    Notes: Abstract This paper describes a neural network model whose structure is designed to closely fit neuroanatomical and-physiological data, and not to be most suitable for rigorous mathematical analysis. It is shown by computer simulation that a process of self-organization that departs from a fixed retinotopic order at peripheral layers and includes hebbian modifications of synaptic connectivity at higher processing levels leads to a system that is capable of mimicking various functions of visual systems: In the initial state the overall structure of the network is preset, individual connections at higher levels are randomly selected and their strength is initialized with random numbers. For this model the outcome of the self-organization process is determined by the stimulation during the developmental phase. Depending on the type of stimuli used the model can either develop towards a featureselective “preprocessor” stage in a complex vision system or towards a subsystem for associative recall of abstract patterns. This flexibility supports the hypothesis that the principles embodied are rather universal and can account for the development of various nervous system structures.

    Notes: Summary Psychophysical experiments were conducted to determine the conditions under which human observers can detect small differences between two successively presented random-dot patterns. The inter-stimulus interval (ISI) varied from 20 to 500 msec. It was found that the appearance of an additional light spot in the second pattern was detectable with ISIs of up to 120 msec. The disappearance of a light spot from the first pattern was detectable with ISIs up to 60 msec. Decreasing the exposure duration of the first pattern worsened the detection of appearance but improved the detection of disappearance. In contrast, decreasing the exposure duration of the second pattern improved the detection of appearance but worsened the detection of disappearance. If the first pattern was presented to one eye and the second pattern to the other under conditions of binocular fusion neither appearance nor disappearance could be detected at any ISI. The results are interpreted as evidence that the transient neural response to the onset of a stimulus is significantly affected by the offset of that stimulus within the preceding 120 msec, and that the transient neural response to the offset of a stimulus is significantly affected by the onset of that stimulus within the following 60 msec. It is suggested that reciprocal inhibition between on- and off-activity could account for these results.

    Notes: Summary Cats were raised under conditions of daily alternating monocular exposure, so that each eye received normal input, but the animals were never allowed to use both eyes simultaneously. With single cell recording techniques it could be shown that this led to a severe disturbance of the normal binocularity of cortical neurons. The cats were trained by operant techniques in a two-choice box to discriminate with each eye non-retinotopic stimulus pairs. To test the ability to combine binocular signals it was tested whether the learned discrimination would transfer to red/green colored stimuli which were viewed dichoptically through appropriately colored contact lenses. The arrangement was such that the positive stimulus could be distinguished from the negative stimuli only when the signals from both eyes were combined and used simultaneously. All cats showed immediate transfer, from the monocularly learned discrimination task to the dichoptic paradigm. This indicates that the presence of a normal population of binocular cortical cells in area 17 is not a prerequisite for the ability to use binocular cues for the solution of a pattern discrimination task.

    Notes: Summary The effects of monocular deprivation from contour vision were investigated in the striate cortex of cats. In addition to the receptive field (RF) properties of single cells responses to electrical stimulation of the deprived and the experienced optic nerve were analyzed: Evoked potentials as well as intra- and extracellularly recorded single unit responses were evaluated. The main goals were: 1. to determine to what extent the responses to electrical stimulation reflected the shift in ocular dominance apparent from the RF analysis, 2. to determine the relative effects of deprivation on excitatory and inhibitory responses and 3. to locate the site of impaired transmission in the pathway from the deprived eye. The results show that the responses to electrical stimulation reflect precisely the shift in ocular dominance apparent from the RF analysis. The evoked potentials elicited from the deprived nerve further indicate that deprivation had also affected the afferent system at the LGN level or (and) at the terminal field of the thalamo-cortical fibers. In contrast to the reduction of short latency excitatory responses to stimulation of the deprived nerve, oligosynaptic inhibition with latencies of 4–6 msec was equally well elicited by stimulation of either eye. The same was true for delayed excitatory responses which frequently occur with latencies between 40 and 80 msec after nerve stimulation. It is concluded from these results 1. that transmission between thalamic afferents and inhibitory interneurones in the cortex is less affected by deprivation than transmission in those pathways which relay cortical excitation, 2. that there is another deprivation resistant indirect pathway from the retina to the visual cortex which is probably relayed through mesencephalic structures and 3. that deprivation effects are not confined to transmission failure at the thalamo-cortical synapses but include alterations already at the presynaptic level.

    Notes: Summary In two dark reared, 40 day old kittens unilateral divergent squint was induced be resecting the insertion of the medial rectus muscle. Behavioural testing revealed that the kittens used only the normal eye for fixation. Contrast sensitivity functions of the two eyes and visual acuity were determined behaviourally in a jumping stand whereby the kittens had to discriminate sine-wave gratings or variable spatial frequency and contrast from a flux equated homogeneous field. At photopic luminance levels the deviated eye showed a significant deficit in both kittens. This impairment was apparent over the whole range of spatial frequencies (0.18–0.99 c/deg) except for the lowest spatial frequency in one kitten. The interocular difference of visual acuity disappeared at scotopic luminance levels. In subsequent electrophysiological experiments contrast sensitivity functions were determined from cortical evoked potentials that were elicited by phase reversing square wave gratings. Comparison between behavioural and electrophysiological results revealed a very good correspondence between the two sets of data. It is concluded that exotropia without alternating fixation leads to functional amblyopia of the deviated eye.

    Notes: Summary In two cats in which surgically induced, unilateral divergent strabismus had led to behaviourally determined amblyopia, a variety of electrophysiological parameters were determined in search of neuronal correlates of squint amblyopia. Tests that assess global neuronal excitability along the pathways from the two eyes to the visual cortex (areas 17 and 18) failed to reflect the functional inferiority of the squinting eye: retinographic responses and cortical evoked potentials elicited by Ganzfeld-stimulation and by stimulation of the optic nerves were identical for the two eyes. The ocular dominance distribution of neurons in area 17 showed the expected disruption of binocularity but failed to provide clear evidence for a functional inferiority of the squinting eye. At other levels of analysis, however, a clear difference between the two eyes was apparent: 1. Responses to optimally aligned light stimuli tended to be more sluggish and the under-representation of neurons with vertically oriented receptive fields was more pronounced in neurons driven from the deviated eye than in cells dominated by the normal one. 2. Interocular inhibition as assessed from electrically evoked potentials was found to be asymmetric; responses evoked from the amblyopic eye were suppressed more readily and over longer periods by conditioning shocks applied to the normal nerve than vice versa. 3. Numerous abnormalities reflecting the functional inferiority of the squinting eye became apparent in cortical potentials evoked by phase reversal of gratings of variable spatial frequency and contrast. A laminar analysis of these field potentials suggests impaired transmission along the intracortical pathways which relay activity to supragranular layers as a major cause for abnormal responses from the squinting eye. It is concluded that squint amblyopia is associated with a variety of neuronal changes at various levels of the visual system, the present data providing evidence for alterations at the cortical level.

    Notes: Summary Fifteen dark-reared, 4- to 5-week-old kittens were stimulated monocularly with patterned light while they were anesthetized and paralyzed. Six of these kittens were exposed to the light stimuli only, in four kittens the light stimuli were paired with electric stimulation of the mesencephalic reticular formation and in five kittens with electric activation of the medial thalamic nuclei. Throughout the conditioning period, the ocular dominance of neurons in the visual cortex was determined from evoked potentials that were elicited either with electric stimulation of the optic nerves or with phase reversing gratings of variable spatial frequencies. In two kittens, ocular dominance changes were assessed after the end of the conditioning period by analyzing single unit receptive fields. Monocular stimulation with patterned light induced a marked shift of ocular dominance toward the stimulated eye, when the light stimulus was paired with electric activation of either the mesencephalic reticular formation or of the medial thalamus. Moreover, a substantial fraction of cells acquired mature receptive fields. No such changes occurred with light or electric stimulation alone. It is concluded that central core projections which modulate cortical excitability gate experience-dependent modifications of connections in the kitten visual cortex.

    Notes: Summary The perigeniculate nucleus of the cat (PGN) was examined at light and electron microscopic levels after immunocytochemical labeling for the gamma-aminobutyric acid (GABA) synthesizing enzyme, glutamic acid decarboxylase (GAD). In light microscopic sections, virtually all perikarya were found to be labeled (GAD+), as well as proximal dendrites, fibres and punctiform elements. Cells in the thalamic reticular nucleus (TRN) dorsal to PGN were also labeled. Ultrastructural analysis of PGN showed immunoreactivity in all somata, in dendrites and in the following vesicle containing profiles: 1.) F1 terminals, which are characterized by large size, dark mitochondria, and pleomorphic vesicles. These terminals form symmetrical synaptic contacts with somata, somatic spines and with dendrites of GAD+ PGN cells. 2.) F2 terminals, which are smaller than F1 terminals, contain also pleomorphic vesicles and frequently make serial synapses of the symmetric type with other F2 terminals. Presumably, F1 terminals are formed by collaterals of PGN-cell axons and F2 terminals by vesicle containing dendrites of PGN cells. Terminals devoid of immunoreactivity included: 1.) RLD terminals characterized by large size, round vesicles, dark mitochondria, and by asymmetric synaptic contacts with somata, especially with somatic spines, and with dendrites of GAD+ perigeniculate neurons; 2.) RSD terminals, characterized by small size, round vesicles and dark mitochondria, which make asymmetric synapses with GAD+ dendrites of medium and small size; 3.) Multivesicular (MV) terminals with variably shaped vesicles including dense core vesicles synapsing on GAD+ dendrites. There are reasons to believe that RSD terminals belong to corticofugal axons and RLD terminals to collateral axons of LGN relay cells. The origin of MV terminals remains to be determined. The GABAergic nature of the PGN cells conforms with the presumed function of these cells as mediators of inhibition of LGN relay cells. The complex synaptic relations observed between GAD+ elements in the PGN would allow for reciprocal inhibition between perigeniculate cells.

    Notes: Summary The method of one-dimensional current source density (CSD) analysis was applied to field potentials recorded from 350 μm thick slices of the primary visual cortex of rats and cats. Field potentials were elicited by stimulation of the white matter and recorded along trajectories perpendicular to the cortical layers at spatial intervals of 25 to 50 μm. The resulting CSD distributions resembled closely those recorded from the cat visual cortex “in vivo”. The responses with the shortest latency were distinct sinks in layers IV and VI probably reflecting monosynaptic EPSP's from specific thalamic afferents. From layer IV activity was relayed along three major routes: 1. to the supragranular layers via strong local connections to layer III and from there via both short and long range connections to layer II, 2. to targets within layer IV, and 3. to layer V. The source distributions suggest that the projections to layers III and II terminate on the proximal and distal segments, respectively, of apical dendrites of layer III pyramidal cells while the projection to layer V contacts the apical dendrites of layer VI pyramidal cells. These results indicate that all the excitatory pathways that are detectable with the CSD technique in the “in vivo” preparation remain intact in 350 μm thick cortical slices. However, in the slice paired pulse stimulation did not lead to a depression of the response to the second stimulus while this is the case “in vivo”. This might be due to reduced inhibition in the slice which has been reported by several authors.
