    Notes: Abstract Exudation of phlorotannins (polyphenolics) was measured in situ from the sublittoral kelp Ecklonia radiata at two locations near Sydney, New South Wales, Australia during 1992–1993. Minimally disruptive techniques were used in which individual plants were enclosed within clear plastic bags in order to concentrate exudates. Rates of exudation from E. radiata were low relative to most previous studies, with a mean rate (averaged across four seasons and two sites) of 5.5 μg g (dry wt)-1 h-1. Exudation was greatest in summer and least in winter at one site, but there were no seasonal differences at a second, more protected site. There was no measurable diurnal variation in exudation rates. Exudation after a period of heavy storms was not significantly different from exudation during calm weather, but severe physical damage to the kelp did increase exudation. Our results suggest that exudation of phlorotannins in temperate Australian waters may be less ecologically important than has been suggested for coastal systems in the Northern Hemisphere.

    Notes: Abstract High levels of polyphloroglucinol phenolics in marine brown algae are usually interpreted as a defensive response to herbivory. However, tropical brown algae generally contain very low levels of phenolics, even though herbivory in many tropical systems (e.g. coral reefs) is intense. This apparent paradox would be explained if polyphenolics did not deter tropical herbivores, in which case selection by herbivores for high levels of phenolics in tropical algae would be weak. To examine this hypothesis, in February 1989 we presented mixed assemblages of herbivorous fishes on the Great Barrier Reef with tropical, phenolic-poor brown algae (primarilySargassum spp.) and closely related (conspecifics in one instance) phenolic-rich temperate species. Different species of brown algae were eaten at very different rates, but these differences were not correlated with variation in the phenolic levels among the plants. TLC and NMR analyses showed no evidence of other, non-polar, metabolites in these algae, with the exception of the temperate speciesHomoeostrichus sinclairii. Thus, variation in non-polar metabolites also did not explain the differences in susceptibility to herbivores among these algae. We conclude that the herbivorous fishes studied here were not deterred by phenolic-rich algae, which suggests that levels of phenolics in many tropical algae may generally be low due to their ineffectiveness as defences. However, alternative explanations for the pattern are possible, and these are discussed.

    Notes: Abstract We used random amplified polymorphic DNA (RAPDs) to examine small-scale spatial genetic structure in the red alga Delisea pulchra (Greville) Montagne at two locations near Sydney, Australia. We examined genetic structure among plants at four spatial scales ranging from 2 km apart down to 〈50 cm apart between locations, among sites within locations, among quadrats within sites, and among plants within quadrats. Haploid stages of D. pulchra were absent from the populations studied, suggesting that they are maintained through asexual reproduction of diploid plants. Consistent with this, we found that 19 RAPD phenotypes scored in this study had multiple individuals, indicating the presence of clones in these populations. However, there were no RAPD phenotypes common to two locations separated by only 2 km. Analysis of molecular variance revealed that strong genetic differences occurred between plants from these two locations, with 46.3% of the total genetic variation occurring at this scale, most probably reflecting limited gene flow. Within each location, 〈25% of the genetic variation was attributable to differences among sites or quadrats, indicating gene flow at those smaller scales. Most of the variation within each location occurred at the smallest spatial scale, among plants within 0.25 m2 quadrats. Nonetheless, some pairwise genetic distances (φST) between sites or quadrats within locations were large, indicating some genetic divergence on smaller scales. Genetic distance was independent of spatial distance within both locations, suggesting that fine-scale differences within locations were most probably caused by variation in fine-scale patterns of water movement or fine-scale natural selection. We assessed the impact of one potential selective agent, grazing sea urchins, on the fine-scale genetic structure of D. pulchra. There was no evidence that grazing by sea urchins affected the genetic structure of D. pulchra. In combination with demographic data, our results indicated that local populations of D. pulchra within locations were relatively open and that fine-scale genetic structure was probably constrained by gene flow. At the larger scale however, strong genetic differentiation indicated little gene flow between locations and restricted dispersal of spores.

    Notes: Abstract The localisation of halogenated furanones, the biologically active secondary metabolites from the red alga Delisea pulchra (Greville), was determined by a combination of fluorescence microscopy, culture studies and quantitative chemical analyses. All types of evidence showed that furanones are localised in the central vesicle of gland cells in D. pulchra. These cells release furanones onto the surface of the plant, where they can be quantified using a newly developed surface extraction-technique. Levels of furanones on the surface of the plant were highest near the apical tips (≃100 ng cm−2), and decreased towards the base of the alga. Variation in furanone levels within the plant and variation in the number of gland cells followed a similar pattern. The localisation of furanones within gland cells in D. pulchra and the presence and concentrations of furanones on the surface of the plant are consistent with furanones functioning as antifoulants and in mediating other ecological interactions at the surface of the alga.

    Notes: Abstract  Chronic administration of aflatoxin B1 (AFB1) to rats gives rise to hepatocellular and cholangiocellular carcinomas without affecting Kupffer and endothelial cells. The enzymatic conversion of AFB1 to AFB1-8,9-epoxide is the critical step in the activation of the mycotoxin, while the conversion of AFB1 to aflatoxin M1 (AFM1), catalyzed by the AFB1-9a-hydroxylase, is considered to be a detoxication route for the toxin. In the present study the distribution and inducibility of AFB1-9a-hydroxylase were analyzed in microsomes derived from freshly isolated liver parenchymal (PC) and nonparenchymal cells (i.e. Kupffer+endothelial cells, NPC). AFB1-9a-hydroxylase activity was clearly measurable in NPC and similar to that of PC. In NPC the rate of formation of AFM1 was higher (when incubating with 16 μM AFB1) than or similar (with 128 μM AFB1) to that of AFB1-8,9-epoxide, while in PC it was significantly lower. Taken together, these results suggest that the AFB1-9a-hydroxylase activity might be particularly important in NPC to protect these cells from AFB1 by converting it to a significantly less mutagenic metabolite and by reducing the amount of AFB1 available for epoxidation. Furthermore, it is shown that AFB1-9a-hydroxylase activity is inducible by phenobarbital (only in PC), 3-methylcholanthrene, isosafrole and Aroclor 1254, thus indicating that in rat liver the conversion of AFB1 to AFM1 is catalyzed by members of the cytochrome 1A and 2B families.

    Notes: Abstract  The expression of the pyruvate kinase (PK) isoenzymes L and M2 was analysed in the livers of rats treated with the hepatocarcinogenic agent N-nitrosomorpholine (NNM) in the drinking water. In control animals L-PK expression was restricted to liver parenchymal cells, whereas M2-PK was detected in bile duct epithelial, blood vessel wall, endothelial and Kupffer cells. In rats treated with NNM proliferating oval cells were consistently L-PK negative and M2-PK positive, while the ductal cells of cholangiofibroses were clearly L-PK positive and coexpressed M2-PK. However, no morphological differentiation of ductal cells into hepatocyte-like cells was observed. In the clear and acidophilic cell foci storing glycogen in excess strong staining for L-PK was observed. In glycogen-poor foci induced by NNM a shift from L-PK to M2-PK expression takes place.

    Notes: Summary The influence of inhibitors of metabolism and uptake of noradrenaline on the 3H-noradrenaline removal from the perfusion fluid by the isolated rat liver was studied. Livers were perfused with 3 nmol/l 3H-noradrenaline and 3H-noradrenaline and 3H-metabolites were determined in effluent, liver and bile. After the perfusion with 14,900 ±920 dpm · g−1 · min−1 during 90 min, cumulative removal of tritium was 323,574 ± 63,103 dpm/g. 3H-metabolites recovered from the liver after 90 min perfusion represented 71.1 ± 9.0% of total metabolite formation. Only the OMDA-fraction appeared in the perfusate; its approach to steady state of efflux was slow. The inhibition either of MAO or COMT changed neither the total removal of tritium nor the 3H-metabolites recovered from the liver. Cocaine (10 μmol/l) reduced the accumulation of 3H-noradrenaline in the liver. The uptake2 inhibitor corticosterone (30 μmol/l) diminished total removal of tritium and the 3H-metabolites recovered from the liver without changing the accumulation of 3H-noradrenaline. The hypothesis of two different compartments, one responsible for the metabolism and the other for the accumulation of the amine is discussed.

    Notes: Abstract We examined factors affecting the abundance and distribution of epiphytes (fouling) on the sublittoral kelp Ecklonia radiata. We first assessed the importance of phlorotannins as chemical defences against epiphytes by (a) correlating epiphyte loads on different parts of the thallus with the phlorotannin content of those tissues, and (b) experimentally testing the effects of variation in phlorotannin concentration against the settlement and growth of gametes of Ulva lactuca, a common epiphyte in the system. Tissue phlorotannin content was, at best, only weakly related to epiphyte loads, with r 2 values typically 〈0.10. Inhibition of Ulva gametes only occurred at concentrations 〉10 mg l−1, which is 5 orders of magnitude greater than levels of phlorotannins in the water column around beds of E. radiata, and 1–3 orders of magnitude greater than estimated levels in the boundary layer at the surface of the plant. We concluded that phlorotannins have a negligible impact on patterns of epiphytism on E. radiata, and proceeded to investigate other factors influencing the distribution and abundance of epiphytes. In our samples the relative age of different parts of the thallus was strongly correlated with epiphyte abundance, with epiphyte densities greatest on the oldest tissue and least on the youngest. Distal parts of the thalli also had greater epiphyte loads than basal parts. Field experiments in which kelp tissue was suspended at two heights in an E. radiata forest for varying lengths of time confirmed the importance of the length of time that the tissue was in the water, and its height in the water column, to the development of an epiphyte community. Comparison of epiphyte loads on tissue from primary (smooth) and secondary (rough) laminae in these experiments indicated that surface rugosity also affected fouling. Macroherbivores were rare on E. radiata, and abundances of mesofauna and epiphytes were positively related, suggesting that grazers were not important determinants of patterns of epiphyte abundance. Although phlorotannins have been previously suggested to play an important role as defences against epiphytes, we suggest that water-soluble compounds such as phlorotannins are less likely to be effective defences against epiphytes than non-polar metabolites, which can adhere to the surface of the producing organism.
