surface emitting GaAs/AlGaAs laser diodes and qualitatively explained by a model based on ray optics. The main advantage of this type of laser diode is its simple fabrication and the possibility of adjusting the desired wavelength independent of the growth process by external technological parameters. © 1995 American Institute of Physics.

    Notizen: It is traditionally assumed that the pair energies of atoms on the surface are site independent and equal to the pair energy in the bulk of the crystal (constant bond energy approach). We have assumed that the internal energy of an atom, ψ, depends nonlinearly on the number of nearest neighbors n in the form ψ(n)=−(an+bn2), where a and b are material constants that can be evaluated from experimentally determined heats of sublimation and energies of bulk vacancy formation. This simple approximation of the actual many-body interactions accommodates site-dependent pair energies. Based on this phenomenological "variable bond model'' and the Bragg–Williams approximation, occupation fractions of an equilibrium surface are obtained in two-level and three-level models. These provide a criterion of surface roughness for specific orientations. Application of the criterion to copper, lead, and zinc yields good agreement with experimental observations of anisotropic melting, while the traditional constant bond approach fails. It is shown that in addition to these systems many other metals possess negative b values, i.e., exhibit "bond strengthening'' on surfaces. Similar conditions are shown to exist in numerous ionic, covalent, and van der Waals solids. Hence, one must expect that constant bond models underestimate the surface roughness for a large group of materials.
