    Notes: Abstract. The chloride conductance of inner medullary collecting duct cells (mIMCD-3 cell line) has been investigated using the whole cell configuration of the patch clamp technique. Seventy-seven percent of cells were chloride selective when measured with a NaCl-rich bathing solution and a TEACl-rich pipette solution. Seventy-five percent of chloride-selective cells (90/144) had whole cell currents which exhibited an outwardly-rectifying (OR) current-voltage (I/V) relationship, while the remaining cells exhibited a linear (L) I/V relationship. The properties of the OR and L chloride currents were distinct. OR currents (mean current densities at ±60 mV of 66 ± 5 pA/pF and 44 ± 3 pA/pF), were time- and voltage-independent with an anion selectivity (from calculated permeability ratios) of SCN− (2.3), NO− 3 (1.8), ClO− 4 (1.7), Br− (1.7), I− (1.6), Cl− (1.0), HCO− 3 (0.5), gluconate− (0.2). Bath additions of NPPB, flufenamate, glibenclamide (all 100 μm) and DIDS (500 μm) produced varying degrees of block of OR currents with NPPB being the most potent (IC50 of approximately 50 μm) while DIDS was the least effective. Linear chloride currents had similar current densities to the OR chloride currents and were also time- and voltage-independent. The anion selectivity sequence was SCN− (2.5), NO− 3 (1.9), Br− (1.4), I− (1.1), Cl− (1.0), ClO− 4 (0.5), HCO− 3 (0.5), gluconate− (0.3). In contrast to the OR conductance, glibenclamide was the most potent and DIDS the least potent blocker of L currents. An IC50 of 〉100 μm was observed for NPPB block. Neither OR of L chloride currents were affected by acutely or chronically increased intracellular cAMP and were not affected when intracellular Ca2+ levels were increased or decreased. The molecular identity and physiological role of OR and linear currents in mIMCD-3 cells are discussed.

    Notes: Summary Transmission in the Ia inhibitory pathway from wrist extensor muscles onto flexor MNs was studied at various times after the onset of voluntary wrist extension or flexion. At the very onset of wrist movements Ia inhibition was not changed, as compared to at rest, whereas later it progressively increased during wrist extension and decreased during wrist flexion. These results are discussed in relation to the different inputs converging onto Ia interneurones and it is suggested that their inhibition by Renshaw cells might be responsible for the results found at the onset of contraction

    Notes: Summary Homonymous Ia facilitation of quadriceps (Q) motoneurones (MNs) was significantly larger during (and at the onset of) a very weak Q voluntary contraction than at rest. This increase in Ia facilitation only appeared with a conditioning-test interval within the narrow range of 5–9 ms, which fits the time course of the recently described polysynaptic Ia excitation to Q MNs. This suggests that interneurones mediating polysynaptic Ia excitatory effects to Q MNs receive a strong descending excitation during such a contraction. It is therefore argued that these interneurones might mediate part of the descending command to Q MNs during voluntary contraction.

    Notes: Summary Reciprocal Ia inhibition from ankle flexors to extensors was studied during voluntary tonic isometric dorsiflexion and plantar flexion in five normal subjects. The Ia inhibition was examined as the short-latency suppression of the soleus H-reflexes by stimulation of the low-threshold afferents in the common peroneal nerve (Mizuno et al. 1971). At rest, weak Ia inhibition was demonstrated in four subjects out of five, the maximal amount being 14.1 ± 5.0% suppression of the control H-reflex. The absolute amount of inhibition, which was calculated by subtracting the mean size of the conditioned H-reflex from that of the control H-reflex and expressed as a percentage of the maximal M-response, increased during ankle dorsiflexion, and decreased or disappeared during plantar flexion in parallel with the amount of contraction. The neural mechanisms for facilitation of the Ia inhibitory pathway during dorsiflexion were considered to support the hypothesis of “α-γ-linkage in reciprocal inhibition”, i.e. combined facilitatory effects on the Ia inhibitory interneurone from the supraspinal centers directly and indirectly via the γ motoneurone — Ia afferent route. The mechanism for inhibition of the pathway during plantar flexion was considered to be inhibition of the Ia interneurone of the flexor side by Ia interneurone of antagonist extensors. A quantitative aspect of activity in the reciprocal Ia inhibitory pathway on the performance of voluntary movement is revealed in this study.

    Notes: Summary The H-reflex technique was used to collect indirect evidence for changes in excitability of the interneurones mediating reciprocal Ia inhibition between wrist extensors and flexors. Stimulating the radial nerve results in an inhibition of the flexor carpi radialis (FCR) H-reflex and evidence has previously been presented that the early phase of inhibition is mediated by extensor-coupled Ia interneurones (Ext Ia INs), i.e. by inhibitory interneurones fed by muscle spindle Ia afferents from wrist extensors. Variations in the level of this inhibition were used to assess changes in excitability of Ext Ia INs. Stimulation of group I fibres from flexors was shown to depress the “reference” Ia inhibition, i.e. to inhibit the Ext Ia INs. The central latency of this interneuronal inhibition was compatible with a disynaptic linkage between flexor Ia afferents and Ext Ia INs. Its threshold and time course profile could almost exactly be superimposed on those of reciprocal Ia inhibition from flexors to extensor carpi radialis (ECR) motoneurones (MNs). This suggests that the Ia inhibitions to extensor MNs and extensor Ia INs are collateral effects mediated by the same flexor-coupled Ia interneurones. In two subjects, in whom it was possible to elicit an H-reflex in the ECR, inhibition of flexor-coupled Ia interneurones by activation of extensor Ia interneurones could similarly be demonstrated.

    Notes: Summary Group I effects from wrist extensors to flexors, i.e. an early inhibition followed by a relative facilitation which are thought to be Ia and Ib in origin respectively, were compared in control conditions and when preceded by a weak cutaneous stimulation. Stimulating the skin of the dorsal side of fingers II–III, which did not modify the test reflex size when applied alone, increased Ib facilitation. By contrast this Ib effect was not changed by stimulation of the skin of the palmar side. It has previously been shown that heteronymous Ib inhibition in the lower limb during voluntary contraction is facilitated by cutaneous stimulation from restricted receptive fields. Thus, present results lend support to the idea that cutaneous facilitation of transmission in Ib reflex pathways might be functional in curtailing exploratory movements.

    Notes: The multilayer thin films having the form of Ti(30 nm)/Fe(dFe)/[Nd–Fe–B(dNd–Fe–B)/Fe(dFe)] ×5/Ti(30 nm)/glass were fabricated on glass substrates by means of radio frequency (rf) sputtering, with dFe varied from 0 to 50 nm and dNd–Fe–B from 0 to 100 nm. Magnetization measurements have revealed that the minor loops of the films are reversible in a certain range of demagnetization fields except for the films with dFe∼0 nm or dNd–Fe–B〈10 nm. Micromagnetic calculation of magnetization curves has been performed, which has well reproduced the observed hysteresis loops including spring-back behavior and the dependence of the coercive field HcJ on dFe and dNd–Fe–B. From the comparison between experiment and calculation, we can infer that the interlayer exchange-coupling strength is about 10% of the intralayer couplings, which are found to be almost independent of dFe and dNd–Fe–B. © 1997 American Institute of Physics.
