    Notes: Abstract The conversion of serine and tetrahydrofolate to glycine and 5,10 methylene tetrahydrofolate by serine hydroxymethyltransferase (SHMT, EC 2.1.2.1) is the major route for the provision of one-carbon units for biosynthetic reactions. SHMT cDNAs have been cloned from both rabbit and man, and a human mitochondrial SHMT gene sequence has recently been reported. We have isolated phage clones containing human genomic sequences homologous to cytosolic SHMT and have found these to contain a processed pseudogene (SHMT-psl) with a 90% identity to cloned SHMT cDNAs. SHMT ps1 contains 2335 nt that are homologous to SHMT but it has an additional leader sequence of 203 nt of unknown origin. The complete SHMT-ps.1 sequence of 2538 nt is bounded by two 16 nt direct repeats that are characteristic of retroelement insertion sites. Two phage clones containing SHMT-ps1 have been mapped by fluorescence in situ hybridisation to 1p32.3–33. ln addition, an SHMT cDNA clone hybridized to the same region and to 17p11.2–12. The latter is consistent with a previous localisation of the gene for cytosolic SHMT.

    Notes: Service industries, such as public bus transport, are time-bound, which makes it impossible to inventory their service output. The potential revenue from an empty seat on a bus is lost for good once the service run is complete. Conversely, when demand for a seat on a service run exceeds supply, the revenue is also lost. As public bus transport has a high fixed to variable cost ratio, these demand and supply imbalances have a significant impact on cost recovery performance. Addresses a number of factors that influence the cost recovery performance of public bus transport using data from one of Australia's largest operators. It considers the shortcomings of current fare price structures and how these may be changed to reflect operational cost drivers in a way that improves cost recovery performance. The various non-monetary costs passengers incur when purchasing and using public transport are also considered along with methods of reducing these to increase the revenue-generating performance of operators' fixed capacity.

    Notes: As an enzyme of the thymidylate synthase cycle, serine hydroxymethyltransferase (SHMT) has a key role in nucleotide biosynthesis. Elevated activities of SHMT have been correlated with the increased demand for nucleotide biosynthesis in tumors of human and rodent origin, making this enzyme a novel target for cancer chemotherapy. Here the purification and crystallization of recombinant human cytosolic SHMT are reported. Crystals belong to space group P6222 or P6422 with cell parameters a = b = 155.0, c = 235.5 Å and diffract to at least 3.0 Å resolution.

    Notes: Before the difaqane, warfare among the Sotho was usually little more than cattle-raiding. Some attacks were combined operations executed by all the fighting men of a chiefdom, but most were the exploits of a few adventurous individuals. The raiders would each be armed with a bunch of long spears, a knobkerrie and a light oxhide shield, and they would usually approach the enemy's cattle along river beds and through mountain kloofs, relying partly on surprise to achieve their ends. Occasionally the men who were guarding the herds would have prior warning of the attack, in which case they would be specially reinforced and would offer a spirited resistance, but more often they would be taken unawares, and they would then beat a hasty retreat and sound the alarm in the village: all the able-bodied men would thereupon join together in pursuit of the attackers in the hope of recovering their stock as it was being driven away. When the warriors of two chiefdoms clashed, they generally conducted their fighting at a considerable distance from each other, for their spears were more suitable for throwing than for stabbing, and their small shields were not designed to be impenetrable barriers in close conflict but to deflect missiles. If the two groups did come to grips with each other, the spears' bamboo handles could be broken and they could then be used for stabbing, but the most favoured weapon in this situation was the knobkerrie. Desperate battles, however, were rare, and in most of the Sotho's skirmishes their casualties were light.

    Notes: Abstract Performance aspects of a Lagrangian relaxation based heuristic for solving large 0-1 integer linear programs are discussed. In particular, we look at its application to airline and railway crew scheduling problems. We present a scalable parallelization of the original algorithm used in production at Carmen Systems AB, Göteborg, Sweden, based on distributing the variables. A lazy variant of this approach which decouples communication and computation is even useful on networks of workstations. Furthermore, we develop a new sequential active set strategy which requires less work and is better adapted to the memory hierarchy properties of modern RISC processors. This algorithm is also suited for parallelization on a moderate number of networked workstations.
