    Notes: Abstract. Molecular analyses have been used recently to refine our knowledge of phylogenetic relationships within the ciliated protozoa (phylum Ciliophora). A current Hennigian phylogeny of the orders in the class Colpodea, based on light and electron microscopic analyses, makes three important assumptions with regard to apomorphic character states, namely, (1) that the kreyellid silver line evolved early in colpodean phylogeny, separating bryometopids, such as Bryometopus, from all other colpodeans; (2) that the macro–micronuclear complex is an autapomorphy of the cyrtolophosidids, such as Platyophrya; and (3) that merotelokinetal stomatogenesis is an apomorphic character of colpodids, such as Colpoda, Bresslaua, and Pseudoplatyophrya. These predictions of relationships within the class Colpodea were investigated by determining the complete small subunit rRNA gene sequences for the colpodid Bresslaua vorax, the grossglockneriid Pseudoplatyophrya nana, and the cyrtolophosidid Platyophrya vorax and a partial sequence for the bryometopid Bryometopus sphagni. These sequences were combined with the previously published complete SSrRNA sequences for the colpodid Colpoda inflata and the bursariomorphid Bursaria truncatella. The affiliations were assessed using both distance matrix and maximum-parsimony analyses. The tree topologies for the class Colpodea were identical in all analyses, with bootstrap support for bifurcations always exceeding 60%. The results suggest the following. (1) Since the clade including Bryometopus and its sister taxon, Bursaria, is never basal, the kreyellid silver-line system evolved later in colpodean phylogeny and does not separate bryometopids from all other colpodeans. (2) Since Platyophrya is always the sister taxon to the other five genera, there is a fundamental phylogenetic significance for its macro–micronuclear complex. (3) Since the colpodids, Colpoda, Bresslaua, and Pseudoplatyophrya, always group in one clade, merotelokinetal stomatogenesis appears to be a derived character state.

    Notes: Abstract. The histones H4 are known as highly conserved proteins. However, in ciliates a high degree of variation was found compared both to other eukaryotes and between the ciliate species. To date, only H4 histones of species belonging to two distantly related classes have been investigated. In order to obtain more detailed information on histone H4 variation in ciliates we undertook a comprehensive sequence analysis of PCR-amplified internal H4 fragments from 12 species belonging to seven out of the nine currently recognized ciliate classes. In addition, we used PCR primers to amplify longer fragments of H3 and H4 genes including the intergenic region. The encoded amino acid sequences reveal a high number of differences when compared with those of other eukaryotes and the ciliate species investigated. Furthermore, in some species H4 gene variants were detected, which result in amino acid differences. The greatest number of substitutions and insertions found was in the amino terminal region of the H4 histones. However, all sequences possess a conserved region corresponding to those of all other eukaryotic H4 histones. The histone gene variations were used to reconstruct phylogenetic relationships. The tree from our data matches perfectly with the ribosomal RNA data: The heterotrichs, which were considered as a late branching lineage, diverge at the base of the ciliate tree and groups formerly thought to represent ancestral lineages now appear as highly derived ciliates.

    Notes: A new species, Pleurotricha indica n. sp., is described, characterized by an average size of 220 × 119 μm, a firm and inflexible body, six rows of dorsal kineties, one left and two right rows of marginal cirri, and an “Oxytricha-like” pattern of ventral cirri. The parts of the macronucleus are variable in shape and number. The comparison with other members of the genus shows that P. indica differs from other congeneric species in the combination of these characters.

    Notes: The small subunit rRNA gene sequences of the karyorelictean ciliates, Loxodes striatus and Protocruzia sp., and the heterotrichian ciliates, Climacostomum virens and Eufolliculina uhligi, were used to test the evolution of nuclear dualism in the Phylum Ciliophora. Phylogenies derived using a least squares distance method, neighbour joining, and maximum parsimony demonstrate that the karyorelictean ciliates sensu Small and Lynn, 1985 do not form a monophyletic group. However, Loxodes and the heterotrich ciliates form the first branch in the ciliate lineage, and Protocruzia branches, in distance methods, basal to the spirotrich lineage. It is proposed that Protocruzia be removed from the Class Karyorelictea, and placed in closer taxonomic association with the spirotrich lineage. The distribution of nuclear division types along the phylogenetic tree is consistent with the notion that macronuclei incapable of division represent a derived rather than a primitive or “karyorelictid” character trait.

    Notes: The complete small subunit rRNA gene sequences of the heterotrich Blepharisma americanum and the colpodid Colpoda inflata were determined to be 1719 and 1786 nucleotides respectively. the phylogeny produced by comparisons with other ciliates indicated that C. inflata is allied more closely with the nassophoreans and oligohymenophoreans than the spirotrichs. This is consistent with the placement of the colpodids in the Class Copodea. Blepharisma americanum was not grouped with the hypotrichs but instead was placed as the earliest branching ciliate. the distinct separation of B. americanum supports the elevation to class status given the heterotrichs based on morphological characters.

    Notes: The morphology and the morphogenesis of the freshwater hypotrich ciliate Onychodromus quadricornutus n. sp. have been investigated using living organisms, protargol impregnation, and scanning electron microscopy. Some preliminary and supplementary results about the morphogenesis of O. grandis and Laurentiella acuminata are included. The new species is unique among all described hypotrichs in having four dorsal horns, whose function is unknown. In addition, O. quadricornutus is probably the most voluminous hypotrich ciliate known (2 times 10-6-5 times 106μm3). Its morphogenetic pattern resembles the oxytrichids O. grandis and L. acuminata. The strongest apomorphic character, which unites these three species, is probably the multiple fragmentation of the dorsal primordia during morphogenesis. This fragmentation causes the characteristic high number and more or less irregular distribution of the dorsal kineties in the non-dividing individuals.

    Notes: The two closely related hypotrichous ciliate species, Stylonychia lemnae and S. mytilus, have been compared with respect to their isoenzyme patterns, their macronuclear DNA banding patterns, and micronuclear DNA banding patterns after restriction enzyme digestion. Since macronuclear DNA contains mainly protein-coding sequences and since the micronuclear DNA patterns represent mainly the repetitive fraction of the genome, these results, together with the isoenzyme patterns, reflect differences on three different levels of molecular evolution between morphologically very similar species. Each of the three methods allows an unequivocal identification of each of the two species. Intraspecific variation seems to be greatest among the repetitive sequences of the micronuclear genomes. By using the isoenzyme data and the formula of Nei (19) the genetic distance between the two species is calculated and compared with the results from other protozoa and different Drosophila species. Despite their morphological similarity, the two species show a considerable amount of evolutionary divergence on the three molecular levels which have been investigated.

    Notes: All Stylonychia mytilus-like ciliates which were collected and sent to us during the last 20 years belonged either to S. mytilus or to a new species, S. lemnae, which is described here. The only morphological differences are the shape and the size. Stylonychia mytilus that have been starved for one day average 300 μm in length, and S. lemnae starved for a day are 230 μm long. The occurrence of mating types is described.

    Notes: . The Testaceafilosia includes amoebae with filopodia and with a proteinaceous, agglutinated or siliceous test. To explore the deeper phylogeny of this group, we sequenced the small subunit ribosomal RNA coding region of 13 species, including the first sequence of an amoeba with an agglutinated test, Pseudodifflugia sp. Phylogenetic analyses using maximum parsimony and maximum likelihood methods as well as neighbor joining method yielded the following results: the order Euglyphida forms a monophyletic lineage with the sarcomonads as sister group. The next related taxa are the Chlorarachnea and the unidentified filose strain N-Por. In agreement with the previous studies the Phytomyxea branch off at the base of this lineage. The Monadofilosa (Testaceafilosia and Sarcomonadea) appear monophyletic. The Testaceafilosia are polyphyletic, because Pseudodifflugia sp. is positioned as the sister taxon to the sarcomonads. Within the order Euglyphida Paulinella branches off first, together with Cyphoderia followed by Tracheleuglypha. In maximum likelihood and neighbor joining analyses, the genus Euglypha is monophyletic. The branching pattern within the order Euglyphida reflects the evolution of shell morphology from simple to complex built test.

    Notes: Abstract Ten species of orchid plants belonging to the generaOrchis (7),Dactylorhiza (2), andGymnadenia (1) were analyzed by enzyme electrophoresis. Each species can be identified by a combination of enzyme bands different from those of all other species examined. The electrophoretic data were used for the construction of phenetic and phylogenetic trees with the help of computer programs. The trees were almost identical regardless which method was used. Our results differ considerably from a classification based on morphological evidence. The electrophoretic data indicate that the genusOrchis is not a monophyletic group.
