Notes: : A water-soluble extract of dry-cured ham was fractionated by gel filtration chromatography. Collected fractions (below 1200 Da) were sensory-analyzed and subjected to amino acid analysis with and without previous acid hydrolysis, in order to determine the amino acid and peptide distribution. Those fractions with the highest peptide amount were further separated by reverse-phase and cation-exchange high-performance liquid chromatography in order to isolate the peptides and sequence them. The results demonstrated the presence of small peptides, mainly dipeptides. The combination of these peptides and free amino acids may contribute to the characteristic taste in the savory fractions of dry-cured ham.

Notes: Abstract In the course of the study of the primary structures and molecular mechanisms of action of immunologically active compounds of the nervous system we have isolated from the soluble fraction of total bovine brain two heat-stable proteins. The purification procedure was mainly based on DEAE-Servacel ion-exchange chromatography and reversed-phase HPLC. The proteins were identified by the N-terminal Edman microsequence analysis and database searching as macrophage migration inhibitory factor (MIF). The N-terminal sequences for MIF1 and MIF2 were found to be identical. According to mass spectral analysis, the molecular masses for MIF1 and MIF2 were determined respectively as 12,369.21 and 12,299.7 Da. In addition, we have also isolated a third peptide having the same N-terminal sequence and Mr 9,496.2 that seems to be a proteolytic fragment of MIF. Using p-hydroxyphenylpyruvate as a substrate, we have not revealed tautomerase activity of either MIF1 or MIF2. As both the immunologic and enzymatic activities were reported to be expressed by the oligomeric structure of MIF, we suggest that the present study may give additional information on MIF in terms of structural properties of this protein. A comparatively simple purification procedure is presented that may be widely used for simultaneous isolation in one run of MIF isoforms.

Notes: Abstract Of four enzymatically active forms of Alcaligenes faecalis penicillin amidase (EC 3.5.1.11) observed in sonicated cells, two (PA5.5 and PA5.3; subscript denotes pI) could be isolated and purified in two steps from the cells destroyed by osmotic shock. Active enzyme was only found in the periplasm. PA5.5 converts further to PA5.3 which differs in the molecular mass of the A-chain. The origin of these differences is a conversion of the N-terminal Gln to pyrolideno-carboxilic acid and a loss of three amino acids from the C-terminus of the A-chain. PA5.3 had higher specific activity (10–30%) for the hydrolysis of benzylpenicillin and 6-nitro-3-phenylacetamidobenzoic acid than PA5.5. © Rapid Science Ltd. 1998

Notes: A number of blends based on suspension poly(vinyl chloride) and stabilizers with poly(ethylene) chlorinated in a fluidized-bed reactor containing 21.8% chlorine, hydroxyl-terminated poly(butadiene), and ethylene-propylene-diene terpolymer have been studied using such methods as thermally stimulated current depolarization and dynamic mechanical analysis. Some dielectric and thermodynamic parameters (τmax, τo, Ea, ΔH*, ΔSE*, ΔG*, μeff) have been determined. Blends containing randomly chlorinated poly(ethylene) exhibited dipole-dipole interactions between the macromolecules of poly(vinyl chloride) which decreased at the expense of the long sequences of nonchlorinated methylene groups. Simultaneously, an increased physical interaction between poly(vinyl chloride) and the additives was observed in blends containing chlorinated poly(ethylene) and/or hydroxyl-terminated poly(butadiene), and ethylene-propylene-diene terpolymer. On the basis of the data of dynamic mechanical analysis obtained a heterogeneous structure of the blends is suggested. The development of a boundary interfacial layer with a proper region of relaxation proves the formation of compatible structures between the components. © 1998 John Wiley & Sons, Inc. Description / Table of Contents: With a series of CPE samples chlorinated in the solid state the composition, degree of chlorination and sequential chlorine distribution have been investigated by 13C-NMR microstructure analysis. The measuring conditions were chosen thus as to avoid the effect of incompatibility caused by chemical inhomogeneity of the degree of chlorination on the results. The samples were separated spectroscopically (without prior fractionation) into two fractions. This evaluation provides two quantitative informations on the maximum PE-like fraction γ as well as on the maximum degree of chlorination of the CPE fraction αCPE and thus on the inhomogeneity of the degree of chlorination.

Notes: The influence of natural, activated, and modified microzeolite with the general formula Me2/m · Al2O3 · nSiO2 · pH2O (where Me = Fe3+, Ca2+, Mg2+, Zn2+, K+, Na+) and an organic additive, such as ammonium sulfamate combined with ammonium sulfate on smoke formation and flammability of suspension poly(vinyl chloride) has been studied in the present work. Microzeolite in native (i.e., natural), activated, and modified forms has been found to possess both fire-retardant and smoke-suppressant effects in solid poly(vinyl chloride) compositions. Such effects have been best revealed by modified microzeolite containing 3,5-4% zinc. On the other hand, the ammonium sulfamate combined with ammonium sulfate is believed to act as a strong smoke-suppressant additive. Thermogravimetric examinations and limited oxygen index measurements show that microzeolite acts as a fireresistant and smoke-suppressant additive predominantly in the condensed phase. The ammonium sulfamate in combination with ammonium sulfate gives evidence of the same effects, mostly in the gaseous phase.

Notes: Natural, activated, and modified microzeolites were studied individually and in combination with ammonium sulfamate as high-melting dispersed additives. The strength - deformation properties of poly(vinyl chloride) were improved within the interval of 3-8 wt % additive. The strengthening effect due to the considered dispersed additives can be ascribed to the optimal adhesive interaction of the phase-boundary surface and to the formation of a partially ordered dispersed phase. The tetrahedral configuration of the modified microzeolite that contains 3.5-4% zinc in its crystal lattice remains unchanged, but exhibits a reduced birefringence. These relationships are of exceptional importance for the use of poly(vinyl chloride). The introduction of certain amounts of the additives improves the melt flow of the polymer and facilitates the processibility of the compositions. © 1993 John Wiley & Sons, Inc.
