    Notes: Abstract: The actions of purified β-bungarotoxin (5 or 10 μg/ml) on the metabolic and transmitter-releasing properties of rat cortical synaptosomes was studied. The toxin stimulated control respiratory rates, but prevented the respiratory response to veratrine. Tissue potassium levels were greatly reduced (54%) and endogenous glutamate, aspartate and GABA showed increased levels of release (10- to 25-fold), but other amino acids were unaffected or showed much smaller changes. Tissue levels of these amino acids were reduced in proportion. The toxin inhibited the uptake of [U-14C]GABA (35%) and [U-14C]glutamate (53%) over 5-min incubation periods. This uptake-inhibition was Ca2+-dependent and was reduced by tetrodotoxin. Miniature-end-plate-potential (m.e.p. p.) frequencies at the locust neuromuscular junction (extensor tibialis) were greatly (four- to sevenfold) accelerated by local application of the toxin (5 μg/ml). This effect was reversible and occupied about 20 min. Amplitudes of m.e.p. p.'s were also increased and muscle membrane depolarization occurred. The results are interpreted as being due to a depolarizing action of the toxin.

    Notes: Abstract: Indices of presynaptic cholinergic nerve endings were assayed in neocortical biopsy samples from patients with presenile dementia. For those patients in whom Alzheimer's disease was histologically confirmed, [14C]acetylcholine synthesis, choline acetyltransferase activity and choline uptake were all found to be markedly reduced (at least 40%) below mean control values. The changes occurred in samples from both the frontal and temporal lobes and for [14C]acetylcholine synthesis the decrease was similar under conditions of high and low neuronal activity (as assessed by incubations in 31 mM and 5 mM K+ respectively). Samples from other demented patients, in whom the histological features of Alzheimer's disease were not detected, produced values for all three biochemical parameters which were similar to controls. For the total group of patients with presenile dementia there were correlations between values for the three markers of presynaptic cholinergic nerve endings suggestive of a loss of functional activity at these sites in Alzheimer's disease.

    Notes: We report the application of a technique for performing opacity measurements on hot, dense plasmas. Plasmas were generated by using the x rays emitted from a laser-produced gold plasma to heat an aluminum sample. Observations were made using the technique of point projection spectroscopy. The distribution of ion stages observed is compared with predictions based on Saha distributions, at densities and temperatures deduced from hydrodynamic simulations.

    Notes: Aims:  To report a series of four paediatric cases of interdigitating dendritic cell sarcoma (IDCS) and add to the known extranodal sites of occurrence for this tumour. Neoplasms derived from interdigitating dendritic cells are rare, with only 33 cases being reported in the literature (Medline search). These tumours usually occur in lymph nodes in the adult population.Methods and results:  The patients were a 10-year-old girl with a large soft tissue mass bulging into the left chest, a 12-year-old girl with a right paraspinal mass, a 21-month-old boy with generalized lymphadenopathy and hepatosplenomegaly and a 6-year-old girl with a large bladder mass. Paraffin blocks and haematoxylin and eosin slides were available in all cases. In addition, immunohistochemistry and electron microscopy were performed. A diagnosis of IDCS was made in all cases.Conclusion:  The diagnosis of IDCS can rarely be entertained on clinical information alone. Microscopically, there is a wide spectrum of features. Thus, immunohistochemistry and electron microscopy are crucial in making the diagnosis. The differential diagnosis includes inflammatory pseudotumour, follicular dendritic cell sarcoma, true histiocytic lymphoma, malignant Langerhans cell histiocytosis, anaplastic large-cell lymphoma, melanoma, and a range of sarcomas. IDCS displays aggressive behaviour and approximately half of the patients die of the disease.
